Microwave-assisted Solid-phase Synthesis of N-substituted-2-aminobenzo[d][1,3] Thiazine Derivatives from a BOMBA Resin

An efficient solid-phase methodology has been developed for the synthesis of N-acyl and N-sulfonyl substituted 2-aminobenzo[d][1,3] thiazine derivatives. The key step in this methodology is the preparation of backbone amide linker-bound 2-aminobenzo[d][1,3] thiazine resin through cyclization reaction between isothiocyanates and BOMBA resin under microwave irradiation. This 2-aminobenzo[d][1,3] thiazine core skeleton resin undergoes substitution reactions with various electrophiles, such as acid halides and sulfonyl halides, to generate N-acyl and N-sulfonyl substituted 2-aminobenzo[d][1,3] thiazine resins, respectively. Finally, N-acyl and N-sulfonyl-substituted 2-aminobenzo[d][1,3] thiazine derivatives are prepared in good yields and purities by cleavage of the respective resins under trifluoroacetic acid (TFA) in dichloromethane (DCM).

Media polarity and concentration roles on the third order nonlinear behaviors of thiazine dyes

Nonlinear optical materials play important roles in optics and photonics. In order to improve the performance of these materials, in this paper, we tried to find effective methods for controlling the nonlinear responses of thiazine dyes. Our experimental results indicate that molecular linear properties, media polarity, concentration and excitation beam properties with different contributions play significant roles on the saturable and reverse saturable absorption characteristics of thiazine dyes. In real, competition between these effective parameters leads to appearance of various third order nonlinear responses. Although by excitation wavelengths near the absorption peaks and at high powers, dye molecules tend to indicate reverse saturable absorption characteristics in polar protic solvents by increasing the concentration, saturable absorption characteristics were observed in polar aprotic solvents. Furthermore, the samples displayed the same behaviors in different solvent media at low powers and for excitation wavelengths far from absorption peaks. Therefore, by controlling the nonlinear responses of thiazine dyes, these materials can indicate both saturable and reverse saturable absorption characteristics that increase their applications in optics and photonics devices. Luminescence and Nonlinear Optical Properties of Hybrid Associates of Ag2S Quantum Dots with Molecules of Thiazine Dyes

When the IR luminescence of Ag2S quantum dots (QDs) is excited by radiation from the thionine absorption region, its enhancement due to nonradiative resonant energy transfer is observed. The association with methylene-blue molecules blocked the IR luminescence of Ag2S QDs upon its excitation by radiation from the absorption region of the dye due to the transfer of charge carriers. It is demonstrated that the hybrid association of thionine molecules and Ag2S QDs adversely affects the nonlinear optical properties of the latter, which manifests itself in inverse saturated absorption by the action of 10-ns second-harmonic pulses (532 nm) of a Nd3+:YAG laser. For the associates of Ag2S QDs with methylene-blue molecules, the radiation focusing caused by the transfer of charge carriers from the dye and the change in the population of small traps in nanocrystals is found. It is concluded that the direction of the transfer of electronic excitations and the photophysical processes in these objects are determined by the mutual arrangement of the HOMO-LUMO levels of the dye with respect to the levels of dimensional quantization of the Ag2S QDs.

Breast cancer is one of the leading causes of death in cancer categories, followed by lung, colorectal, and ovarian among the female gender across the world. 10H-3,6-diazaphenothiazine (PTZ) is a thiazine derivative compound that exhibits many pharmacological activities. Herein, we proceed to investigate the pharmacological activities of PTZ toward breast cancer MCF-7 cells as a representative in vitro breast cancer cell model. The PTZ exhibited a proliferation inhibition (IC50 = 0.895 mu M) toward MCF-7 cells. Further, cell cycle analysis illustrated that the S-phase checkpoint was activated to achieve proliferation inhibition. In vitro cytotoxicity test on three normal cell lines (HEK293 normal kidney cells, MCF-10A normal breast cells, and H9C2 normal heart cells) demonstrated that PTZ was more potent toward cancer cells. Increase in the levels of reactive oxygen species results in polarization of mitochondrial membrane potential (Delta psi m), together with suppression of mitochondrial thioredoxin reductase enzymatic activity suggested that PTZ induced oxidative damages toward mitochondria and contributed to improved drug efficacy toward treatment. The (RTPCR)-P-2 Profiler Array (human apoptosis pathways) proved that PTZ induced cell death via mitochondria-dependent and cell death receptor-dependent pathways, through a series of modulation of caspases, and the respective morphology of apoptosis was observed. Mechanistic studies of apoptosis suggested that PTZ inhibited AKT1 pathways resulting in enhanced drug efficacy despite it preventing invasion of cancer cells. These results showed the effectiveness of PTZ in initiation of apoptosis, programmed cell death, toward highly chemoresistant MCF-7 cells, thus suggesting its potential as a chemotherapeutic drug.

Synthesis, Characterization and Molecular Docking of Novel Bioactive Thiazolyl-Thiazole Derivatives as Promising Cytotoxic Antitumor Drug

Reactions of ethylidenethiocarbohydrazide with hydrazonoyl halides gave 1,3-thiazole or 1,3,4-thiadiazole derivatives according to the type of hydrazonoyl halides. Treatment of ethylidenethiosemicarbazide with hydrazonoyl halides and dimethylacetylene dicarboxylate (DMAD) afforded the corresponding arylazothiazoles and 1,3-thiazolidin-4-one derivatives, respectively. The structures of the synthesized products were confirmed by IR, H-1-NMR, C-13-NMR and mass spectral techniques. The cytotoxic activity of the selected products against the Hepatic carcinoma cell line (Hepg-2) was determined by MTT assay indicating a concentration dependent cellular growth inhibitory effect, especially for compounds 14c and 14e. The dose response curves indicated the IC50 (the concentration of test compounds required to kill 50% of cell population) were 0.54 M and 0.50 M, respectively. Confocal laser scanning imaging of the treated cells stained by Rhodamin 123 and Acridine orange dyes confirmed that the selected compounds inhibit the mitochondrial lactate dehydrogenase enzymes. The binding mode of the active compounds was interpreted by a molecular docking study. The obtained results revealed promising cytotoxic activity.

Visible light-driven oxidation of arsenite, sulfite and thiazine dyes: A new strategy for using waste to treat waste

Recently, the advanced oxidation processes (AOPs) based on sulfate radical (SO4 center dot-) using sulfite has received increasing concerns. Metal-free activation of sulfite is still in urgent demands from the perspective of cleaner production since the reported efficient systems containing sulfite has been relied on the transition metals. In this work, methylene blue (MB+), a well-known photosensitizer and thiazine dye contaminant, was utilized to activate sulfite for the efficient photooxidation of arsenite (As(III)) to arsenate (As(V)) under visible light (LED lamps with wavelength 640 nm) and sunlight, respectively. The initial oxidation rates of 5-40 mu M As(III) are 28-folde116-fold as that of the decolorization of 0.27 mu M MB+ with 50 mu M sulfite at pH 7.3. The maximum stoichiometric ratio MB+: As(III): sulfite was obtained to be 1 : 142: 247, suggesting an efficient utilization of both MB+ and sulfite as waste resources. The electrical energy per order (EE/O) index was calculated to be 1.83 x 10(-3) kWh L-1 for the visible light-MB+-sulfite system, which is much less than those reported systems. Oxysulfur radicals (mainly peroxymonosulfate radical, SO5 center dot-) produced in this photochemical system are responsible for As(III) oxidation. The other two common thiazine dyes (thionine and Azure B) are also capable of activating sulfite for As(III) oxidation due to the same chromophores. Thus the novel strategy of using waste sulfite and the thiazine dyes is promising for As(III) oxidation under sunlight, with which low-concentration wastes of thiazine dye and sulfite can be utilized to achieve the goal of cleaner production.
