The mixture was stirred at 30 C for 48 h and then heated to 80 C for 2 h. The mixture was cooled to rt then H20 was added. The mixture was extracted with PE three times (200 mL x 3). The combined organic phase was washed with brine, dried over Na2S04, and concentrated in vacuo. The residue was purified by column chromatography (PE 100%, PE:EtOAc = 60: 1) to get 10.8 g pure and 19 g crude. LC-MS: Rt = 2.29 min; ESI, m/z 344 [M+l]+.

To a solution of 3-oxo-3, 4-dihydro-2H-1, 4-benzothiazin-6- carboxylic acid (3.6 g, 17 mmol) in DMF (50 ML) were added WSC (3.8 g, 20 mmol), HOBt (3.1 g, 20 mmol) and 2.0 M dimethylamine-THF solution (12.5 ML, 25 mmol), and the mixture was stirred at room temperature for 12 hrs. Saturated aqueous sodium hydrogen carbonate was poured into the reaction solution and the mixture was extracted with ethyl acetate. The extract was washed with 1N hydrochloric acid and saturated brine, dried (MGS04) and concentrated under reduced pressure. The residue was crystallized from diisopropyl ether to give the title compound as pale yellow crystals (3.6 g, yield 90%). OH NMR (300 MHz, DMSO-D6) 8 ppm: 2.91 (br, 6 H), 3.49 (s, 2 H), 6. 96-7. 00 (m, 2 H), 7. 35 (d, J = 8. 4 Hz, 1 H).

The reaction mixture was cooled to 0 C. and treated with H2O (0.5 mL), 15% aq. NaOH (0.5 mL), and H2O (1.5 mL). The resulting suspension was filtered. The filtrate was concentrated to provide 340 mg (58%) of the title compound as an amber oil. MS (ESI): exact mass calculated for C9H11NOS, 181.06; m/z found, 182.1 [M+H]+. 1H NMR (500 MHz, CDCl3): 6.96 (d, J=7.9, 1H), 6.60 (dd, J=7.9, 1.6, 1H), 6.49 (d, J=7.9, 1H), 4.52 (s, 2H), 3.65-3.62 (m, 2H), 3.06-3.04 (m, 2H).

A mixture of 1-1 [2-(trifluoromethyl)-l0i7-phenothiazine] (300 mg, 1.12 mmol), 1- bromo-4-chlorobutane (391 mg, 2.28 mmol) and CS2CO3 (1.04 g, 3.19 mmol) in DMF (15 mL) was stirred at rt overnight. The mixture was poured into cool water (20 mL) and extracted with EtOAc (15 mL x 3). The combined organic extracts were washed with brine, dried over NaiSCE. fdtered and concentrated under reduced pressure. The residue was purified by column chromatography (petrol ether: EtOAc =10: 1) to provide a yellow solid (240 mg, 60% yield). LC-MS: Rt = 2.30 min; ESI, m/r 358 [M + 1] +

92-30-8, Compound 2; 10-(3-Chloropropyl)-2-trifluoromethylphenothiazine; To a stirred solution of 2-trifluoromethyl phenothiazine (compound 1) (2g, 7.49 mmol) and sodium hydride (0.5 g, 10.42 mmol) in dry toluene (30 mL) was added l-bromo-3-chloropropane (1.57g, 10 mmol). The reaction mixture was stirred for 18 hours at 110 C under an atmosphere of argon. The solution was cooled to room temperature and poured into an ice- water mixture, the crude product was extracted with ethyl acetate (3×50 mL) and the combined organic phase dried over anhydrous sodium sulphate. Final purification was performed by column chromatography (9:1 hexane: ethyl acetate) on silica gel to give 10-(3- chloropropyl)- 2-trifluoromethylphenothiazine (1.5 g, 58% ) as a solid.

The combined organic layers are dried over sodium sulfate, filtered and concentrated to give a crude product that is purified by preparative HPLC to afford 3-oxo-3,4- dihydro-2H-benzo[l,4]thiazine-6-carboxylic acid [tra?s-4-(8-methoxy-l,2-dihydro-3-oxa- – -5,9-diaza-cyclopenta[a]naphthalen-2-ylmethyl)-cyclohexyl]-amide as a white lyophilized powder (16 mg, 21% yield).1H-NMR (400 MHz, DMSO- 6) delta ppm: 10.67 (s, 1H), 8.49 (s, 1H), 8.23 (d, J = 7.9 Hz, 1H), 8.19 (d, J = 9.0 Hz, 1H), 7.44 (m, 3H), 7.06 (d, J = 9.1 Hz, 1H), 5.21 (m, 1H), 4.01 (s, 3H), 3.74 (m, 2H), 3.51 (s, 2H), 3.14 (dd, J = 7.9, 16.8 Hz, 1H), 1.80-2.00 (m, 5H), 1.66 (m, 1H), 1.57 (m, 1H), 1.39 (m, 2H), 1.13 (m, 2H).MS m/z (+ESI): 505.5 [M+H]+.

After dissolving 2-(trifluoromethyl)-10H-phenothiazine (2.0 g, 7.48 mmol) in N,N-dimethylformamide (2 mL), 60% sodium hydride (149.70 mg, 3.74 mmol) and 1-bromo-4-chlorobutane (1.16 mL, 10.10 mmol) were added at 0 C. The reactants were heated at 100 C. for 12 hours under reflux. After the reaction was completed, the reaction solution was extracted with ethyl acetate and the organic layer was dried with magnesium sulfate, filtered under reduced pressure and then concentrated under reduced pressure. The target compound (1.52 g) was obtained with a yield of 56.72% by separating the residue by chromatography (ethyl acetate/n-hexane=1/20). 1H NMR (400 MHz, CDCl3): delta 1.89-2.00 (4H, m, 2CH2), 3.40 (1H, t, J=6.02 Hz, CH), 3.54 (1H, t, 6.18 Hz, CH), 3.94 (2H, t, 6.02 Hz, CH2), 6.89 (1H, d, J=8.12 Hz, CH), 6.96 (1H, t, 7.44 Hz, CH) 7.03 (1H, s, CH), 7.13-7.17 (2H, m, 2CH), 7.21 (2H, t, 8.16 Hz, 2CH).
