Recently, interest in stimuli-responsive core-shell nanogels as drug delivery systems for tumor therapy has increased. Here, a temperature-activated drug locking and glutathione-triggered drug unlocking nanogel is designed, which is composed of hyaluronic acid (HA) conjugated with poly(N-isopropylacrylamide) (PNIPAAm) using a disulfide bond as the linker (HA-SS-PNIPAAm, H-SS-P). After injection into the systemic circulation, these synthetic copolymers endure temperature-motivated lock behaviors to form nanogels due to the thermosensitive lipophilic transformation of PNIPAAm, accompanied by doxorubicin (DOX) locking into the cavities of the nanogels. When they reach tumor cells, these nanogels exhibit glutathione (GSH)-triggered opening behavior to unlock the drugs for tumor therapy. The transmission electron microscopy (TEM) results demonstrate that the H-SS-P copolymer solutions are irregular at room temperature, while spherical structures (similar to 30 nm) can be observed below 37 degrees C, but dissociate in the presence of 40 mM GSH. Based on flow cytometry and fluorescence microscopy analyses, observations reveal that H-SS-P@DOX nanogels are intracellularly taken up into human lung cancer cells (A549) via HA-receptor mediated endocytosis. More importantly, these nanogels possess much higher tumor targeting capacity than free DOX and efficiently enhance the antitumor effect with reduced systemic toxicity in 4T1 tumor-bearing mice.

The study of multifunctional polymer micelles combined with chemotherapy due to reduced systemic toxicity and enhanced efficacy has attracted intensive attention. Herein, a multifunctional core-cross-linked hybrid micelle system based on mPEG-b-PGu(BA-TPE) and OCT-PEG-b-PGu(DA-TPE) with pH- and redox-triggered drug release and aggregation-induced emission (AIE) active imaging has been developed for active targeting of neuroendocrine neoplasms (NENs), especially neuroendocrine carcinomas (NECs) with poor prognosis. These micelles showed excellent biocompatibility and stability. After the formation of borate ester bonds, core-cross-linked micelles (CCLMs) showed enhanced emission properties. In addition, etoposide (ETO), one of the most important anticancer drugs of NECs, was loaded into the hydrophobic core of micelles by self-assembly with an average diameter of 274.6 nm and spherical morphology. Octreotide (OCT) conjugated onto the micelles enhanced cellular uptake by receptor-mediated endocytosis. ETO-loaded micelles demonstrated the dual-responsive triggered intracellular drug release and great tumor suppression ability in vitro. Compared with free ETO, ETO-loaded CCLMs exhibited a considerable antitumor effect and significantly reduced side effects. Considering the active tumor targeting, dual-responsive drug release and the AIE effect, the polymer micelle system will be a potential candidate for diagnosis and oncotherapy of NENs.

Glutathione (GSH) plays a vital role in maintaining biological redox homeostasis. Accordingly, accurate imaging of glutathione in vivo is of great significance. Herein, we propose a magnetic resonance energy transfer (MRET) strategy based on a distance-dependent magnetic exchange coupling effect (MECE), which can realize GSH detection within tumors in vivo by susceptibility weighted imaging (SWI). Fe3O4 nanoparticles (NPs) and CoFe2O4 NPs linked with cystamine (Fe3O4-S-S-CoFe2O4) have been successfully designed as SWI nanoprobes. After the disulfide bonds are broken by excess GSH in the tumor, the increase in the distance between Fe3O4 NPs and CoFe2O4 NPs will induce a decrease of MECE and magnetic susceptibility. As a result, the changes in the SWI signals are used for tumor GSH detection in vivo. Experimental results in vitro and in vivo demonstrate that the Fe3O4-S-S-CoFe2O4 SWI nanoprobe can sensitively detect concentrations of GSH in tumors. Hence, this strategy not only improves the sensitivity of the GSH response in SWI but also provides a powerful basis for the design of other responsive functional MRI nanoprobes.

Compared with liquid electrolytes, the solid polymer electrolyte (SPE), which possesses improved thermal and mechanical stability, is believed the broadest potential application for satisfying the safety needs of advanced electro-chemical devices. The completely healed SPE with extreme damage shows a high self-healing efficiency and no changes in the ionic conductivity and cycling performance of the solid-state lithium-metal/LiFePO4 cell compared to the pristine one.

Amphiphilic derivatives of (+/-)-trans-1,2-diselenane-4,5-diol (DSTox) decorated with long alkyl chains or aromatic substituents via ester linkages were applied as glutathione peroxidase (GPx)-like catalysts. The reduction of H2O2 with the diselenide catalysts was accelerated through a GPx-like catalytic cycle, in which the diselenide (Se-Se) bond was reduced to the diselenolate form ([Se-,Se-]) by coexisting dithiothreitol, and the generated highly active [Se-,Se-] subsequently reduced H2O2 to H2O retrieving the original Se-Se form. In the lipid peroxidation of lecithin/cholesterol liposomes induced by 2,2′-azobis(2-amidinopmpane) dihydrochloride (AAPH), on the other hand, the Se-Se form directly reduced lipid peroxide (LOOH) to the corresponding alcohol (LOH), inhibiting the radical chain reaction, to exert the antioxidative effect. Thus, the two GPx-like catalytic cycles can be switched depending on the peroxide substrates. Furthermore, hydrophilic compounds with no or short alkyl groups (C3) showed high antioxidative activities for the catalytic reduction of H2O2, while lipophilic compounds with long alkyl chains (C6-C14) or aromatic substituents were more effective antioxidants against lipid peroxidation. Polymers of intrinsic microporosity (PIMs) are a class of microporous polymers with rigid and contorted molecular structures. The special structures lead to incomplete space occupation, and the pores of PIMs originate from the formed void. In this work, we made attempts to expand the applications of PIMs to the aspect of heavy metal removal. First, PIM-1 was synthesized using tetrafluoroterephthalonitrile (TFTPN) and 5,5′,6,6′-tetrahydroxy-3,3,3′,3′-tetramethyl-1,1′-spirobisindane (TTSBI) as precursors. The primary PIM-1 was quite hydrophobic, and thus could not be evenly dispersed in water, resulting in low adsorption capacity for mercury ions (Hg2+). A conversion of nitrile group in PIM-1 to thiol group was then carried out by two steps, namely carboxylation and introduction of thiol groups. The carboxylation made the polymers more active, and the final thiol-functionalization provided the polymers with hydrophilicity and affinity for Hg2+. The thiolethyl modified PIM-1 (assigned as PIM-G) and thiophenyl modified (assigned as PIM-B) possessed maximum Hg2+ adsorption capacity of 136 mg g(-1) and 127 mg g(-1) at pH 5, respectively. Besides, the thiol-functionalized PIMs had fluorescence property and showed potential in sensing for Hg2+.

Polymer based core cross-linked nanoparticles (CCNPs) have generated a lot of interest as potential stimuli-responsive drug delivery systems. In the present work, we have synthesized smart redox-responsive water soluble polymeric CCNPs by cross-linking water-soluble PEG based copolymers with bis(acryloyl)cystamine via isoxazoline bond formation through a 1,3-dipolar cycloaddition reaction (click reaction) without using a copper catalyst, in a water-THF mixed solvent. The successful synthesis of CCNPs was confirmed by NMR, GPC and FT-IR measurements. Size distribution of the precursor copolymers and the CCNPs was determined by DLS measurement. AFM and FESEM images have confirmed globular morphology of these CCNPs. Their high stability in a physiological environment makes them effective as potent drug carriers with high loading capacity. MTT assays confirmed the biocompatibility of the synthesized CCNPs. Favourable cellular internalization of these DOX loaded CCNPs into cancer cells and redox-responsive release of DOX therefrom make these CCNP potentially smart vehicles to deliver anticancer drugs into cancer cells.

Background Turf toe injuries, though most common in athletes, can also occur in non-athletes. No study exists in the current literature investigating operative outcomes in non-athlete patients with chronic turf toe injury. In this study, we present our outcomes on operatively treated turf toe injuries in non-athletes in the only cohort yet studied. Methods Using ICD-10 codes, we assembled a cohort of 12 patients who underwent operative repair of chronic turf toe injury from January 2012 through January 2018 at the investigating institution. These 12 patients were evaluated to determine demographic information, method of injury, length of time from injury to surgery, clinical and radiologic characteristics of the injury, and operative outcomes including mean preoperative and postoperative VAS (Visual Analog Scale) scores, preoperative and postoperative FFI (Foot Function Index) scores, and postoperative complications. Results On initial clinical presentation, all 12 patients had local tenderness with associated painful range of motion. Four patients had restricted range of motion, all patients had a positive Lachman test, two had local edema, and eight had hallux valgus deformity. Mean VAS improved from 4.6 (range 2-9) to 1 (range 0-4). Mean FFI improved from 102.5 (range 56-177) to 61.75 (range 23-144). All patients had a negative Lachman test at final follow-up. No patients developed major complications or required revision surgery. Conclusions Our study is the first to investigate operative outcomes following chronic turf toe injury in non-athlete patients. Based on our study, surgeons and patients can expect significant improvement in overall pain and function following surgery.

Tedious polymeric modifications, laborious syntheses, low water solubility, high cytotoxicity and low quantum yields are still the major concerns related to fluorescent materials used for biosensing applications. Here we report one-pot facile synthesis of polyethylene glycol (PEG)-modified cystamine-based multifunctional fluorescent dyes named as CPEG-168, CPEG-1000 and CPEG-2000, which can be used for biosensing and detection of both cations and anions in pure aqueous medium. Rapid response toward thiols could be achieved due to the presence of disulfide functionality and reversible redox property between thiol and disulfide. A cyclic structure is formed by (NH-H) intramolecular hydrogen bonding to produce electron dense region for strong fluorescence emission. The presence of -NH group made them sensitive toward Cu2+ ions, while excellent sensitivity toward pH and F- ions was provided by intramolecular hydrogen bonding between -NH groups. The increase in chain length of PEGs enhanced the fluorescence emission intensities due to the formation of more rigid structure, and hence, the desired quantum yields can be tuned by changing the chain length of PEG. The aforementioned properties along with high brightness in aqueous solution, at biological pH and temperature range, make these fluorescent dyes potential candidates for biosensing applications and detection of cations and anions.

The CAS is 56-17-7. Through research, I have a further understanding and discovery of 2,2′-Disulfanediyldiethanamine dihydrochloride.

As a potential method for tumor treatment, tumor vaccine immunization induces a tumor-specific cellular immune response via immunization with tumor antigens. The delivery of exogenous antigen proteins into the cytoplasm of antigen-presenting cells is well known to induce an intensive cellular immune response for tumor treatment. In this work, we fluorinated a redox-responsive hyperbranched poly(amidoamine) (HPAA) with heptafluorobutyric anhydride to prepare a fluorinated HPAA (HPAA-F7) for use as a vaccine delivery system for antitumor therapy. The immunization results show that HPAA-F7 as a vaccine carrier could effectively promote the intracellular uptake and cytoplasmatic delivery of antigen proteins and induce potent antitumor cellular immunity. The novel vaccine carrier HPAA-F7 could be further developed for antitumor immunotherapy.
