Synthesis and selected immunological properties of 10-substituted 1,8-diazaphenothiazines

A new type of tricyclic azaphenothiazines-1,8-diazaphenothiazines-was obtained in the reaction of 2,3- and 3,4-disubstituted pyridines. The reaction ran as the Smiles rearrangement. The 1,8-diazaphenothiazine system was determined using NOE experiment and 2D NMR spectra (COSY, HSQC, HMBC). 10H-1,8-diazaphenothiazine was transformed into 10-derivatives with alkyl, aminoalkyl, amidoalkyl, sulfonamidoalkyl, and nitrogen half-mustard groups. The compounds were tested for their effects on phytohemagglutinin A-induced proliferative response of human peripheral blood mononuclear cells (PBMC) and lipopolysaccharide-induced tumor necrosis factor alpha production by human whole blood cultures. The compounds exhibited differential, dose-dependent inhibitory activities in these tests. All the compounds were low toxic against PBMC. The compounds showing the highest antiproliferative activity strongly inhibited the growth of leukemia L-1210 and colon cancer SW-948 cell lines, similarly as cisplatin, a reference drug.

Realizing the Trifunctional Potential of Alkyl 4-Chloro-2-diazo-3-oxobutanoates: Convenient Assembly of 6,7-Dihydro-4H-[1,2,3]triazolo[5,1-c][1,4]thiazine Core

The first example of exploiting the trifunctional character of alkyl 4-chloro-2-diazo-3-oxobutanoates in the reactions with vicinal N , S -bis-nucleophiles leading to the formation of bicyclic 6,7-dihydro-4 H -[1,2,3]triazolo[5,1- c ][1,4]thiazines is presented. The key to the successful realization of the atom-economical synthetic strategy is the initial S (N) 2 event, which facilitates the subsequent domino 1,2,3-triazole formation via the Wolff reaction. Equally important is the choice of sodium acetate as the base: acetic acid formed in the course of the initial nucleophilic substitution acts as an efficient catalyst for the Wolff reaction, which suppresses the competing, unwanted fragmentation path (observed when other bases are used).

Application of 92-30-8, Consequently, the presence of a catalyst will permit a system to reach equilibrium more quickly, but it has no effect on the position of the equilibrium as reflected in the value of its equilibrium constant.I hope my blog about 92-30-8 is helpful to your research.

Cyclization of N-benzyl cyanoacetamide: Novel synthesis and biological activity of pyrrole, pyrimidine, and pyran derivatives

Heteroannulation of N-Benzyl cyanoacetamide 1 to a new series of heterocycles has been developed. Thus, reaction of 1 with different polarized pi systems afforded pyrrolo 4, pyridone 6, pyridine 8, and diazapene 10 derivatives, respectively. N-Benzyl cyanoacetamide that undergo condensation reaction with salicylaldehyde yielded pyran derivative 11. Nitrosation of 11 furnished condensed pyran 13. Compound 11 reacted with benzaldehyde, carbon disulfide (cyclizing agent), and ammonium thiocyanate to provide pyrane 17, thiazine 18, and thiourea 20 derivatives, respectively. Cinnamoyl isothiocyanate was reacted with compound 11 to produce non-isolable thiourea derivative 21. The newly synthesized compounds have been characterized by infrared (IR), proton nuclear magnetic resonance (H-1 NMR), and carbon nuclear magnetic resonance (C-13 NMR) spectral data. The compounds were then evaluated for antibacterial and anticancer activities.

Synthesis of the symmetrical 3,5-bis(difluoromethyl)-1,4-thiazine 1,1-dioxides

A convenient synthesis of the symmetrical bis(difluoromethyl)-1,4-thiazine 1,1-dioxides was carried out via heterocyclization of bis(2,2,3,3-tetrafluoropropyl)sulfone with ammonia and primary amines. The reaction with excess of ammonia leads to the formation of 3,5-bis(difluoromethyl)-4H-1,4-thiazine 1,1-dioxide. The result of the reaction with primary amines is determined by spatial nature of substituent at the nitrogen atom. 3,5-Bis(difluoromethyl)-4-propyl-4H-1,4-thiazine 1,1-dioxide and 3,5-bis(difluoromethyl)-4-(p-tolyl)-4H-1,4-thiazine 1,1-dioxide were prepared by the reactions of bis(2,2,3,3-tetrafluoropropyl)sulfone with n-propylamine and p-toluidine accordingly. The reaction of bis(2,2,3,3-tetrafluoropropyl)sulfone with tert-butylamine gives the 1,1′-sulfonylbis(N-(tert-butyl)-3,3-difluoroprop-1-en-2-amine). (C) 2015 Elsevier B.V. All rights reserved.

Arachis hypogaea derived activated carbon/Pt catalyst: Reduction of organic dyes

Activated carbon from agro waste groundnut (Arachis hypogaea) shell was prepared by chemical activation and used as support for dispersion of 5% platinum nanoparticles. The Pt nanoparticles were obtained by the reduction in hydrogen gas medium. The synthesized groundnut activated carbon/platinum catalyst was characterized by various techniques such as X-ray powder diffraction, electron microscopies and X-ray photoelectron spectroscopy. The catalytic behaviour of the synthesized catalyst was investigated by exploring it as catalyst for the reduction of various classes of dyes; namely, triphenylmethane dyes such as malachite green, phenol red and bromophenol blue, xanthene dyes: rose bengal, rhodamine 6 G, rhodamine B, thiazine dye: methyelene blue, azo dye: congo red and 4-nitrophenol by sodium borohydride in aqueous medium. Under suitable reaction conditions, for all tested dyes, cationic dyes were reduced at a faster rate than anionic dyes. The rate of reduction on the structure of dye and nature of catalyst was employed.

Selected azaphenothiazines inhibit delayed type hypersensitivity and carrageenan reaction in mice

Several, previously selected azaphenothiazines, as strongly antiproliferative agents in in vitro models, were subjected to evaluation for their potential immunosuppressive effects in the model of delayed type hypersensitivity (DTH) to ovalbumin (OVA) in BALB/c mice and in foot pad inflammation induced by carrageenan in CBA mice. In the DTH model the compounds were given to mice intraperitoneally (i.p.) in 50 mu g or 250 mu g doses, 1 h before the elicitation of the response. In the carrageenan-induced foot pad inflammation the compounds were given i.p. in 50 mu g or 250 mu g doses, 24 h and 2 h before administration of carrageenan. Among the compounds, the significantly suppressive activities in both models were exhibited only by compound 5 (6-chloroethylureidoethyldiquino[3,2-b;2′,3′-e][1,4]thiazine) and compound 4 (6-acetylaminobuty1-9-chloroquino[3,2-blbenzo[1,4]thiazine).
