Synthesis, Characterization and Molecular Docking of Novel Bioactive Thiazolyl-Thiazole Derivatives as Promising Cytotoxic Antitumor Drug

Reactions of ethylidenethiocarbohydrazide with hydrazonoyl halides gave 1,3-thiazole or 1,3,4-thiadiazole derivatives according to the type of hydrazonoyl halides. Treatment of ethylidenethiosemicarbazide with hydrazonoyl halides and dimethylacetylene dicarboxylate (DMAD) afforded the corresponding arylazothiazoles and 1,3-thiazolidin-4-one derivatives, respectively. The structures of the synthesized products were confirmed by IR, H-1-NMR, C-13-NMR and mass spectral techniques. The cytotoxic activity of the selected products against the Hepatic carcinoma cell line (Hepg-2) was determined by MTT assay indicating a concentration dependent cellular growth inhibitory effect, especially for compounds 14c and 14e. The dose response curves indicated the IC50 (the concentration of test compounds required to kill 50% of cell population) were 0.54 M and 0.50 M, respectively. Confocal laser scanning imaging of the treated cells stained by Rhodamin 123 and Acridine orange dyes confirmed that the selected compounds inhibit the mitochondrial lactate dehydrogenase enzymes. The binding mode of the active compounds was interpreted by a molecular docking study. The obtained results revealed promising cytotoxic activity.

An Anti-Inflammatory Azaphenothiazine Inhibits Interferon beta Expression and CXCL10 Production in KERTr Cells

An azaphenothiazine derivative, 6-chloroethylureidoethyldiquino[3,2-b; 2 0,3 0 -e][1,4]thiazine (DQT), has recently been shown to exhibit immunosuppressive activities in mouse models. It also inhibited the expression of CXCL10 at the protein level, at non-toxic concentrations, in the culture of KERTr cells treated with double-stranded RNA, poly(I:C). In this report, we demonstrated that DQT inhibits the transcription of the CXCL10 gene. Although CXCL10 is an IFN gamma-inducible protein, we found that the CXCL10 protein was induced without the detectable release of IFN gamma or I kappa B degradation. Hence, we concluded that IFN gamma or NF kappa B was not involved in the regulation of the CXCL10 gene in KERTr cells transfected with poly(I: C), nor in the inhibitory activity of DQT. On the other hand, we found that IFN beta was induced under the same conditions and that its expression was inhibited by DQT. Kinetic analysis showed that an increase in IFN beta concentrations occurred 4-8 h after poly(I: C) treatment, while the concentration of CXCL10 was undetectable at that time and started to increase later, when IFN beta reached high levels. Therefore, DQT may be regarded as a new promising inhibitor of IFN beta expression and IFN beta-dependent downstream genes and proteins, e.g., CXCL10 chemokine, which is implicated in the pathogenesis of autoimmune diseases.

Chemometric Characterization of Synthetic Dye Sorption onto Slovakian River Sediments: A Laboratory Batch Experiment

The aim of the work was to characterize the sorption of cationic dyes thioflavine T (ThT) and methylene blue (MB) onto selected Slovakian river sediments using chemometric approaches including principal component analysis (PCA) and cluster analysis (CA). Also, the potential of mentioned multivariate analyses for comparison of studied objects (river sediments or river and model waters) as well as in finding relationships between the variables describing the physico-chemical characteristics of studied matrices or waters and sorption/desorption characteristics of matrices for dyes binding under laboratory conditions was evaluated. Parameters describing the physico-chemical characteristics of sediments include: pH, pH(zpc), or cation-exchange capacity; and in the case of waters: pH, conductivity, water hardness, content of dissolved solids or presence of organic compounds. From the comparison of dye sorption onto sediments, it was found that sorption of thiazine dye MB was minimally 1.5-times higher than sorption of benzothiazole dye ThT. Sorption capacities Q(s) reached the maximum values in the case of sediments originated from Dudvah River (MB-Q(s) = 8.70 +/- 0.42 mg g(-1); ThT-Q(s) = 5.03 +/- 0.28 mg g(-1); +/- SD). Obtained results showed that applied methods of multivariate analyses represent a suitable tool for evaluation of sorption/desorption processes of organic xenobiotics binding in sediments.

Synthesis of Polynuclear Heterocyclic Compounds Derived from 1-Biphenyl-4-yl-4-(1-methyl-1H-benzimidazol-2-yl)-But-2-en-1-One with Expected Biological Activity

NUCLEOPHILIC addition of barbituric acid, thioglycolic acid with malononitrile and 3-Amino-5-pyrazolinone with 1-Biphenyl-4-yl-4-(1-methyl-1H-benzimidazol-2-yl)-but-2-en-1-one (1) gives pyrimidine, Thiazol and pyrazolinone derivatives (2-4), repectively. Compound 4 which reacted with Arylidene malononitrile in ethanol to give pyrano derivatives (5). Cyclocondensation of chalcone (1) with thiourea under reflux in ethanolic NaOH led to thiazine derivative (6) and treatment of (6) with bromine-acetic acid mixture afforded compound (7) similarly, cyclization of chalcone (1) with orthophenylinediamine in Dimethylformamide (DMF) gave diazepine derivatives (8). Also, chalcone (1) reacted with 3-Amino-5-pyrazolinone in ethanol to give pyrazolinone derivative (9). (Scheme 1). The condensation of compound (1) with 2-cyanoacetohydrazide through the acid-catalyzed reaction to form the Schiff base (10) by heating in 96% acetic acid gave the diazetidine derivative (11) which upon reaction with phosphorous oxychloride gave the chlorodiazetidine derivative (12), the latter compound reacted with 2ry amines and yielded the substitueted amino diazetidine derivatives (13(a-c)). On the other hand, the Schiff bases (14) produced from condensed compound (1) with 2-Amino-2-(hydroxy methyl) propane-1,3-diol in ethyl alcohol which reacted with phosphorous oxychloride gave the propanol derivatives (15). Finally, the cyclization reaction of compound (1) with 2-aminophenol and 2-aminothiophenol in dry benzene formed compounds (16, 17), repectively.(Scheme 2).

Therapeutic effects of an azaphenothiazine derivative in mouse experimental colitis

Phenothiazines represent a class of compounds of potential therapeutic utility. In this report we evaluated therapeutic value of an azaphenothiazine derivative, 6-acetylaminobutyl-9-chloroquino[3,2-b]benzo[1,4]thiazine (QBT), given intragastrically, in the model of dextran sodium sulfate-induced colitis in C57BL/6 mice using 5-aminosalicylic acid (5-ASA) as a reference drug. Colitis symptoms such as body weight loss, diarrhea and hematochezia (blood in stool) were observed and registered and disease activity index (DAI) was calculated. In addition, weight and cell numbers in the lymphatic organs and histological parameters of the colon wall were analyzed. The effects of QBT on viability of colon epithelial cell lines were also determined. We showed that weight and cell number of draining mesenteric lymph nodes were lower in mice treated with QBT in comparison to their control counterparts. The number of thymocytes, drastically reduced in control mice, was elevated in mice treated with the compounds with a significant effect of 5-ASA. In addition, an abnormal composition of blood cell types was partially corrected in these groups. Histological analysis of the colon revealed that the pathological changes were partially normalized by QBT and even to a higher degree by 5-ASA. In conclusion we demonstrated a therapeutic efficacy of the compound in amelioration of local and systemic pathological changes associated with chemically-induced colitis in mice. A possible mechanism of action of the compound is discussed.

The complex of zinc with N-(5,6-dihydro-4H-1,3-thiazine-2-yl)benzamide

A complex of zinc with N-(5,6-dihydro-4H-1,3-thiazin-2-yl)benzamide (L) ligand, LZnCl2, was synthesized for subsequent medical trials. The molar extinction coefficients were determined for LHBr solutions in water and physiological saline, and for LZnCl2 ethanol solution. The ligand stability in various solvents was evaluated and the value of its protonation constant was found for the physiological saline solution, logK = 5.3 +/- 0.2. The impossibility of determination of the complex stability constant by the potentiometric titration method was demonstrated. The complex exhibited an insufficient stability in aqueous and physiological saline solutions, but was stable as the solution in alcohol. There was no sorption observed upon the treatment of ligand with hydroxyapatite nanoparticles, which could be a potential carrier for the therapeutic form of LZnCl2, providing additional degrees of freedom for the interaction of ligand with cell membranes and a prolonged action of zinc ions.

Visible light-driven oxidation of arsenite, sulfite and thiazine dyes: A new strategy for using waste to treat waste

Recently, the advanced oxidation processes (AOPs) based on sulfate radical (SO4 center dot-) using sulfite has received increasing concerns. Metal-free activation of sulfite is still in urgent demands from the perspective of cleaner production since the reported efficient systems containing sulfite has been relied on the transition metals. In this work, methylene blue (MB+), a well-known photosensitizer and thiazine dye contaminant, was utilized to activate sulfite for the efficient photooxidation of arsenite (As(III)) to arsenate (As(V)) under visible light (LED lamps with wavelength 640 nm) and sunlight, respectively. The initial oxidation rates of 5-40 mu M As(III) are 28-folde116-fold as that of the decolorization of 0.27 mu M MB+ with 50 mu M sulfite at pH 7.3. The maximum stoichiometric ratio MB+: As(III): sulfite was obtained to be 1 : 142: 247, suggesting an efficient utilization of both MB+ and sulfite as waste resources. The electrical energy per order (EE/O) index was calculated to be 1.83 x 10(-3) kWh L-1 for the visible light-MB+-sulfite system, which is much less than those reported systems. Oxysulfur radicals (mainly peroxymonosulfate radical, SO5 center dot-) produced in this photochemical system are responsible for As(III) oxidation. The other two common thiazine dyes (thionine and Azure B) are also capable of activating sulfite for As(III) oxidation due to the same chromophores. Thus the novel strategy of using waste sulfite and the thiazine dyes is promising for As(III) oxidation under sunlight, with which low-concentration wastes of thiazine dye and sulfite can be utilized to achieve the goal of cleaner production. Methylene blue was the first synthetic drug ever used in medicine, having been used to treat clinical pain syndromes, malaria, and psychotic disorders more than one century ago. Methylene blue is a cationic thiazine dye with redox-cycling properties and a selective affinity for the nervous system. This drug also inhibits the activity of monoamine oxidase, nitric oxide synthase, and guanylyl cyclase, as well as tau protein aggregation; increases the release of neurotransmitters, such as serotonin and norepinephrine; reduces amyloid-beta levels; and increases cholinergic transmission. The action of methylene blue on multiple cellular and molecular targets justifies its investigation in various neuropsychiatric disorders. Investigations of methylene blue were instrumental in the serendipitous development of phenothiazine antipsychotic drugs. ZrCl4/TMSCl as an Efficient Catalyst for Synthesis of 4,6-Substituted 2-Alkylthio-6H-1,3-thiazines
