EXPERIMENTAL AND THEORETICAL INVESTIGATION OF OPTICAL SPECTRA OF METHYLENE GREEN IN SOLUTIONS

The hybrid multilevel approach based on molecular dynamics, quantum mechanics, and statistical theory has been applied to profiles of electronic absorption bands of a number of thiazine dyes (phenothiazine, methylene blue, and methylene green), whose photochemical and biological properties depend on the external medium. The effect of single fragments of the methylene green (MG) molecule on its spectral-luminescent properties is studied. The most preferred geometry for the MG molecule is identified, and its relations with the spectral properties and the solvent nature are established. The energies of the electron states are calculated, and their nature is revealed. The electrostatic potential is analyzed, and the places of the most probable interaction of the molecule with the medium are found. Conclusions on the mechanism of forming the MG absorption spectrum are made based on the observations and calculations performed within the framework of the present work. It is established that the formation of the N-H bonds leads to the formation of an inactive lower excited singlet state and delocalization of the electron charge upon excitation. The position of the maximum of the long-wavelength absorption band of the MG correlates with the donor number of solvents in the series acetonitrile < water < ethanol < isopropanol < dimethyl sulfoxide. The position of the MG electron transition in the region of 630 nm is determined by the interaction of nitrogen and sulfur atoms of the central ring rather than by the increased number of substituents in the structure of the molecule. Protective Effects of 2-Amino-5,6-dihydro-4H-1,3-thiazine and Its Derivative against Radiation-Induced Hematopoietic and Intestinal Injury in Mice

Ionizing radiation (IR) acts as an external stimulating factor, when it acts on the body, it will activate NF-kappa B and cause the up-regulation of inducible nitric oxide synthase (iNOS) and induce a large amount of nitric oxide (NO) production. NO and other reactive nitrogen and oxygen species (RNS and ROS) can cause damage to biological molecules and affect their physiological functions. Our study investigated the protective role of 2-amino-5,6-dihydro-4H-1,3-thiazine hydrobromide (2-ADT) and 2-acetylamino-5,6-dihydro-4H-1,3-thiazine hydrobromide (2-AADT), two nitric oxide synthase inhibitors, against radiation-induced hematopoietic and intestinal injury in mice. Pretreatment with 2-ADT and 2-AADT improved the survival of mice exposed to a lethal dose of radiation, especially, the survival rate of the 2-ADT 20 mg/kg group was significantly higher than that of the vehicle group (p < 0.001). Our findings indicated that the radioprotective actions of 2-ADT and 2-AADT are achieved via accelerating hematopoietic system recovery, decreasing oxidative and nitrosative stress by enhancing the antioxidant defense system and reducing NO as well as peroxynitrite (ONOO-) content, and mitigating the radiation-induced DNA damage evaluated by comet assay. These results suggest that 2-ADT and 2-AADT may have great application potential in ameliorating the damages of radiotherapy. Inclusion of triethylsilane in the cleavage cocktail resulted in the stereoselective formation of the corresponding morpholine/thiomorpholine-3-carboxylic acids. Stereochemical studies revealed the specific configuration of the newly formed stereocenter and also the formation of stable N-acylmorpholine rotamers.

NOVEL SYNTHESIS, RING TRANSFORMATION AND ANTICANCER ACTIVITY OF 1,3-THIAZINE, PYRIMIDINE AND TRIAZOLO[1,5-a]PYRIMIDINE DERIVATIVES

Synthesis, heterocyclization and anticancer activity of a new series of heterocyclic compounds are described. Aminothiazine 1 was obtained from the base induced condensation of thiourea, benzaldehyde and ethyl cyanoacetate. The synthesis of N-phenyl amino pyrimidine derivative 2 was obtained as a result of reaction of aniline with compound 1. Compound 2 underwent ring opening and recyclization upon reaction with HCl or H2O2/NaOH to afford the acid derivative 3 or oxazine 4, respectively. Thiazine 1 undergoes ring transformation upon the effect of NH2OH center dot HCl to produce pyrimidine derivative 5. Heterocyclization of compound 1 with thiosemicarbazide followed by oxidation with I-2/AcOH afforded triazolopyrimidine 6 and 7, respectively. Alkylation of compound 1 was promoted by reaction of 1 with ethyl iodide to give alkylated thiazine 8 which in turn undergo ring transformation when subjected to reaction with hydrazine hydrate to give pyrazole derivative 9. Refluxing of amino-1,3-thiazine derivative 1 with ethyl bromoacetate in the presence of Et3N produce the alkylated pyrimidine product 10. Hydrazonolysis of 1,3-thiazine 1 with hydrazine or phenyhydrazine gave pyrimidine derivatives 11a,b, respectively. Compound 11b was cyclized with carbon disulfide or formaldehyde to produce triazolopyrimidines 12 and 13, respectively. Some of the new compounds were screened for anticancer activity and significant results were found for some compounds.

Development of HPLC method for estimation of glyoxylic acid after pre-column fluorescence derivatization approach based on thiazine derivative formation: A new application in healthy and cardiovascular patients’ sera

Glyoxylic acid (GA) is the intermediate metabolite in various mammalian metabolic pathways. GA showed high reactivity towards formation of advanced glycation end-products (AGEs); the main cause of pathogenesis and complications of many diseases. The presented study aimed to detect GA in healthy and cardiovascular patients’ (CV) sera; however analysis of GA in biological fluid is a challenge and requires chemical derivatization. Hence, a new, highly sensitive, time saving and reproducible pre-column fluorescence derivatization procedure coupled with high performance liquid chromatography (HPLC) method was developed. The derivatization method was based on reaction of 2-aminobenzenthiol (2-ABT), a fluorogenic reagent, with GA in acidic medium to form highly fluorescent thiazine derivative (290 and 390 nm for excitation and emission wavelengths respectively). The fluorescent derivative was separated within 6 min on a reversed-phase ODS column using an isocratic elution with a mixture of methanol-water (70:30, v/v%). The proposed method parameters were optimized and the method was validated. A good linearity in the concentration range (0.05-5.0 mu M) was obtained with detection limit (LOD) of 10 nM (200 fmol/injection), which is more sensitive than several previous methods. Moreover, the recovery results were within the range of 85.0-95.5 % and the intra- and inter-day precision results were <= 3.5%. It should be emphasized that this method is the first one for monitoring of GA in CV patients; to investigate its role for diagnosis and monitoring the severity and complications of this disease in clinical laboratory. Organic dyes festooned with fluorene and fused thiazine for efficient dye-sensitized solar cells

Five new metal free organic dyes festooned with fluorene and fused thiazine structural units such as FPAA1, FPAA2 and FPTAA1-FPTAA3 have been synthesized by adopting multistep synthetic strategy and investigated nanocrystalline TiO2 based dye-sensitized solar cell performance by utilizing the same as sensitizers. Of the devices fabricated, the ones incorporated with cyanoacrylic acid acceptor exhibit greater power conversion efficiencies along with superior electron life time, short-circuit current and open-circuit voltage than the others integrated with (4-oxo-2-thioxothiazolidin-3-yl)acetic acid acceptor. Among them, the dye FPAA1 possessing device exhibited highest PCE of 7.54% besides open circuit voltage (V-OC) of 751 mV, short circuit current density (J(SC)) of 15.65 mA cm(-2), and fill factor (ff) of 0.60.
