The title adduct, chlorido(2,3-diphenyl-3,4,5,6-tetrahydro-2H-1,3-thiazin-4-one-kappa O) triphenyltin, [Sn(C6H5)(3)Cl(C16H15NOS)], resulted from reaction of 2,3-diphenyl- 3,4,5,6-tetrahydro-2H-1,3-thiazin-4-one with triphenyltin chloride. The thiazine ring has an envelope conformation with the S atom forming the flap. The molecule has five phenyl rings, two of them attached to the thiazine ring at positions 2 and 3, and three in coordination with the Sn-IV atom. The three rings of the triphenyltin group are involved in intramolecular interactions of different types, C-H center dot center dot center dot O, edge-to-face (or T-type) pi-pi interactions with the 3-phenyl ring of the thiazine, T-type interactions with both phenyl rings of the thiazine etc. On the other hand, all the phenyl rings participate in intermolecular pi-pi interactions. There is one instance of a ‘parallel-displaced’-type interaction extending continuously along the a-axis direction and seven instances of T-type interactions stabilizing the crystal lattice.

Synthesis of New Derivatives of Heterocyclic Systems Containing Triazolopyrimidine, thiazolo[3,2-a]pyrimidine and pyrimido[2,1-b] thiazine Moiety Showing Promising Antimicrobial Activity

Background and Objective: Continuing our research in the field of heterocyclic compounds and exploiting our experience in the synthesis of pyridothieno(furo)[3,2-d]pyrimidine derivatives decorated by suitable reactive groups (in turn able to give rise to the formation of new heterocyclic rings), we now carried out a research with the aim of building in different position of the pyrimidine some new hetero-rings: the 1,3,4-triazole, thiazole and thiazine. Moreover their possible antimicrobial activity against some gram-positive and gram-negative bacilli strains has been evaluated. Methods: Starting from the hydrazino derivative of pyrido[3′,2′:4,5]thieno(furo)[3,2-d]pyrimidines 2 (7) by reflux with triethyl orthoformate or formic acid, some substituted isomeric pyrido[3′,2′:4,5]thieno(furo)[2,3-e][1,2,4]triazolopyrimidines 3 and 4 (10) have been synthesized. By alkylation of compounds 8 with alkyl dichlorides, the cyclization to a thiazoline or to a thiazine ring on the [a] side of the pyrimidine ring occurs, with the formation of the new pentacyclic systems: pyrido[3′,2′:4,5]thieno[3,2-d][1,3]thiazolo[3,2-a]pyrimidin-7(8)-one 15 and pyrido[3 ”,2 ”:4′,5′]furo(thieno)[3′,2′:4,5]pyrimido[2,1-b][1,3]thiazin-7(8)-ones 16. Results: 11-Alkyl(aryl)-8,8-dimethyl-7,10-dihydro-8H-pyrano[4 ”,3 ”:4′,]pyrido[3′,2′:4,5]thieno[2,3-e][1,2,4] triazolo[4,3-c]pyrimidines 3 gave a Dimroth rearrangement in acidic media. It was observed that the cyclization of compounds 7 and 8 occurred linearly with the expected formation of pyrido[3′,2′:4,5]thieno(furo)[3,2-d][1,2,4]triazolo[4,3-a]pyrimidin-7(8)-ones 10 and pyrido[3′,2′:4,5]thieno[3,2-d][1,3]thiazolo[3,2-a]pyrimidin-7(8)-one 15 and pyrido[3 ”,2 ”:4′,5′]furo(thieno)[3′,2′:4,5]pyrimido[2,1-b][1,3]thiazin-7(8)-ones 16: that is triazole, thiazole and thiazine ring condensation occurs at the [b] side of the pyrimidine ring. During the alkylation of compounds 7, the unusual cleavage of the hydrazino group took place with the formation of 8(9)-methylpyrido[3′,2′:4,5]thieno(furo)[3,2-d]pyrimidin-7(8)-ones 12. Conclusion: The synthesis of some new ‘complex’ pentaheterocyclic systems containing triazolopyrimidine, thiazolo[3,2-a]pyrimidine and pyrimido[2,1-b]thiazine moiety based on the pyrido[3′,2′:4,5]thieno(furo)[3,2-d]pyrimidines is described. During the alkylation of 9(10)-hydrazinopyrido[3′,2′:4,5]thieno(furo)[3,2-d]pyrimidin-7(8)-ones 7 an unexpected cleavage of the hydrazino group has been observed with the formation of 8(9)-methylpyrido[3′,2′:4,5]thieno(furo)[3,2-d]pyrimidin-7(8)-ones 12. Biological tests on the newly synthesized compounds evidenced that some of them showed promising antimicrobial activity.

Breslow Intermediates from a Thiazolin-2-ylidene and Fluorinated Aldehydes: XRD and Solution-Phase NMR Spectroscopic Characterization

The first generation and X-ray diffraction (XRD) analysis of a crystalline Breslow intermediate (BI) derived from a thiazolin-2-ylidene, that is, the aromatic heterocycle present in vitamin B-1, is reported. Key to success was the combined use of pentafluorobenzaldehyde and a thiazolin-2-ylidene carrying an enol-stabilizing dispersion energy donor as N-substituent. A so-called primary intermediate (PI) could be isolated in protonated form (pPI) as well and analyzed by XRD. When switching to a saturated thiazolidin-2-ylidene, reaction with pentafluorobenzaldehyde afforded a new BI in solution (NMR spectroscopy). Attempts to crystallize the latter BI resulted in the isolation of a novel thiazolidin-2-ylidene dimer that had undergone rearrangement to a hexahydro[1,4]-thiazino[3,2-b]-1,4-thiazine.

Visible light-driven oxidation of arsenite, sulfite and thiazine dyes: A new strategy for using waste to treat waste

Recently, the advanced oxidation processes (AOPs) based on sulfate radical (SO4 center dot-) using sulfite has received increasing concerns. Metal-free activation of sulfite is still in urgent demands from the perspective of cleaner production since the reported efficient systems containing sulfite has been relied on the transition metals. In this work, methylene blue (MB+), a well-known photosensitizer and thiazine dye contaminant, was utilized to activate sulfite for the efficient photooxidation of arsenite (As(III)) to arsenate (As(V)) under visible light (LED lamps with wavelength 640 nm) and sunlight, respectively. The initial oxidation rates of 5-40 mu M As(III) are 28-folde116-fold as that of the decolorization of 0.27 mu M MB+ with 50 mu M sulfite at pH 7.3. The maximum stoichiometric ratio MB+: As(III): sulfite was obtained to be 1 : 142: 247, suggesting an efficient utilization of both MB+ and sulfite as waste resources. The electrical energy per order (EE/O) index was calculated to be 1.83 x 10(-3) kWh L-1 for the visible light-MB+-sulfite system, which is much less than those reported systems. Oxysulfur radicals (mainly peroxymonosulfate radical, SO5 center dot-) produced in this photochemical system are responsible for As(III) oxidation. The other two common thiazine dyes (thionine and Azure B) are also capable of activating sulfite for As(III) oxidation due to the same chromophores. Thus the novel strategy of using waste sulfite and the thiazine dyes is promising for As(III) oxidation under sunlight, with which low-concentration wastes of thiazine dye and sulfite can be utilized to achieve the goal of cleaner production. Paper-based diagnostic devices for evaluating the quality of human sperm

Male infertility, which amounts to half of all infertility cases, is a serious problem worldwide. The percentage of fertility-related patients in sub-Saharan African countries is higher than that for the developed countries. In low-resource countries, examination of sperm characteristics for male infertility cannot be undertaken because of poor clinical access. To evaluate male fertility in assisted reproductive medicine laboratories, the numbers of motile human sperm, the degree of sperm motility, and sperm morphology have been commonly analyzed using a microscope. It is challenging to monitor the health status of human sperm in resource-limited or remote settings for two primary reasons: (1) high capital cost (equipment for currently accepted procedural standard), and (2) complexity of the currently accepted procedural standard used to simultaneously measure human sperm concentration and motility by skillful embryologists. Determining the health status of human sperm in order to evaluate fertilization capacity using various types of low-cost, easy-to-use, and rapid devices (or systems) is a longstanding but interesting biotechnologically relevant issue in various scientific communities such as male reproduction. Furthering such efforts will inherently influence birth rate in both developed and developing nations. We have demonstrated an inexpensive but robust and easy-to-handle device for monitoring the health status of human sperm made by patterning a piece of paper and measuring the activity of a specific enzyme-a simple and elegant solution. After applying semen to the hydrophilic center circle of our patterned paper, a thiazine assay can be used to suggest sperm concentration in semen, and a tetrazolium-based colorimetric assay (MTT assay) data can be used to help estimate the percentage of motile human sperm (sperm motility) in semen based on the character that motile human sperm moved in and on the paper. Using this paper-based device, we can evaluate fertility levels without consulting doctors and use our assay to compare results with World Health Organization (WHO) reference values for sperm concentration (> 2×10(7)) and motility (> 50 %). The duration and cost of one entire test are 30 min and 0.1 USD, respectively. We believe that this paper-based assay system would be useful for fertility checks based on WHO references, without need of a microscope, at home. Using this assay method, males in developed or developing countries who are reluctant or unable to consult assisted reproductive technologies clinics can self-analyze their sperm characteristics. We further note that our approach adheres to WHO regulations, especially in regard to in vitro diagnostic device performance with an associated diagnostic algorithm to enhance diagnostic accuracy (compared with just one diagnostic output), and we wish to emphasize that our research could significantly advance a broad range of diagnostic developments including paper-based diagnostic devices, in vitro diagnostic devices, and diagnosis of other diseases in various divisions of translational medicine. These results, we believe, will be of interest to a wide scientific audience working in materials science (biomaterials), chemistry (analytical and clinical), lab-on-a-chip technologies (the development of diagnostic tools), reproductive medicine, bioengineering, and translational medicine.

Evaluation of the effect of 2-(2,4-dihydroxyphenyl)-4H-benzofuro[3,2-d][1,3]thiazin-4-one on colon cells and its anticancer potential

In this paper, we present the biological effect of the newly synthesized 2-(2,4-dihydroxyphenyl)-4H-benzofuro[3,2-d][1,3]thiazin-4-one (DPBT) on human colon adenocarcinoma cell lines (HT-29 and LS180). Additionally, DPBT cytotoxicity was examined in human colon epithelial cells (CCD 841 CoTr) and human skin fibroblasts (HSF). The studies revealed a significant decrease in the proliferation of cancer cells after exposure to DPBT at concentrations in the range of 10-100 mu M. Additionally, DPBT was not toxic to normal CCD 841 CoTr and HSF cells at concentrations that induced inhibition of cancer cell proliferation. The nature of the anti-proliferative action of DPBT in the cell cycle progression in colon cancer cells and the expression of proteins involved in this process were examined by flow cytometry and western blotting, respectively. The investigations demonstrated higher sensitivity of LS180 than HT-29 to the DPBT treatment. The anti-proliferative action of DPBT in LS180 was attributed to cell cycle arrest in the G(1) phase via up-regulation of p27(KIP1) and down-regulation of cyclin D1 and CDK4 proteins.
