Human histone deacetylase 8 (HDAC8) is a highly promising target for neuroblastoma and other types of cancer. Several HDAC inhibitors are approved for the treatment of special cancer subtypes or are evaluated in clinical trials. By far the most drugs or drug candidates contain a hydroxamate group that chelates the catalytic zinc ion within HDACs. Most hydroxamate inhibitors are more or less unselective, although there are considerable exceptions demonstrating the general feasibility to develop at least HDAC isoenzyme selective inhibitors. In addition, hydroxamates have recently come under discussion regarding their potential for mutagenicity. Recently, PD-404,182 was discovered as a selective and potent non-hydroxamate inhibitor of HDAC8. However, this active compound turned out to be decomposed in the presence of glutathion (GSH). Here, we describe the synthesis of significantly improved analogs of PD-404,182 that demonstrate both, great selectivity for HDAC8 and also chemical stability in the presence of GSH. The compounds are characterized with respect to structure-activity relationship, binding mode and target engagement in neuroblastoma cells by combining biochemical and biophysical methods with chemoinformatics. Synthesis, spectroscopic structure identification, X-ray study and anticancer activities of new angularly fused quinobenzothiazines

Synthesis of 16 new tetracyclic angularly fused azaphenothiazines, 8-, 9- and 10-substituted quinobenzo1,4-thiazines (benzo[a]-3-azaphenothiazines) was based on the reactions of dichlorodiquinolinyl disulfide and diquinodithiin with substituted anilines. Whereas the reactions with p-fiuoroaniline and p-methylthioaniline led to only one product, the reaction with m-triflouromethylaniline led to isomeric compounds. The obtained 8-10-substituted 12H-quinobenzothiazines were further transformed into 12 substituted derivatives through allcylation of the thiazine nitrogen atom. The structure analysis was based on 1D and 2D NMR (NOESY, COSY, HSQC and HMBC) spectra which enabled to distinguish the isomers and to exclude retro-Smiles rearrangement and the azine nitrogen atom alkylation pathways. This supposition was fully confirmed by X-ray analysis showing the quinobenzothiazine system to be folded and the substituent at the thiazine nitrogen atom in an equatorial position. Some compounds exhibited anticancer activity against MCF-7, MDA-MB-231 and SNB-19 cell lines similar to a reference drug cisplatin. 10H-1,9-diazaphenothiazine and its 10-derivatives: synthesis, characterisation and biological evaluation as potential anticancer agents

10H-1,9-diazaphenothiazine was obtained in the sulphurisation reaction of diphenylamine with elemental sulphur and transformed into new 10-substituted derivatives, containing alkyl and dialkylaminoalkyl groups at the thiazine nitrogen atom. The 1,9-diazaphenothiazine ring system was identified with advanced H-1 and C-13 NMR techniques (COSY, NOESY, HSQC and HMBC) and confirmed by X-ray diffraction analysis of the methyl derivative. The compounds exhibited significant anticancer activities against the human glioblastoma SNB-19, melanoma C-32 and breast cancer MDA-MB-231 cell lines. The most active 1,9-diazaphenothiazines were the derivatives with the propynyl and N, N-diethylaminoethyl groups being more potent than cisplatin. For those two compounds, the expression of H3, TP53, CDKN1A, BCL-2 and BAX genes was detected by the RT-QPCR method. The proteome profiling study showed the most probable compound action on SNB-19 cells through the intrinsic mitochondrial pathway of apoptosis. The 1,9-diazaphenotiazine system seems to be more potent than known isomeric ones (1,6-diaza-, 1,8-diaza-, 2,7-diaza- and 3,6-diazaphenothiazine). [GRAPHICS] .

Removal Bismarck Brown G dye from Aqueous Solution Over a Composite of Triazole-Polyvinyl Chloride Polymer and Zinc Oxide

The present study involves synthesis a composite of triazole-poly vinyl chloride (PVC) polymer with nano-crystalline zinc oxide to yield triazole-PVC/ZnO. The produced composite was characterized using fouier infrared spectroscopy (FTIR), X-Rays diffraction (XRD) and micro-elemental analysis (CHN). The activity of the produced composite with respect to neat zinc oxide was investigated by following both of adsorption and photocatalytic removal of Bismarck Brown G (BBG) dye from their aqueous solutions. The obtained results in this study showed that triazole-PVC/ZnO was more efficient than neat ZnO on dye removal for both of adsorption and photocatalytic reaction.

A comparative study for removal of different dyes over M/TiO2 (M = Cu, Ni, Co, Fe, Mn and Cr) photocatalysts under visible light irradiation

The purpose of this work is to design and develop a series of stable and effective TiO2 photocatalysts prepared by a modified precipitation method. In order to improve the photo response of the TiO2 to the visible region, the effect of addition of six successive transition metal elements of fourth period, namely chromium (Cr), manganese (Mn), iron (Fe), cobalt (Co), nickel (Ni), and copper (Cu) was studied. Two different types of dyes, methyl orange (MO, azo dye) and methylene blue (MB, thiazine dye group), were used to investigate the influence of structure of dyes on the photocatalytic degradation rates. The results showed that loading titania with 0.3 wt.% of different transition metal elements was found to have significant influences on the crystallographic structure, physical properties and optical absorption properties of titania based catalysts, as well as catalytic activity during dye degradation reactions. The optical reflection edge was obviously shifted to the visible light range with the transition metal additives. The band gap energies of the doped TiO2 samples were considerably narrower than that of plain TiO2. The different activity levels of the catalysts for both dyes clearly show that the photocatalytic activity of samples strongly depended on the metal dopant used. Among all the catalysts, Cu/TiO2 sample exhibited the highest photocatalytic activity under visible light for both of the dyes owing to the low band gap energy and delayed electron-hole recombination.
