The four-arm star copolymers poly(methacrylic acid)-poly(2-hydroxyethyl methacrylate-disulfide similar to)-poly(poly(ethylene glycol) methyl ether methacrylate) (4AS-PMAA(x)-(PHEMA-SS similar to)(y)-PPEGMA(z)) with four different block ratios were synthesized and could self-assembled into cross-linked polymer micelles for the exploration of the structure-property relationship. The cross-linked polymer micelles in aqueous solution had low critical micelle concentration (CMC) values (1.9-4.6 mg/L), which exhibited better stability than non-cross-linked micelles. The CMC value decreased with the increase of the length of inner PMAA core and hydrophobic PHEMA cross-linked middle layer. The blank and doxorubicin (DOX)-loaded micelles with different block ratios were prepared by dialysis with the particle sizes of 120-240 nm. The longer inner PMAA core and cross-linked middle layer enhanced the drug loading content (DLC) results and led to relatively bigger particle sizes of polymer micelles. The in vitro DOX release data revealed that DOX-loaded micelles had low DOX cumulative release percentages of 18-37% after 110 h at pH 7.4, but up to 83-90% when introducing reductant GSH at pH 5.0. The 4AS-PMAA(21.2)-(PHEMASS approximate to)(13.1)-PPEGMA(5.1) micelles with the longest PMAA core had the largest cumulative release of 90.1%. The DOX release process and mechanism of the micelles at different conditions fitted well with the semi-empirical equation. Overall, the results demonstrated that the block ratios and pH/redox-responsiveness of these four-arm star copolymers could be well-controlled and their self-assembled cross-linked micelles as anticancer drug carrier system could be improved by optimizing the different ratios.

Objective: To determine the pattern of presentations and characteristics of patients at a psychiatric emergency services facility. Methods: The cross-sectional study was conducted in November 2016 at the Department of Psychiatry and Behavioural Sciences, Jinnah Postgraduate Medical Centre, Karachi, and comprised all patients presenting to the departmental emergency services. Data was documented on a pre-designed semi-structured proforma, and analysed using SPSS 22. Results: Of the 316 cases, 162(51.3%) were males and 154(48.7%) were females. The overall mean age was 30.78+/-13.09 years (range: 8-80 years). Psychiatric emergencies comprised suicidal attempt, excitement, violence, altered sensorium, altered/inappropriate behaviour and extrapyramidal symptoms / acute dystonia together constituting for 123(38.9%) of the total presentations, while there were 20(6.33%) cases of hysterical fits and 6(1.89%) with acute anxiety symptoms. Overall, 56(17.7%) subjects were found to have no psychiatric diagnosis, and 46(81.5%) of these were referred cases. Conclusion: Non-emergency visits constituted high percentage of emergency presentation.

A water-soluble cyclophane dimer having two disulfide groups as a reduction-responsive cleavable bond as well as several acidic and basic functional groups as a pH-responsive ionizable group 1 was successfully synthesized. It was found that 1 showed pH-dependent guest-binding behavior. That is, 1 strongly bound an anionic guest, 6-p-toluidinonaphthalene-2-sulfonate (TNS) with binding constant (K/M-1) for 1:1 host-guest complexes of 9.6 x 10(4) M-1 at pH 3.8, which was larger than those at pH 7.4 and 10.7 (6.0 x 10(4) and 2.4 x 10(4) M-1, respectively), indicating a favorable electrostatic interaction between anionic guest and net cationic 1. What is more, release of the entrapped guest molecules by 1 was easily controlled by pH stimulus. Large favorable enthalpies (Delta H) for formation of host-guest complexes were obtained under the pH conditions employed, suggesting that electrostatic interaction between anionic TNS and 1 was the most important driving force for host-guest complexation. Such contributions of Delta H for formation of host-guest complexes decreased along with increased pH values from acidic to basic solutions. Upon addition of dithiothreitol (DTT) as a reducing reagent to an aqueous PBS buffer (pH 7.4) containing 1 and TNS, the fluorescence intensity originating from the bound guest molecules decreased gradually. A treatment of 1 with DTT gave 2, having less guest-binding affinity by the cleavage of disulfide bonds of 1. Consequently, almost all entrapped guest molecules by 1 were released from the host. Moreover, such reduction-responsive cleavage of 1 and release of bound guest molecules was performed more rapidly in aqueous buffer at pH 10.7.

Designed oligourea sequences with an amphipathic distribution of Arg- and His-type residues were shown to form tight complexes with plasmid DNA, and to effectively promote the release of DNA from the endosomes. Herein, we report the synthesis of new cell-penetrating foldamer sequences in which the foldamer segment is conjugated via a reducible disulfide bond to a ligand that binds cell-surface expressed nucleoproteins with the idea that this system could facilitate both assemblies with nucleic acids and cell entry. This new system was evaluated for delivery of DNA in several cell lines and was found to compare favorably with all comparators tested (DOTAP and b-PEI as well as a number of known cell penetrating peptides) in various cell lines and particularly in culture medium containing up to 50% of serum. These results suggest that this dual molecular platform which is long lasting and noncytotoxic could be of practical use for in vivo applications.

Polymer based core cross-linked nanoparticles (CCNPs) have generated a lot of interest as potential stimuli-responsive drug delivery systems. In the present work, we have synthesized smart redox-responsive water soluble polymeric CCNPs by cross-linking water-soluble PEG based copolymers with bis(acryloyl)cystamine via isoxazoline bond formation through a 1,3-dipolar cycloaddition reaction (click reaction) without using a copper catalyst, in a water-THF mixed solvent. The successful synthesis of CCNPs was confirmed by NMR, GPC and FT-IR measurements. Size distribution of the precursor copolymers and the CCNPs was determined by DLS measurement. AFM and FESEM images have confirmed globular morphology of these CCNPs. Their high stability in a physiological environment makes them effective as potent drug carriers with high loading capacity. MTT assays confirmed the biocompatibility of the synthesized CCNPs. Favourable cellular internalization of these DOX loaded CCNPs into cancer cells and redox-responsive release of DOX therefrom make these CCNP potentially smart vehicles to deliver anticancer drugs into cancer cells.

Introducing clickable groups onto polymer microspheres allows the microspheres to be facilely functionalized via a click reaction. In this report, we have synthesized monodisperse PMMA microspheres with clickable double bonds on the surface by one-step RAFT-assisted dispersion photopolymerization using a cyclohexyl-containing macro-RAFT agent as a stabilizer. This macro-RAFT agent, named P(THPAA-b-AA)-TTC, was synthesized by RAFT-mediated block copolymerization of a cyclohexyl-containing monomer (THPAA, obtained by esterification of cis-1,2,3,6-tetrahydrophthalic with 2-hydroxyethyl acrylate) and acrylic acid. The obtained microspheres were analyzed by H-1 NMR and Iodometric method, indicating that the content of carbon double bonds on the microspheres increased with increasing the amount of P(THPAA-b-AA)-TTC in the polymerization feed. We then used a thiolated biotin to perform the photo-click reaction with the cyclohexene double bond on the micro spheres, thereby attaching the biotin unit onto the microspheres. The biotinylated PMMA microspheres were found to be able to effectively immobilize fluorescein isothiocyanate streptavidin (FITC-SAv) via the specific adsorption of biotin to streptavidin, and thus exhibited clear fluorescence under confocal microscope. This study provides a convenient and feasible way to prepare surface-functionalized microspheres, which has great potential in biological applications as well as other fields.

Tedious polymeric modifications, laborious syntheses, low water solubility, high cytotoxicity and low quantum yields are still the major concerns related to fluorescent materials used for biosensing applications. Here we report one-pot facile synthesis of polyethylene glycol (PEG)-modified cystamine-based multifunctional fluorescent dyes named as CPEG-168, CPEG-1000 and CPEG-2000, which can be used for biosensing and detection of both cations and anions in pure aqueous medium. Rapid response toward thiols could be achieved due to the presence of disulfide functionality and reversible redox property between thiol and disulfide. A cyclic structure is formed by (NH-H) intramolecular hydrogen bonding to produce electron dense region for strong fluorescence emission. The presence of -NH group made them sensitive toward Cu2+ ions, while excellent sensitivity toward pH and F- ions was provided by intramolecular hydrogen bonding between -NH groups. The increase in chain length of PEGs enhanced the fluorescence emission intensities due to the formation of more rigid structure, and hence, the desired quantum yields can be tuned by changing the chain length of PEG. The aforementioned properties along with high brightness in aqueous solution, at biological pH and temperature range, make these fluorescent dyes potential candidates for biosensing applications and detection of cations and anions.

We present a sensitive single step nanobiosensor for thyroid disease diagnosis. Nanoparticles modified screen printed carbon electrode catalyzes the formation of peptide bond between anti-TSH antibody and amino coated gold nanoparticles rendering a covalently coupled antibody. The nanobiosensor detects and quantifies the thyroid stimulating hormone in the sample by sensing the effective resistance offered by electrode. As the TSH concentration increases in serum sample, more is the immunocomplex formed and higher is the resistance offered by electrode. Gold nanoparticles functionalization with cystamine dihydrochloride facilitates a covalent bonding between surface amino group and carboxylic Fc ensuring maximizing available active antibody. This strategy ensures a much lower limit of detection in addition to improved detection range due to increased loading capacity as a result of larger effective surface area offered by gold nanoparticles. These two aspects of immunosensor fabrication resulted in limit of detection as low as 0.001 mu IU/mL and an enhanced detection range of 0.001-150 mu IU/mL. This makes the developed immunosensor suitable for diagnostic purpose covering the clinically relevant range and a simple detection technique makes it potential candidate for fabrication of a Point-of-Care device for detection of Thyroid dysfunctioning. Introducing clickable groups onto polymer microspheres allows the microspheres to be facilely functionalized via a click reaction. In this report, we have synthesized monodisperse PMMA microspheres with clickable double bonds on the surface by one-step RAFT-assisted dispersion photopolymerization using a cyclohexyl-containing macro-RAFT agent as a stabilizer. This macro-RAFT agent, named P(THPAA-b-AA)-TTC, was synthesized by RAFT-mediated block copolymerization of a cyclohexyl-containing monomer (THPAA, obtained by esterification of cis-1,2,3,6-tetrahydrophthalic with 2-hydroxyethyl acrylate) and acrylic acid. The obtained microspheres were analyzed by H-1 NMR and Iodometric method, indicating that the content of carbon double bonds on the microspheres increased with increasing the amount of P(THPAA-b-AA)-TTC in the polymerization feed. We then used a thiolated biotin to perform the photo-click reaction with the cyclohexene double bond on the micro spheres, thereby attaching the biotin unit onto the microspheres. The biotinylated PMMA microspheres were found to be able to effectively immobilize fluorescein isothiocyanate streptavidin (FITC-SAv) via the specific adsorption of biotin to streptavidin, and thus exhibited clear fluorescence under confocal microscope. This study provides a convenient and feasible way to prepare surface-functionalized microspheres, which has great potential in biological applications as well as other fields.
