Novel temperature-activated humidity-sensitive optical sensor

A novel, colorimetric, temperature-activated humidity indicator is presented, with a colour change based on the semi-reversible aggregation of thiazine dyes (esp. methylene blue, MB) encapsulated within the polymer, hydroxypropyl cellulose (HPC). The initially purple MB/HPC film is activated by heat treatment at 370 degrees C for 4 s, at which point the film (with a colour associated with that of a highly aggregated form of MB; lambda(max) = 530 nm) becomes blue (indicating the presence of monomeric and dimeric MB; i.e. with lambda(max) = 665; 605 nm respectively). The blue, heat-treated MB/HPC films respond to an ambient environment with a relative humidity (RH) exceeding 70% at 21 degrees C within seconds, returning to their initial purple colour. This colour change is irreversible until the film is heat-treated once more. When exposed to a lower RH of up to ca. 47%, the film is stable in its blue form. In contrast, a MB/HPC film treated only at 220 degrees C for 15 s also turns a blue colour and responds in the same way to a RH value of ca. 70%, but it is unstable at moderate RH 37-50% values, so that it gradually returns to its purple form over a period of approximately 6 h. The possible use of the high heat-treated MB/HPC humidity indicator in the packaging of goods that cannot tolerate high RH, such as dry foods and electronics, is discussed. (C) 2016 Elsevier B.V. In the crystal, two sets of weak C-HPh center dot center dot center dot ODbt (Ph = phenyl and Dbt = dihydrobenzothiazine) hydrogen bonds form layers of molecules parallel to the bc plane. The layers stack along the aaxis direction with intercalation of the ester chains. The crystal studied was a two component twin with a refined BASF of 0.34961 (5). The Hirshfeld surface analysis of the crystal structure indicates that the most important contributions to the crystal packing are from H center dot center dot center dot H (37.5%), H center dot center dot center dot C/C center dot center dot center dot H (24.6%) and H center dot center dot center dot O/O center dot center dot center dot H (16.7%) interactions. Hydrogen-bonding and van der Waals interactions are the dominant interactions in the crystal packing. Computational chemistry indicates that in the crystal, C-H-Ph center dot center dot center dot O-Dbt hydrogen bond energies are 38.3 and 30.3 kJ mol(-1). Density functional theory (DFT) optimized structures at the B3LYP/ 6-311 G(d,p) level are compared with the experimentally determined molecular structure in the solid state. The syntheses and crystal structures of 2,3-bis[3-(trifluoromethyl)phenyl]-2,3dihydro-4H-1,3-benzothiazin-4-one (di-m-CF3; C22H13F6NOS) (1) and 2,3-bis(4-methylphenyl)-2,3-dihydro-4H-1,3-benzothiazin-4-one (di-p-CH3; C22H9NOS) (2) are reported. Therapeutic effects of an azaphenothiazine derivative in mouse experimental colitis

Phenothiazines represent a class of compounds of potential therapeutic utility. In this report we evaluated therapeutic value of an azaphenothiazine derivative, 6-acetylaminobutyl-9-chloroquino[3,2-b]benzo[1,4]thiazine (QBT), given intragastrically, in the model of dextran sodium sulfate-induced colitis in C57BL/6 mice using 5-aminosalicylic acid (5-ASA) as a reference drug. Colitis symptoms such as body weight loss, diarrhea and hematochezia (blood in stool) were observed and registered and disease activity index (DAI) was calculated. In addition, weight and cell numbers in the lymphatic organs and histological parameters of the colon wall were analyzed. The effects of QBT on viability of colon epithelial cell lines were also determined. We showed that weight and cell number of draining mesenteric lymph nodes were lower in mice treated with QBT in comparison to their control counterparts. The number of thymocytes, drastically reduced in control mice, was elevated in mice treated with the compounds with a significant effect of 5-ASA. In addition, an abnormal composition of blood cell types was partially corrected in these groups. Histological analysis of the colon revealed that the pathological changes were partially normalized by QBT and even to a higher degree by 5-ASA. In conclusion we demonstrated a therapeutic efficacy of the compound in amelioration of local and systemic pathological changes associated with chemically-induced colitis in mice. A possible mechanism of action of the compound is discussed.

Synthesis, Characterization and Molecular Docking of Novel Bioactive Thiazolyl-Thiazole Derivatives as Promising Cytotoxic Antitumor Drug

Reactions of ethylidenethiocarbohydrazide with hydrazonoyl halides gave 1,3-thiazole or 1,3,4-thiadiazole derivatives according to the type of hydrazonoyl halides. Treatment of ethylidenethiosemicarbazide with hydrazonoyl halides and dimethylacetylene dicarboxylate (DMAD) afforded the corresponding arylazothiazoles and 1,3-thiazolidin-4-one derivatives, respectively. The structures of the synthesized products were confirmed by IR, H-1-NMR, C-13-NMR and mass spectral techniques. The cytotoxic activity of the selected products against the Hepatic carcinoma cell line (Hepg-2) was determined by MTT assay indicating a concentration dependent cellular growth inhibitory effect, especially for compounds 14c and 14e. The dose response curves indicated the IC50 (the concentration of test compounds required to kill 50% of cell population) were 0.54 M and 0.50 M, respectively. Confocal laser scanning imaging of the treated cells stained by Rhodamin 123 and Acridine orange dyes confirmed that the selected compounds inhibit the mitochondrial lactate dehydrogenase enzymes. The binding mode of the active compounds was interpreted by a molecular docking study. The obtained results revealed promising cytotoxic activity.

Decolorization of heterocycle dye Neutral Red by white-rot fungus Perenniporia subacida

A white-rot fungus Perenniporia subacida was developed for decolorizing twelve structurally various dyes from anthraquinone, azo, heterocycle, thiazine, and triphenylmethane groups. Among them, heterocycle dye Neutral Red with preferable decolorization performance was selected for further experiments and this strain could be utilized sequentially for three consecutive cycles with declining decolorization (%). After a 10-day incubation period, higher dye uptake (96.56%) was obtained at constant inoculum and agitation speed with the optimum physicochemical parameters like initial pH at 4.0, temperature at 35 degrees C, initial dye concentration at 100 mg/L, and ionic strength at 0.1 mol/L. Noteworthy induction of various dye decolorizing enzymes viz. lignin peroxidase, laccase, manganese peroxidase, tyrosinase, and nicotinamide-adenine dinucleotide hydrogen-2,6-dichlorophenol indophenol reductase compared to control, point out toward their involvement in overall decolorization and degradation process. Analytical studies like fourier transform infrared spectroscopy and gas chromatography-mass spectroscopy were used to identify the degraded metabolites and scrutinize the degradation process. Phytotoxicity studies indicated that the fungal treatment favors detoxification of dye Neutral Red. It is suggested that P. subacida has great potential for decolorizing heterocycle dyes.

SYNTHESIS OF SOME NOVEL THIAZOLE, THIADIAZOLE AND 1,4-PHENYLENE-BIS-THIAZOLE DERIVATIVES AS POTENT ANTITUMOR AGENTS

A novel series of 2-ethylidenehydrazono-5-arylazothiazoles 5a-h and 2-ethylidenehydrazono-5-arylazothiazolones 9a-d were prepared by cyclocondensation of hydrazonyl halides 3a-h and 7a-d with ethylidenethiosemicarbazide 2. In addition, reaction of 2 with N-phenyl-carbohydrazonyl chloride (14), afforded 1,3,4-thiadiazole derivative 17 as the end product. Moreover, the thiosemicarbazide derivative 2 was reacted with various bromoacetyl compounds 19a-d and 1,1′-(1,4-phenylene)bis(2-bromoethanone) (21) furnished the respective thiazole derivatives 20a-d and 1,4-phenylene-bis-thiazole derivative 22. The structures of the newly synthesized compounds were established on the basis of spectroscopic evidences and their alternative syntheses. The newly synthesized compounds were evaluated for their antitumor activities against hepatocellular carcinoma (HepG2) cell line and the results revealed promising activities of compounds 5h, 5d, 5g, 5f and 5e with IC50 equal 2.23 +/- 0.28, 2.48 +/- 0.34, 2.49 +/- 0.24, 4.03 +/- 0.11, and 5.32 +/- 0.27 mu M, respectively.

Spectral and kinetic features of nonradiative energy transfer in hybrid associates of colloidal quantum dots and organic dyes

The features of nonradiative resonance energy transfer in hybrid associates of colloidal CdS quantum dots (QDs) with molecules of a thiazine dye and J-aggregates of a carbocyanine dye are discussed. The case of an inhomogeneously broadened recombination band of donor (CdS QDs) luminescence is considered. We detected spectral-selective quenching of recombination luminescence of colloidal CdS QDs in association with organic dyes. The maximum of quenching occurs in the region of absorption of the acceptor (organic dye molecule). It is shown using the time-correlated single photon counting technique that the lifetime of the QD luminescence decreases synchronously with the quenching of the luminescence, but only at the wavelengths falling in the region of absorption of the dye. Based on the concept of a donor-acceptor nature of emission of CdS QDs, the dependence of the efficiency of nonradiative energy transfer on the luminescence wavelength is analyzed taking into account the Poisson distribution of its quenchers.
