The seed ofCasuarina equisetifolia, the genus of Pinus of the familyPinaceaewas examined for the potentiality of adsorbing a thiazine-based hetero-polyaromatic dye from water system. For which, the surface functionalization ofCasuarina equisetifolia(SFCE) was achieved through H(2)SO(4)acidic bath to treat methylene blue (MB) dye contaminated water. The SFCE was characterised using scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) to analyse morphological, physical and chemical behaviour after surface modification. The effects of adsorption process influencing variables such as pH, concentration of MB dye, contact time and temperature was examined. The maximum adsorption capacity of 0.5 gL(-1)SFCE for the concentration ranging from 20 to 100 mgL(-1)of MB was investigated using different variables. The effect of parameters including pH, time and temperature were optimised at a speed of agitation of 150 rpm. The adsorption data retrieved from batch experiments were fit with Langmuir, Freundlich, Temkin and Dubinin-Raduskevich (D-R) isotherm models, and as a result, Freundlich presents the best fit (R-2 > 0.97). Pseudo-second order kinetic was superior indicating that SFCE-MB system has undergone the chemisorption mechanism (R-2 > 0.99) spontaneously. Overall, the tested system with synergistic effect was witnessed with maximum adsorption efficiency of 42.19 mg g(-1)to remove dye from industrial wastewater.

Comparative transcriptome analysis of Anguilla japonica livers following exposure to methylene blue

The thiazine dye methylene blue is widely used as a disinfectant and antidote in veterinary medicine. Here, we compared transcriptome differences in the liver of Anguilla japonica exposed to methylene blue using Illumina RNA sequencing technology. A total of 75,953,174 and 86,499,866 clean reads with an average length of 126bp from control and treatment groups respectively. A total of 148,238 transcripts were assembled, with 62,152 components were assembled. Overall, 416,991 unigenes were checked against the National Center of Biotechnology Information, Non-redundant, Cluster of Orthologous Groups (COG), and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases. Of these, 18,541 genes were judged to be differently expressed. The gene ontology (GO) terms molecular function and biological process were the most enriched in protein binding, followed by ATP binding and regulation of transcription, and DNA-dependent. KEGG pathway analysis revealed significant enrichment of N-glycan biosynthesis and the citrate cycle. This study determined that unigenes related to transmembrane transport and drug metabolism in A.japonica were differentially expressed following exposure to methylene blue. These findings can shed insight into the mechanism of methylene blue metabolism in this economically important species.

Epinephrine electrochemical sensor based on a carbon paste electrode modified with hydroquinone derivative and graphene oxide nano-sheets: Simultaneous determination of epinephrine, acetaminophen and dopamine

In this paper, an electrochemical sensor was constructed for determination of epinephrine based on carbon paste electrode (CPE) modified with graphene oxide (GO) and 2-(5-Ethyl-2,4-dihydroxyphenyl)-5,7-dimethy1-4H-pyrido[2,3-d][1,3]thiazine-4-one (EDDPT) as modifiers. The modified electrode was applied as an electrochentical sensor for oxidation of epinephrine (EP). Under the optimum conditions, the over potential of EP oxidation decreased about 279 mV at the modified CPE more than a non-modified CPE. Electrochemical behavior of EP was investigated on the fabricated electrode by cyclic voltammetry (CV) and differential pulse voltammetry (DPV) methods, and some kinetic parameters of EP Were obtained, too. The linear range and the detection limit for EP were found to be 1.5-600.0 WM and 0.65 uM (based on 3 sum), respectively using the EDDPT/GO/CPE sensor and the differential pulse voltammetry (DPV) method. Also, the designed electrochemical sensor was applied to determine EP in the drug sample and simultaneous determination of epinephrine (EP), acetaminophen (AC) and dopamine (DA) in human serum solutions. Antimicrobial Properties of Substituted Quino [3,2-b]benzo[1,4] thiazines

Our previous studies demonstrated that among phenothiazines several derivatives could be found showing strong antiproliferative actions and the property of inhibiting inducible tumor necrosis factor alpha (TNF alpha) production in human blood cultures. The aim of this investigation was to determine potential antimicrobial actions of forty four new phenothiazine derivatives with the quinobenzothiazine structure. The compounds showed differential antibacterial and antifungal activities against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Candida albicans depending on the compound structures, concentrations and bacterial strains. More specifically, 6-(1-methyl-2-piperidylethyl) quinobenzothiazine displayed strongest actions against S. aureus and E. coli whereas 6-methanesulfonylaminobutyl-9-methylthioquinobenzothiazine exhibited the most universal antimicrobial properties. The correlation between antimicrobial activity and the chemical structure of quinobenzothiazines was discussed.

An Efficient One-Pot Synthesis of Benzo[1,4]Thiazines, Benzo[1,3]Thiazoles and Benzo[1,5] Thiazepines

Background: One-pot reactions have gained considerable and wide range of interests in the field of chemistry due to their simplicity and economy. Comparing to the step-by-step reactions that involve the formation of individual bonds for a given target product, the greatest attribute of one-pot reactions, is the formation of several bonds in one-pot operation without intermediate isolation, reaction condition changing and further reagents adding. Objective: We developed an one-pot, rapid, simple and eco-friendly synthetic strategy for benzothiazines, benzothiazoles and benzothiazpines through the interaction of o-aminothiophenol with ketones, nitriles and chalcones, respectively. Method: A mixture of o-aminothiophenol ketones, nitriles and chalcones was refluxed or stirred at room temperature in glacial acetic acid containing a catalytic amount of concentrated H2SO4 for 3h. Then, the resulting precipitate was collected by filtration, washed with water and crystallized from the appropriate solvent. Results: The interaction of o-aminothiophenol with aromatic and aliphatic ketones containing active alpha-hydrogens afforded the corresponding 2H-benzo[1,4] thiazines and 4H-benzo[1,4] thiazines, respectively. Similarly, the cyclic ketones afforded the corresponding tricyclic compounds. Treatment of o-aminothiophenol with nitriles under the same reaction condition gave benzo[1,3] thiazoles. Further, cyclocondensation with chalcones under the same reaction condition at room temperature yielded the corresponding benzo[1,5] thiazepines. The reaction mechanism was investigated and the structures of all new compounds were elucidated using spectral analysis. Conclusion: An efficient, simple and environmentally friendly synthesis of benzothiazines, benzothiazoles and benzothiazepines in one-pot synthesis has been developed. With this catalytic reaction, a very cheap sulfuric acid was used as a catalyst.

Synthesis, characterization, monoamine oxidase inhibition, molecular docking and dynamic simulations of novel 2,1-benzothiazine-2,2-dioxide derivatives

In this research work, we report the synthesis and biological evaluation of two new series of 1-benzyl-4-(benzylidenehydrazono)-3,4-dihydro-1H-benzo[c] [1,2]thiazine 2,2-dioxides and 1-benzyl-4((1-phenylethylidene) hydrazono)-3,4-dihydro-1H-benzo [c] [1,2]thiazine 2,2-dioxides. The synthetic plan involves the mesylation of methyl anthranilate with subsequent N-benzylation of the product. The methyl 2-(N-benzylmethylsulfonamido) benzoate was subjected to cyclization reaction in the presence of sodium hydride to obtain 1-benzyl-1H-benzo[c] [1,2]thiazin-4(3H)-one 2,2-dioxide which was treated with hydrazine hydrate to get corresponding hydrazone precursor. Finally, the titled compounds were obtained by reaction of hydrazone with various substituted aldehydes and ketones. The synthesized derivatives were subjected to carry out their inhibition activities against monoamine oxidases along with modelling investigations to evaluate their binding interactions and dynamic stability during the docking studies. The inhibition profile of potent compounds was found as competitive for both the isozymes. The compounds were more selective inhibitors of MAO-A as compared to MAO-B. Moreover, drug likeness profile of the derivatives was evaluated to have an additional insight into the physicochemical properties. The molecular dynamic simulations predicted the behaviour of amino acids with the active site residues.

Revealing electronic features governing hydrolysis of cephalosporins in the active site of the L1 metallo-beta-lactamase

The QM/MM simulations followed by electron density feature analysis are carried out to deepen the understanding of the reaction mechanism of cephalosporin hydrolysis in the active site of the L1 metallo-beta-lactamase. The differences in reactivity of ten similar cephalosporin compounds are explained by using an extended set of bonding descriptors. The limiting step of the reaction is characterized by the proton transfer to the nitrogen atom of the cephalosporin thiazine ring accompanied with formation of the C-4 & xe001;C-3 double bond in its N-C-4-C-3 fragment. The temporary NMIDLINE HORIZONTAL ELLIPSISH-O-w hydrogen bond, which is formed in the transition state of the limiting step of the reaction was recognized as a key atomic interaction governing the reactivity of various cephalosporins. Non-local real-space bonding descriptors show that different extent of localization of electron lone pair at N atom in the transition state affect the reactivity of compounds: smaller electron localization is typical for the less reactive species. In particular, the Fermi hole analysis shows how exchange electron correlation in the NMIDLINE HORIZONTAL ELLIPSISH-O-w fragment control electron lone pair localization. Delocalization tensor, linear response kernel and source function indicate that features of electron delocalization in the N-C-4-C-3 fragment of cephalosporins in the transition state complexes determine the differences in C-4-C-3 bond for substrates with high and low rate constants. The C-4-C-3 bond of the N-C-4-C-3 fragment at the transition state is similar to that of the preceding intermediate for the less reactive species and resembles the features of the enzyme-product complex for more reactive compounds. The power and limitations of the descriptors applied for solving the problem are discussed and the generality of approach is stressed.
