Novel chitosan-cystamine-retinoic acid conjugate (CS-SS-RA) was synthesized and could self-assemble into redox-sensitive micelles in aqueous environment with low critical micelle concentration value. CS-SS-RA micelles were characterized with spherical shape, desirable particle size, negative zeta potential, high paclitaxel (PTX) loading and encapsulation efficiency and redox-sensitivity. Hemolysis and cytotoxicity studies proved the safety of CS-SS-RA micelles for intravenous administration. Cytotoxicity study against HepG2 cells and the growth inhibition study on three-dimensional multicellular tumor spheroids (MCTSs) revealed that PTX-loaded CS-SS-RA micelles exhibited higher antitumor activity than free PTX. The in vitro cellular uptake profiles of FITC-labeled CS-SS-RA micelles evaluated via confocal laser scanning microscopy and flow cytometry indicated that CS-SS-RA micelles could enhance cellular uptake efficiency of PTX, and their internalization by HepG2 cells were mediated by clathrin-mediated endocytosis and macropinocytosis. These results demonstrated that CS-SS-RA micelles could be developed as a promising platform for intracellular delivery of hydrophobic antitumor agents.

Carbon quantum dots (CQDs) show promising potential for tumor imaging owing to their unique superior fluorescent properties. However, the small particle size limits their practical application. Here, pH/reduction dual-responsive comet-shaped PEGylated CQD-DOX conjugate prodrug, DOX-Hy-CQD-SS-PEG with DOX content of 28.5%, was designed with the hydrophobic acid-labile DOX conjugated CQDs as comet nucleus and the few hydrophilic bioreducible detachable PEG brushes as comet tails. The comet-shaped DOX-Hy-CQD-SS-PEG prodrug could self-assemble into unique micelles with mean diameter of 127 nm. The DOX-Hy-CQD-SS-PEG micelles possessed excellent pH/reduction dual-responsive drug release with low drug leakage of 9% in 150 h. Furthermore, the fluorescent CQDs was recovered after DOX release and de-PEGylation, demonstrating their potential application for real-time response of therapy.

A series of isocyanate-free polyurethanes (NIPUs), showing an intrinsic thermo-healing performance, have been synthesized from sustainable vegetable oil-based cyclic carbonates and bio-based amines. The bio-based contents of the NIPUs are higher than 78%. Their thermo-mechanical and self-healing performances are tailored simply by varying the feed ratios of the amines employed. The biomass linoleic acid dimer-based diamine plays a positive role in maintaining the higher thermal stability and good elasticity of the prepared NIPUs, the cycloaliphatic isophorone diamine contributes to high tensile strength, and cysteine-derived disulfide-containing cystamine (CA) enhances the self-healing efficiency. The combination of intrinsic hydrogen bonds existed in the NIPU matrix with the dynamic exchange reactions of disulfide bonds and the enhanced flexibility of NIPU networks results in 98.1% self-healing efficiency at 25 degrees C. The contributions of disulfide bonds and hydrogen bonds to the healing efficiency were estimated by adjusting the composition of NIPUs, showing that the contribution of disulfide bonds to the healing ability is about 45.3% even at a low amount of CA (6.0 wt%).

Therapeutic delivery selectively to lymph nodes has the potential to address a variety of unmet clinical needs. However, owing to the unique structure of the lymphatics and the size-restrictive nature of the lymph node reticular network, delivering cargo to specific cells in the lymph node cortex and paracortex is difficult. Here, we describe a delivery system to overcome lymphatic and intra-lymph node transport barriers by combining nanoparticles that are rapidly conveyed to draining lymph nodes after administration in peripheral tissues with programmable degradable linkers. This platform enables the controlled release of intra-lymph-mobile small-molecular cargo, which can reach vastly more immune cells throughout the lymph node than either the particles or free compounds alone. The release rate can be programmed, allowing access to different lymph node structures and therefore specific lymphocyte subpopulations. We are thus able to alter the subtypes of drugged lymph node cells to improve immunotherapeutic effects. Nanoparticles that access lymphatic vessels and are functionalized with degradable linkers, whose half-lives can be programmed, enable the controlled release of therapeutic cargo in different regions of the lymph nodes, allowing the targeting of otherwise difficult-to-reach lymphocyte subpopulations.

As a potential method for tumor treatment, tumor vaccine immunization induces a tumor-specific cellular immune response via immunization with tumor antigens. The delivery of exogenous antigen proteins into the cytoplasm of antigen-presenting cells is well known to induce an intensive cellular immune response for tumor treatment. In this work, we fluorinated a redox-responsive hyperbranched poly(amidoamine) (HPAA) with heptafluorobutyric anhydride to prepare a fluorinated HPAA (HPAA-F7) for use as a vaccine delivery system for antitumor therapy. The immunization results show that HPAA-F7 as a vaccine carrier could effectively promote the intracellular uptake and cytoplasmatic delivery of antigen proteins and induce potent antitumor cellular immunity. The novel vaccine carrier HPAA-F7 could be further developed for antitumor immunotherapy.

Crosslinked polymer nanomaterials have attracted great attention due to their stability and highly controllable drug delivery; herein, a series of well-defined amphiphilic PDPA-b-P(NMS-co-OEG) diblock terpolymers (P1-P3) were designed and prepared via RAFT polymerization and were self-assembled into non-cross-linked (NCL) nanomicelles, which were further prepared into shell-cross-linked (SCL) micelles via cystamine-based in situ shell cross-linking. Using P3 as an optimized polymer, SCL-P3 micelles were prepared, which demonstrated remarkable pH/redox-dual responsive behaviour. For drug delivery, camptothecin (CPT)-loaded SCL-P3 micelles were prepared and showed much higher CPT-loading capability than their NCL-P3 counterparts. A well-defined functional star polymer nanoparticle was prepared using an “ arm first” approach via RAFT polymerization. In this work, N-adamantylacrylamide and N-succinimidyl acrylate were copolymerized with N-isopropylacrylamide sucessfully. The obtained diblock copolymer regarded as an “ arm” endowed the star polymer with multiple conjugate functions. By adjusting the amount of crosslinking agent, the size and polydispersity of the star polymer were controlled. With the star polymer as the scaffold, a sequential multi-enzyme system was constructed, where HRP was placed in the inner layer through covalent conjugation, and b-cyclodextrin-modified GOx was assembled in the outer layer through hostguest recognition. More interestingly, the activity of the enzyme modified by b-cyclodextrin was improved. In addition to the enhanced thermal stability, this multienzyme system also exhibited a reduced Km value (from 2.18 mM to 0.39 mM) and an excellent substrate affinity. The specificity constant (Kcat/Km) for GOx in the star polymer@ HRP@ GOx system was 1.7-fold higher than that of the free HRP with free GOx system. This strategy will promote the development of biocatalysis reactions and biosensors as a promising method.

A dumbbell-shaped copolymer (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) with a lower critical solution temperature (LCST) and redox responses was first synthesized by the combination of ring opening polymerization (ROP), click chemistry and atom transfer radical polymerization (ATRP). Then a dumbbell-shaped copolymer (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) with an upper critical solution temperature (UCST) and redox responses was obtained through the quaternization reaction between DMAEMA and excess 1,3-propane sultone. Both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) were amphiphilic, and they could self-assemble into spherical micelles under certain conditions. Benefitting from the LCST-type thermoresponse of PDMAEMA and UCST-type thermoresponse of PDMAPS, both dumbbell-shaped copolymers (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) presented temperature-responsive properties. The morphology and size of their assemblies would be changed when the temperature is altered. Meanwhile, due to the redox-response of disulfide bonds, both (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) and (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) showed redox-responsive properties. When dl-dithiothreitol (DTT) was added into the systems, disulfide bonds were broken, causing the change of (PDMAEMA)(4)-PCL-SS-PCL-(PDMAEMA)(4) to PCL-(PDMAEMA)(4), while (PDMAPS)(4)-PCL-SS-PCL-(PDMAPS)(4) changed into PCL-(PDMAPS)(4). Background: Black patients have higher rates of hospitalization for acute heart failure than other race/ethnic groups. We sought to determine whether diuretic efficiency is associated with racial differences in risk for rehospitalization after acute heart failure. Methods: A post hoc analysis was performed on 721 subjects (age, 68 +/- 13 years; 22% black) enrolled in 3 acute heart failure clinical trials: ROSE-AHF (Renal Optimization Strategies Evaluation in Acute Heart Failure), DOSE-AHF (Diuretic Optimization Strategy Evaluation in Acute Decompensated Heart Failure), and CARRESS-HF (Cardiorenal Rescue Study in Acute Decompensated Heart Failure). Repeated-measures ANOVA was used to test for a racextime effect on measures of decongestion. Diuretic efficiency was calculated as net fluid balance per total furosemide equivalents. In a subset of subjects, Cox regression was used to examine the association between race and rehospitalization according to plasma renin activity (PRA). Results: Compared with nonblack patients, black patients were younger and more likely to have nonischemic heart failure. During the first 72 to 96 hours, there was greater fluid loss (P=0.001), decrease in NT-proBNP (N-terminal pro-B-type natriuretic peptide;P=0.002), and lower levels of PRA (P<0.0001) in black patients. Diuretic efficiency was higher in black than in nonblack patients (403 [interquartile range, 221-795] versus 325 [interquartile range, 154-698];P=0.014). However, adjustment for baseline PRA attenuated the association between black race and diuretic efficiency. Over a median follow-up of 68 (interquartile range, 56-177) days, there was an increased risk of all-cause and heart failure-specific rehospitalization in nonblack patients with increasing levels of PRA, while the risk of rehospitalization was relatively constant across levels of PRA in black patients. Conclusions: Higher diuretic efficiency in black patients with acute heart failure may be related to racial differences in activity of the renin-angiotensin-aldosterone system.
