A convenient synthesis of the symmetrical bis(difluoromethyl)-1,4-thiazine 1,1-dioxides was carried out via heterocyclization of bis(2,2,3,3-tetrafluoropropyl)sulfone with ammonia and primary amines. The reaction with excess of ammonia leads to the formation of 3,5-bis(difluoromethyl)-4H-1,4-thiazine 1,1-dioxide. The result of the reaction with primary amines is determined by spatial nature of substituent at the nitrogen atom. 3,5-Bis(difluoromethyl)-4-propyl-4H-1,4-thiazine 1,1-dioxide and 3,5-bis(difluoromethyl)-4-(p-tolyl)-4H-1,4-thiazine 1,1-dioxide were prepared by the reactions of bis(2,2,3,3-tetrafluoropropyl)sulfone with n-propylamine and p-toluidine accordingly. Stereoselective Synthesis and Antiproliferative Activity of Monoterpene-Fused 2-Imino-1,3-oxazines

Background: In the recent years the 2-imino-1,3-thiazine and 2-iminothiazolidine ring systems can be found as moieties in biologically relevant compounds, including BACE1 inhibitors, or cannabinoid receptor agonists, while monoterpene-based 2-imino-1,3-thiazines, prepared from chiral 1,3-amino alcohols exhibiting pronounced antiproliferative activity. Methods: The antiproliferative activities of the prepared compounds were determined in vitro against a panel of human adherent cancer cell lines including HeLa, MCF7 and A431 by MTT assay. Results: Starting from pinane-, apopinane-and carane-based beta-amino acid derivatives, 1,3-amino alcohols were prepared via two-step syntheses. The reactions of the product 1,3-amino alcohols and aryl isothiocyanates yielded gamma-hydroxythioureas, which were transformed to monoterpene-fused 2-imino-1,3-oxazines via base-catalysed ring closure. The antiproliferative activities of these 2-imino-1,3-oxazines were examined and the structure-activity relationships were studied from the aspects of the type and stereochemistry of the monoterpene ring and the substituent effects on the 1,3-oxazine ring system. The N-unsubstituted monoterpene-based derivatives exhibited considerable antiproliferative activity against a panel of human adherent cancer cell lines (HeLa, MCF7 and A431). Conclusions: A mild and efficient method has been developed for the synthesis of 2-imino-1,3-oxazines by the ring closure of thiourea adducts of 1,3-amino alcohols. The resulting 1,3-oxazines exert marked antiproliferative action on a panel of human cancer cell lines.

Photodynamic activity of nanostructured fabrics grafted with xanthene and thiazine dyes against opportunistic fungi

Fungi are an important class of human pathogens for which considerable research has gone into defeating them. The photodynamic effects of rose bengal (RB), phloxine B (PB), azure A (AA), and toluidine blue O (TBO) dyes to inhibit Aspergillus fumigatus, Aspergillus niger, Trichoderma viride, Penicillium funiculosum, and Chaetomium globosum were investigated grafted to nano- and micro-structured fabrics. Three antifungal tests conducted: broth microdilution test of free dyes, zone of inhibition and quantitative antifungal assays on fabrics grafted with dyes. In the broth microdilution test, free RB displayed the lowest MIC at 32 mu M to inhibit visible hyphal growth and germination but the antifungal ability of MIC for other photosensitizers below 63 mu M was insignificant. RB and PB showed lower MIC than AA and TBO. In the inhibition zone tests, nanostructured fabrics grafted with RB and PB did not display fungal growth on the surface. Most microstructured fabrics grafted with AA and TBO showed little inhibition. In quantitative antifungal assay, nanostructured fabrics grafted with RB has the largest inhibition rate on T. viride and the lowest inhibition rate on P. funiculosum and the results showed the increasing inhibition rate in the order of AA < TBO < PB < RB. Design, synthesis and biological evaluation of substituted 2-amino-1,3-thiazine derivatives as antituberculosis and anti-cancer agents

The Coats-Redfern equation was used to calculate the kinetic and thermodynamic parameters for the different thermal decomposition steps of some complexes. All the synthesized compounds were tested for in vitro antibacterial activity against some Gram-positive and Gram-negative bacteria, yeast and fungus.
