Stereoselective Synthesis and Cytoselective Toxicity of Monoterpene-Fused 2-Imino-1,3-thiazines

Starting from pinane-, apopinane- and carane-based 1,3-amino alcohols obtained from monoterpene-based beta-amino acids, a library of monoterpene-fused 2-imino-1,3-thiazines as main products and 2-thioxo-1,3-oxazines as side-products were prepared via two-or three-step syntheses. When thiourea adducts prepared from 1,3-amino alcohols and aryl isothiocyanates were reacted with CDI under mild conditions, O-imidazolylcarbonyl intermediates were isolated which could be transformed to the desired 1,3-thiazines under microwave conditions. 1,3-Thiazines and 2-thioxo-1,3-oxazine side-products could also be prepared in one-step reactions through the application of CDI and microwave irradiation. The ring-closure process was extended to cycloalkane-based gamma-hydroxythioureas. The carane-and apopinane-based derivatives exhibited marked antiproliferative activity against a panel of human adherent cancer cell lines (HeLa, A2780, MCF7 and A431).

Sol-gel (template) synthesis of macroporous Mo-based catalysts for hydrothermal oxidation of radionuclide-organic complexes

Molybdenum compounds are industrially demanding as heterogeneous catalysts for oxidation of various organic substances. Highly porous structure of molybdenum-containing catalysts avoids surface’s colmatation and prevents blocking catalytic sites that makes these materials play a key role in processes of hydrothermal oxidation of radionuclide organic complexes. The study presents an original way of sol-gel synthesis of new macroporous molybdenum compounds using core-shell colloid template (polymer latex) as poreforming agent. We have described three individual routs of template removal via thermal decomposition to obtain porous materials based on molybdenum compounds. Thermal treatment conditions (temperature, gaseous atmosphere) have been studied with respect to their influence on composition, structure and catalytic properties of synthesized molybdenum systems. The optimal way to synthesis of crystal molybdenum (VI) oxide with ordered porous structure (mean pore size 100-160 nm) has been suggested. Catalytic properties of macroporous molybdenum materials have been investigated in the process of liquid phase and hydrothermal oxidation of such organic substances thiazine and stable Co-EDTA complex. It was shown that macroporous molybdenum oxides could be applied as prospective catalysts for hydrothermal oxidation of organic radionuclide complexes during the processing of radioactive waste. Synthesis of 2,5-Disubstituted Derivatives of Pyrano[2,3-d][1,3]thiazines via the Interaction of 2-Substituted 4-Hydroxy-6H-1,3-thiazine-6-ones with Aldehydes

A new and efficient route for the synthesis of derivatives of the poorly investigated pyrano[2,3-d][1,3]thiazine heterocyclic system is disclosed. These compounds were prepared via annulation of 2-aryl-4-hydroxy-6H-1,3-thiazine-6-ones with aliphatic and aromatic aldehydes in the presence of pyridine. The method is general and versatile, and the interaction is independent on the nature of the aldehyde, the only exceptions being formaldehyde and salicylaldehydes.

Mortino (Vaccinium floribundum Kunth) berry assisted green synthesis and photocatalytic performance of Silver-Graphene nanocomposite

Mortitio (Vacciniumf/oribundum Kunth) is a wild shrub native to South America, whose berries are widely consumed in Ecuador as fresh fruit, juice and jam. We reported a green synthesis of Silver-Graphene (Ag-G) nanocomposite through the reduction of Ag+ and graphene oxide using berry extract of Morthio. Silver nanoparticles were anchored onto the graphene sheets by ultrasonication and solar light irradiation. The as-prepared 4g-G nanocomposite was characterized by Fourier transform infrared spectroscopy, UV-vis spectroscopy, Scanning transmission electron microscopy, Scanning electron microscopy with energy dispersive X-ray spectrometry, and X-ray diffraction techniques. The photocatalytic activities of the Ag-G nanocomposite were evaluated by the degradation of methylene blue (MB) and methylene orange (MO) in an aqueous medium under sunlight irradiation. The photocatalytic activity of Ag-G nanocomposite for the degradation of MB (k = 0.0163283 min(-1)) > MO (k=0.0140985 min(-1)), due to the anchoring of Ag nanoparticles on the graphene sheets and thiazine ring of MB is more susceptible to photodegradation than the azo bond of MO. This ecofriendly and green reduction method avoids the use of toxic reagents, and it might be useful for biocompatible materials in the future engineering applications. (C) 2016 Elsevier B.V. An efficient regioselective synthesis of 4-iodo-2,3-disubstituted-2H-thieno[3,2-el[1,2]thiazine-1,1-dioxide derivatives via iodocyclization approach using iodine under mild reaction condition described herein. This coupling iodocyclization strategy tolerated a variety of functional groups such as alkyl, cycloalkyl, phenyl producing the six-membered heterocyclic ring selectively. The resulting 4-iodo-2,3disubstituted-2H-thieno[3,2-e][1,2]thiazine-1,1-dioxide was coupled with a variety of boronic acids (Suzuki coupling) and activated alkenes (Heck coupling). The iodo group was utilized for Sonogashira coupling followed by efficient transformation to azido precursor, which was used for the synthesis of various thieno-sultam linked with triazole. Reinvestigations into synthesis of allyldithiocarbamates and their intramolecular cyclization: synthesis and antihyperglycemic activity of 2-thioxothiazolidine-4-alkanoates

Reinvestigating the synthesis of allylthiocarbamates from MBH derivatives revealed that they are obtained more efficiently from allylbromides in isopropanol rather than acetates in water. Further unlike reported, the base-mediated intramolecular cyclization of these allyldithiocarbamates give substituted 2-thioxothiazolidine-4-alkanoates instead of substituted 1,3-thiazines and the reaction was restricted to 2- or 4-nitrophenyl carrying substrates. Screening of these products for antihyperglycemic activity revealed that 5bC displays significant activity via partial agonist activity on PPAR gamma at transcriptional level, which was corroborated by molecular docking studies. Synthesis, biological activity, distribution and membrane permeability of novel spiro-thiazines as potent neuroprotectors

New spiro-derivatives of 1,3-thiazine-potential neuroprotectors have been synthesized. It has been determined that the obtained compounds are biologically active and capable of blocking the glutamate-induced calcium ion uptake into synaptosomes of rat brain cortex. The inhibitory activity of the test substances was shown to depend on the chemical nature and structure of the substituents bound with an exocyclic nitrogen atom. Non-polar alkyl and polar radicals with halogen, oxygen and nitrogen atoms were used as substituents. It is typical of the active spiro-thiazines to have alkyl substituents in ortho-and para-position of the benzene ring. Among the investigated spiro-thiazines it is the derivatives with ethyl- and isopropyl-groups in the aril part of the molecules that are the lead-compounds with a high inhibitory ability. We measured the distribution coefficients of the substances in octanol/buffer and hexane/buffer systems and made conclusions about the ability of the investigated drug-like compounds to penetrate the biological membranes. By using the parabolic model we derived a quadratic equation that allowed us to evaluate quantitatively the inhibitory activity of spiro-thiazines with hydrophobic substituents based on lipophilicity data. We also studied the permeability through the phospholipidic membrane and introduced a correlation equation describing the dependence of the investigated spiro-thiazines activity on the descriptors characterizing the donor acceptor properties.
