3,6-Diazaphenothiazines as potential lead molecules – synthesis, characterization and anticancer activity

3,6-Diazaphenothiazines were obtained in cyclization of 3-amino-3′-nitro-2,4′-dipyridinyl sulfide and the reaction of sodium 3-amino-2-pyridinethiolate with 4-chloro-3-nitropyridine followed by alkylation and heteroarylation. The thiazine ring formation ran via the Smiles rearrangement. The structure elucidation was based on 2D NMR and X-ray analysis of N-methylated product. 3,6-Diazaphenothiazines were investigated for antitumor activity using glioblastoma SNB-19, melanoma C-32 and breast cancer MCF-7 cells. 10H-3,6-diazaphenothiazine was 10 times more active (IC50 < 0.72 mu g/mL) than cisplatin. Two diazaphenothiazines with the 2-pyrimidinyl and dimethylaminopropyl substituents were selectively active against MCF-7 and C-32 cells. The expressions of H3 (proliferation marker), TP53, CDKN1A (cell cycle regulators), BAX and BCL-2 (proapoptopic and antiapoptopic genes) were detected by RT-QPCR method. Synthesis and Structural Properties of N-(2-bromo-4-nitmphenyl)-3-methoxy-4-oxo-3,4-dihydro-2H-benzole 1,2] thiazine-3-carboxamide 1,1-dioxide: A Comparative Experimental and Quantum Chemical Study

N-(2-bromo-4-nitrophenyl)-3-methoxy-4-oxo-3,4-dihydro-2H-benzo[e] [1,2] thiazine-3-carboxamide-1,1-dioxide was synthesized in three step process with 86 % overall yield. The final structure of compound was evaluated by using spectroscoanalytical methods (H-1-NMR and FT-IR). Suitable crystals were obtained by slow evaporation method, and the final structure was confirmed unequivocally by performing single crystal X-ray diffraction (XRD) studies. Geometric parameters were calculated at B3LYP/6-3 1G (d, p) method with the help of Gaussian 09 software to validate spectrosopic and single crystal X-ray results. The computed data corroborated nicely with the experimental results (spectroscopic and X-ray). Frontier molecular orbitals (FMOs) and reactivity indices revealed the reactivity of benzothiazine derivative. Molecular electrostatic potential (MEP) was measured to understand the electro or nucleophilic nature of compound. Mulliken and natural population charge analysis (NBO) was carried out to prove inter and intramolecular hydrogen bonding.

Synthesis and Characterization of Some New Bis-Pyrazolyl-Thiazoles Incorporating the Thiophene Moiety as Potent Anti-Tumor Agents

A new series of 1,4-bis(1-(5-(aryldiazenyl)thiazol-2-yl)-5-(thiophen-2-yl)-4,5-dihydro-1H-pyrazol-3-yl)benzenes 3a-i were synthesized via reaction of 5,5-(1,4-phenylene)bis(3-(thiophen-2-yl)-4,5-dihydro-1H-pyrazole-1-carbothioamide) (1) with hydrazonoyl halides 2a-i. In addition, reaction of 1 with ethyl chloroacetate afforded bis-thiazolone derivative 8 as the end product. Reaction of compound 8 with methyl glyoxalate gave bis-thiazolone derivative 10. The structures of the newly synthesized compounds were established on the basis of spectroscopic evidences and their alternative syntheses. All the synthesized compounds were evaluated for their anti-tumor activities against hepatocellular carcinoma (HepG2) cell lines, and the results revealed promising activities of compounds 3g, 5e, 3e, 10, 5f, 3i, and 3f with IC50 equal 1.37 +/- 0.15, 1.41 +/- 0.17, 1.62 +/- 0.20, 1.86 +/- 0.20, 1.93 +/- 0.08, 2.03 +/- 0.25, and 2.09 +/- 0.19 M, respectively.

Environment effects on the nonlinear absorption properties of Methylene blue under different power of excitation beam

In this paper, the nonlinear optical absorption properties of Methylene blue solutions were studied by open aperture Z-scan technique under a continuous-wave laser irradiation with various input powers. The experimental results show that the mechanism of nonlinear absorption and the calculated coefficients depend highly onthe molecular surrounding media properties and the nature of intermolecular interactions. From the experimental results, the contributions of various intermolecular interactions were determined as well. Exploiting these results, it is possible to determine the effective media polarity parameters on the nonlinear responses of optical materials. According to the experimental results, by increasing of excitation beam power, the strong hydrogen bonds with high contributions increase the third order nonlinear responses of Methylene blue molecules. Removal of Synthetic Dyes by Dried Biomass of Freshwater Moss Vesicularia Dubyana: A Batch Biosorption Study

In this work the biosorption of cationic dyes thioflavin T (TT) and methylene blue (MB) from single and binary solutions on dried biomass of freshwater moss Vesicularia dubyana as a function of contact time, pH, and biomass or sorbate concentration has been investigated. The prediction of maximum sorption capacities using adsorption isotherm models were also realized. Biosorption of TT and MB is a rapid process strongly affected by solution pH. Maximum sorption capacities Q(max) calculated from Langmuir isotherm were 119 +/- 11 mg/g for TT and 229 +/- 9 mg/g for MB. In binary mixture, the presence of MB caused significant decrease of TT sorption, advocating the competitive sorption between TT and MB. Results revealed that V. dubyana biomass exhibited significantly higher affinity to thiazine dye MB in comparison with benzothiazole dye TT from both single and binary solutions. Based on the obtained results, the competitive effects in binary system can substantially influence the sorption process and should be thoroughly evaluated before application of selected adsorbents for removal of basic dyes from colored effluents.

10H-1,9-diazaphenothiazine was obtained in the sulphurisation reaction of diphenylamine with elemental sulphur and transformed into new 10-substituted derivatives, containing alkyl and dialkylaminoalkyl groups at the thiazine nitrogen atom. The 1,9-diazaphenothiazine ring system was identified with advanced H-1 and C-13 NMR techniques (COSY, NOESY, HSQC and HMBC) and confirmed by X-ray diffraction analysis of the methyl derivative. The compounds exhibited significant anticancer activities against the human glioblastoma SNB-19, melanoma C-32 and breast cancer MDA-MB-231 cell lines. The most active 1,9-diazaphenothiazines were the derivatives with the propynyl and N, N-diethylaminoethyl groups being more potent than cisplatin. For those two compounds, the expression of H3, TP53, CDKN1A, BCL-2 and BAX genes was detected by the RT-QPCR method. The proteome profiling study showed the most probable compound action on SNB-19 cells through the intrinsic mitochondrial pathway of apoptosis. The 1,9-diazaphenotiazine system seems to be more potent than known isomeric ones (1,6-diaza-, 1,8-diaza-, 2,7-diaza- and 3,6-diazaphenothiazine). [GRAPHICS] .

Synthesis and biological evaluation of coumarin clubbed thiazines scaffolds as antimicrobial and antioxidant

A new series of 4-methyl-6-nitro-2-oxo-2H-chroman-7yl-2-(4-(4-fluorophenyl)-6-phenyl-2H-1,3-thiazin-2-yl-amino)acetates 5a-j were synthesized from 6-nitro-4-methyl coumarinyl chloroacetate (5) and 2-amino thiazines (IIIa-j). Final compounds were screened for their antimicrobial, antitubercular, and antioxidant activities. Compounds 5c and 5h found to have antibacterial potency against E. coli with MIC values 50 mu g/mL compared to standard drugs. Compound 5d demonstrated better antifungal potency (MIC = 200 mu g/mL) against C. albicans when compared with griseofulvin. Compounds 5b and 5h found to be encouraging antitubercular (MIC = 62.5 mu g/mL with 98-99% inhibition) against M. tuberculosis H(37)Rv. The newly synthesized 5h and 5b were appeared to have high radical scavenging efficacies as 33.99 +/- 0.301 and 35.35 +/- 0.470 mu g/mL +/- SD of IC50 values, respectively, in DPPH and ABTS bioassay.
