Cresyl Violet Stains Mast Cells Selectively: Its Application to Counterstaining in Immunohistochemistry

The thiazine dye toluidine blue (TB) is well known to stain mast cells and hyaline cartilage metachromatically, and thus is mostly often used for their identification. However, TB is not suitable for counterstaining in immunohistochemistry, because of its high-background staining in the cytoplasm of other cell species and in extracellular structures. To expand the knowledge about dyestuffs staining mast cells in consideration with their usage in immunohistochemistry, we determined the stainability of several thiazines and oxazines, which are structurally related compounds to TB, using sections of mast cell-containing tissues. We found that all azine dyes used metachromatically stained mast cells and cartilage. Among these dyes, an oxazines cresyl violet (CV) stained mast cells with lower background, suggesting that those are useful for detecting mast cells and for counterstaining in immunohistochemistry. To ascertain its utility, CV was used in immunostaining of bHSDs in sections from adult rat ovary. Immunopositive signals reflected by DAB development in brown were clearly detected even after CV staining. We conclude that, similar to thiazines, oxazines stain mast cells metachromatically, and that of these, CV is more useful as a counterstain in immunohistochemistry than TB.

Spectral and kinetic features of nonradiative energy transfer in hybrid associates of colloidal quantum dots and organic dyes

The features of nonradiative resonance energy transfer in hybrid associates of colloidal CdS quantum dots (QDs) with molecules of a thiazine dye and J-aggregates of a carbocyanine dye are discussed. The case of an inhomogeneously broadened recombination band of donor (CdS QDs) luminescence is considered. We detected spectral-selective quenching of recombination luminescence of colloidal CdS QDs in association with organic dyes. The maximum of quenching occurs in the region of absorption of the acceptor (organic dye molecule). It is shown using the time-correlated single photon counting technique that the lifetime of the QD luminescence decreases synchronously with the quenching of the luminescence, but only at the wavelengths falling in the region of absorption of the dye. Based on the concept of a donor-acceptor nature of emission of CdS QDs, the dependence of the efficiency of nonradiative energy transfer on the luminescence wavelength is analyzed taking into account the Poisson distribution of its quenchers.

Metal complexes of copper(II), nickel(II), cobalt(II), oxovanadium(IV), chromium(III) and cadmium(II) with a new bridged ONS dibasic tridentate hydrazone (H2L) derived from 5-acetyl-4-hydroxy-2H-1,3-thiazine-2,6(3H)-dione with S-methyl dithiocarbazate have been synthesized and characterized by elemental analysis, molar conductance, magnetic susceptibility measurements, spectral (infrared, electronic, mass, H-1 NMR and ESR) studies as well as thermal gravimetric analysis (TGA). The synthesized complexes have dimeric structures with the general formula [ML(NO3)(m)(H2O)(x)](2)center dot nH(2)O center dot zMeOH, L = dianion of the hydrazone, m = 0-1, x = 0-2, n = 0-4 and z = 0-1. The metal complexes exhibited square planar, tetrahedral and octahedral geometrical arrangements, the molar conductivity data indicates that all complexes are neutral. The Coats-Redfern equation was used to calculate the kinetic and thermodynamic parameters for the different thermal decomposition stages of some complexes. Structural parameters of the ligand and its metal complexes have been theoretically computed on the basis of semiempirical PM3 level and the results were correlated with their experimental data. Microwave-assisted one pot three-component synthesis of some novel pyrazole scaffolds as potent anticancer agents

Background: Pyrazoles, thiazoles and 1,3,4-thiadiazoles have been reported to possess various pharmacological activities. Results: An efficient and a novel approach for the synthesis of some novel pyrazole based-azoles are described via multi-component reaction under controlled microwave heating conditions. The structures of the synthesized compounds were assigned on the basis of elemental analysis, IR, H-1 NMR and mass spectral data. All the synthesized compounds were tested for in vitro activities against two antitumor cell lines, human lung cancer and human hepatocellular carcinoma compared with the employed standard antitumor drug (cisplatin). Conclusions: All the newly synthesized compounds were evaluated for their anticancer activity against human lung cancer and human hepatocellular carcinoma cell lines using MTT assay. The results obtained exploring the high potency of six of the tested compounds compared with cisplatin.

N-Radical initiated aminosulfonylation of unactivated C(sp(3))-H bond through insertion of sulfur dioxide in the presence of visible light is reported. During the reaction process, an N-radical is initiated by the treatment of O-aryl oximes with DABC center dot(SO2)(2) under visible-light irradiation. It is followed by aminosulfonylation of a nearby C(sp(3)) H bond through 1,5-hydrogen atom transfer with accompanying insertion of sulfur dioxide to provide 1,2-thiazine 1,1-dioxide derivatives.
