Upon completion, the residue waspurified with column chromatography (hexane:EtOAc 9:1) toobtain analogue 13n~13o.

General procedure: To a mixture of sodium hydride suspension (60percent, 0.26 g, 6.67 mmol) and phenothiazine derivative in abs. THF (50 cm3) was added the appropriate 3-aryltetrazolo[1,5-a]pyridin-4-ium tetrafluoroborate (1) in portions over 20 min, and the mixture was stirred at room temperature for 2 days. After evaporation of the reaction mixture, the residue was dissolved in a 1:1 mixture of EtOAc:water (50 cm3) and the solid product was filtered off. The aqueous mother liquor was extracted with EtOAc (thrice) and the organic phase was dried over anhydrous Na2SO4, filtered and evaporated. The combined organic solvent was evaporated, the residue was washed with diethyl ether then it was recrystallized from ethyl acetate – unless otherwise stated – to give the product.

38.1) 2-Methoxy-10H-phenothiazine-1-carbaldehyde: 4.6 g (20 mmol) of 2-methoxy-10H-phenothiazine are dissolved, under an argon atmosphere, in a three-necked flask, containing 140 ml anhydrous ethyl ether. 20 ml (50 mmol) of a solution of nBuLi (2.5 M) in hexane is then added dropwise, at 20 C. The reaction mixture is agitated for 3 hours at 20 C. before the dropwise addition of 6.2 ml (80 mmol) of anhydrous DMF. Agitation is maintained for another 15 hours at 20 C. The whole is then poured into 150 ml ice-cooled water and the product is extracted twice using 200 ml of ethyl acetate. The organic solution is washed with 100 ml of salt water, dried over MgSO4, filtered and concentrated under vacuum. The evaporation residue is taken up in diisopropyl ether, filtered and dried to produce a red solid with a yield of 30%. Melting point: 155-160 C.

After stirring for 1 hour at 40 , under ice-cooling, use 1mol / L diluted hydrochloric acid, and extracted with ethyl acetate. The combined organic layers were dried over sodium sulfate, filtered, concentrated, and the use of silica gel column chromatography (hexane: ethyl acetate = 1: 1) and the residue was purified to obtain the title compound (130mg, 31%) .

Step a 4-(3′-indolyl)-1-[1″-(2′”-trifluoromethyl-10′”-phenothiazinyl)-2″-methyl-3″-propionyl]-piperidine hydrochloride 27 g of 2-trifluoromethyl-phenothiazine and 12 g of alpha-methylacryloyl chloride in 100 ml of toluene were 1 for 2 hours and the mixture was evaporated to dryness. The residue was chromatographed over silica gel and elution with a 1– 1 benzene-cyclohexane mixture gave 18 g of 2-trifluoromethyl-10-(alpha-methyl acryloyl)-phenothiazine in the form of white crystals melting at 103¡ãC.

Preparation of 4-[(3-oxo-3,4-dihydro-2H-benzo[l ,41thiazine-6-carbonyl)-aminol-piperidine- 1-carboxylic acid tert-butyl ester:3-Oxo-3,4-dihydro-2H-benzo[l,4]thiazine-6-carboxylic acid (556 mg, 2.45 mmol, 1.0 eq) is added at room temperature to a stirred solution of 4-amino-piperidine- 1-carboxylic acid tert- butyl ester (500 mg, 2.45 mmol, 1.0 eq) in N,N-dimethylformamide (20 mL), followed by 1- hydroxybenzotriazole (421 mg, 2.69 mmol, 1.1 eq), N-(3-dimethylaminopropyl)-N’- ethylcarbodiimide hydrochloride (550 mg, 2.81 mmol, 1.15 eq) and N,N- diisopropylethylamine (962 L, 5.50 mmol, 2.25 eq). After 15 hours stirring at room temperature, solvent is evaporated and the residue is extracted with dichloromethane (3 x 30 – – mL) and water (30 mL). The combined organic layers are dried over sodium sulfate, filtered and concentrated to give a crude product that is purified by column chromatography (silica gel, eluent: cyclohexane:ethyl acetate :methanol, 1 :3:0 to 0/9/1, v/v/v) to afford 4-[(3-oxo-3,4- dihydro-2H-benzo [ 1 ,4]thiazine-6-carbonyl)-amino ] -piperidine- 1 -carboxylic acid tert-butyl ester as a light yellow solid (621 mg, 62% yield).1H-NMR (400 MHz, DMSO- 6) delta ppm: 10.66 (s, 1H), 8.27 (d, J = 7.8 Hz, 1H), 7.42 (m, 3H), 3.96 (m, 3H), 3.51 (s, 2H), 2.90 (m, 2H), 1.76 (m, 2H), 1.40 (m, 11H).MS m/z (+ESI): 392.3 [M+H]+, 414.3 [M+Na]+.
