Hydrazide-integrated carbazoles: synthesis, computational, anticancer and molecular docking studies

A new class of carbazole-based hydrazides 6a-c, synthesized from carbazole by employing a multistep synthetic strategy, has been described. Detailed insight into their structures (6a-c) has been elucidated by UV-Vis, FT-IR and NMR (H-1 and C-13) spectroscopic studies. Theoretical investigation of the molecules 6a-c has been accomplished utilizing DFT and TD-DFT techniques with the B3LYP/6-311++G(d,p) method. Theoretical findings such as optimized structural, vibrational, and electronic properties, and proton and carbon chemical shifts of the targets 6a-c are in harmony with their experimental results and/or structurally related reported ones. Cytotoxicity of the target hydrazides 6a-c has been evaluated using human pancreatic cancer cells (AsPC1 and SW1990). The hydrazides 6a-c displayed a significant in vitro cytotoxic effect against both the pancreatic cancer cells AsPC1 (concentration that inhibits 50% cell viability, IC50: 3.42 +/- 0.41 mu M for 6a) and SW1990 (IC50: 22.42 +/- 1.40 mu M for 6a). The superior binding energy resulting from the in silico molecular docking approach of the hydrazide 6a indicates its greater affinity towards the receptor (binding energy: -8.63 kcal mol(-1) and IC50: 475.05 nM). Thus, the hydrazide 6a could serve as a new lead for the development of anticancer agents.

Methylene blue induces the soxRS regulon of Escherichia coli

Extensive application of methylene blue (MB) for therapeutic and diagnostic purposes, and reports for unwanted side effects, demand better understanding of the mechanisms of biological action of this thiazine dye. Because MB is redox-active, its biological activities have been attributed to transfer of electrons, generation of reactive oxygen species, and antioxidant action. Results of this study show that MB is more toxic to a superoxide dismutase-deficient Escherichia coli mutant than to its SOD-proficient parent, which indicates that superoxide anion radical is involved. Incubation of E. coli with MB induced the enzymes fumarase C, SOD, nitroreductase A, and glucose-6-phosphate dehydrogenase, all controlled by the soxRS regulon. Induction of these enzymes was prevented by blocking protein synthesis with chloramphenicol and was not observed when soxRS-negative mutants were incubated with MB. These results show that MB is capable of inducing the soxRS regulon of E. coli, which plays a key role in protecting bacteria against oxidative stress and redox-cycling compounds. Irrespective of the abundance of heme-containing proteins in living cells, which are preferred acceptors of electrons from the reduced form of MB, reduction of oxygen to superoxide radical still takes place. Induction of the soxRS regulon suggests that in humans, beneficial effects of MB could be attributed to activation of redox-sensitive transcription factors like Nrf2 and FoxO. If defense systems are compromised or genes coding for protective proteins are not induced, MB would have deleterious effects.

An unusual electrochemical oxidation of phenothiazine dye to phenothiazine-bi-1,4-quinone derivative (a donor-acceptor type molecular hybrid) on MWCNT surface and its cysteine electrocatalytic oxidation function

Phenothiazine (PTZ), a thiazine class heterocyclic compound, is a well-known electron donating system and has been widely used as a starting compound to prepare various phenothiazine dyes and pharmaceutically important compounds. Quinones and its derivatives are constituents of biologically active molecules serve as excellent electron-acceptor systems. Oxidation of PTZ by chemical and electrochemical methods often resulted into monohydroxylation of benzene ring moiety, S-oxidized and polymerized compounds as end products. Electrochemical oxidation of PTZ on a multiwalled carbon nanotube (MWCNT) modified glassy carbon electrode in pH 7 phosphate buffers solution (PBS) has been investigated in this work. A highly redox active surface confined PTZ-bi-1,4-quinone derivative (PTZ-biQ) on MWCNT modified glassy carbon electrode, designated as GCE/MWCNT@PTZ-biQ, as a product was unusually observed. The GCE/MWCNT@PTZ-biQ showed well-defined redox peaks at E-1/2 = -0.07 and +0.29 V vs Ag/AgCl corresponding to surface confined electron-transfer behavior of the bi-quinone (acceptor) and PTZ-cationic radical species (donor) respectively. No such electrochemical characteristics were noticed when unmodified GCE was subjected to the electrochemical oxidation of PTZ. Existence of PTZ-biQ was confirmed by XRD, Raman spectroscopy, FT-IR and GC-MS (methanolic extract of the active layer) analyses. Position of biQ in PTZ-biQ as 1,4-quinone isomer was confirmed by observation of absence of copper-complexation with 1,4-quinone and H2O2 electrochemical reduction reactions at -0.1 V vs Ag/AgCl unlike to the specific copper-complexation and H2O2 reduction with 1,2-quinone isomer in pH 7. Cysteine (CySH) oxidation was studied as a model system to understand the electron-transfer function of the MWCNT@PTZ-biQ. A highly selective electrocatalytic oxidation and sensing by amperometric i-t and flow injection analysis of CySH at low oxidation potential, 0.3 V vs Ag/AgCl in pH 7 PBS with detection limit values (signal-to-noise ratio = 3) of 11.10 mM and 110 nM respectively, without any interference from other biochemicals like uric acid, dopamine, nitrite, citric acid and H2O2, unlike the conventional chemically modified electrodes with serious interference’s, have been demonstrated. Microwave-assisted regioselective reaction of furanone derivative supported by DFT stimulation and molecular docking to afford controlling insecticidal agents

The furanone 1 was allowed to react with different nitrogen nucleophiles such as hydrazine hydrate, hydroxyl amine, anisidine, anthranilic acid, glycine, ethylglycinate and isatin and some carbon nicleophiles under Michael reaction conditions assisted by microwave irradiation. They afforded new synthesized some important heterocyclic compounds 2-16. The structures of the synthesized new compounds were characterized by spectral data and elemental analysis. DFT calculation is serving to clarify not only the active electrophilic sites attacked by suitable nucleophiles in the organic reaction but also the bioactivity of the larvicidal compounds. The larvicidal activities of the synthesized compounds were tested against Plutella xylostella and Helicoverpa armigera larvae in vitro. The results revealed that the synthesized compounds 4, 13 and 14 showed good insecticidal activity against Plutella xylostella larvae with 100%, 87.2% and 91.4% activities, respectively, while 13 and 14 displayed 92% and %, 94.8 activities, respectively, against Helicoverpa armigera larvae. Therefore, they exhibited good larvicidal activity. The executed molecular docking simulations evinced the selective binding of the novel synthesized compounds toward acetylcholinesterase (AchE) of Drosophila Melanogaster, which signifies their role as a prominent larvicides. Compound 4, 13 and 14 showed clear docking affinity and to the assumed binding sites in terms of both calculated binding energies and the types of interactions, which provided a solid rationalization for the conducted experimental results.

Synthesis and Investigation of Pinane-Based Chiral Tridentate Ligands in the Asymmetric Addition of Diethylzinc to Aldehydes

A library of pinane-based chiral aminodiols, derived from natural (-)-beta-pinene, were prepared and applied as chiral catalysts in the addition of diethylzinc to aldehydes. (-)-beta-Pinene was reacted to provide 3-methylenenopinone, followed by a reduction of the carbonyl function to give a key allylic alcohol intermediate. Stereoselective epoxidation of the latter and subsequent ring opening of the resulting oxirane with primary and secondary amines afforded aminodiols. The regioselectivity of the ring closure of the N-substituted secondary aminodiols with formaldehyde was examined and exclusive formation of oxazolidines was observed. Treatment of the allylic alcohol with benzyl bromide provided the corresponding O-benzyl derivative, which was transformed into O-benzyl aminodiols by aminolysis. Ring closure of the N-isopropyl aminodiol derivative with formaldehyde resulted in spirooxazolidine. The obtained potential catalysts were applied in the reaction of both aromatic and aliphatic aldehydes to diethylzinc providing moderate to good enantioselectivities (up to 87% ee). Through the use of molecular modeling at an ab initio level, this phenomenon was interpreted in terms of competing reaction pathways. Molecular modeling at the RHF/LANL2DZ level of theory was successfully applied for interpretation of the stereochemical outcome of the reactions leading to display excellent (R) enantioselectivity in the examined transformation.

Dihydropyrimidinone derivatives were prepared by tri-component reaction of ethyl aceto acetate, aldehydes and thiourea in the presence of modified montmorillonite nanostructure as a catalyst and used as key intermediates for the synthesis of ethyl-5-(aryl)-2-(2-alkokxy-2-oxoethylidene)-7-methyl-3-oxo-3,5-dihydro-2H-thiazolo[3,2-a]pyri midine-6-carboxylate derivatives with use of diethyl and dimethyl acetylene dicarboxylate by two methods: (a) in methanol as a solvent under ultrasonic irradiation at ambient temperature (b) in methanol as a solvent at ambient temperature (conventional magnetic stirring). SYNTHESIS AND ANTIMICROBIAL ACTIVITY OF NOVEL OXAZOLIDINONES HAVING BENZO THIAZINE DERIVATIVES

In order to develop relatively small molecules as pharmacologically active molecules, a series of novel oxazolidinones having benzothiazine and their derivatives were synthesized and characterized by IR, H-1 NMR and Mass spectral studies. Various substituted oxazolidinone benzothiazines were prepared by simple refluxing in the presence of acetonitrile. Treatment of these oxazolidinone benzothiazine derivatives with methanesulfonyl chloride gives its sulphonate derivatives which on further treatment with sodium azide and triphenyl phosphine in acetic anhydride gave acetamide derivatives. Further the synthesized compounds were evaluated for antibacterial activity against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and antifungal activity against Candida albicans and Aspergillus niger.
