Therapeutic effects of an azaphenothiazine derivative in mouse experimental colitis

Phenothiazines represent a class of compounds of potential therapeutic utility. In this report we evaluated therapeutic value of an azaphenothiazine derivative, 6-acetylaminobutyl-9-chloroquino[3,2-b]benzo[1,4]thiazine (QBT), given intragastrically, in the model of dextran sodium sulfate-induced colitis in C57BL/6 mice using 5-aminosalicylic acid (5-ASA) as a reference drug. Colitis symptoms such as body weight loss, diarrhea and hematochezia (blood in stool) were observed and registered and disease activity index (DAI) was calculated. In addition, weight and cell numbers in the lymphatic organs and histological parameters of the colon wall were analyzed. The effects of QBT on viability of colon epithelial cell lines were also determined. We showed that weight and cell number of draining mesenteric lymph nodes were lower in mice treated with QBT in comparison to their control counterparts. The number of thymocytes, drastically reduced in control mice, was elevated in mice treated with the compounds with a significant effect of 5-ASA. In addition, an abnormal composition of blood cell types was partially corrected in these groups. Histological analysis of the colon revealed that the pathological changes were partially normalized by QBT and even to a higher degree by 5-ASA. In conclusion we demonstrated a therapeutic efficacy of the compound in amelioration of local and systemic pathological changes associated with chemically-induced colitis in mice. A possible mechanism of action of the compound is discussed.

Luminescence and Nonlinear Optical Properties of Hybrid Associates of Ag2S Quantum Dots with Molecules of Thiazine Dyes

Spectral, kinetic, and nonlinear optical regularities that demonstrate the exchange of electronic excitations between the components of hybrid associates of Ag2S colloid quantum dots (1.7-1.8 nm) in gelatin with molecules of thiazine dyes (Ds) are found. When the IR luminescence of Ag2S quantum dots (QDs) is excited by radiation from the thionine absorption region, its enhancement due to nonradiative resonant energy transfer is observed. The association with methylene-blue molecules blocked the IR luminescence of Ag2S QDs upon its excitation by radiation from the absorption region of the dye due to the transfer of charge carriers. It is demonstrated that the hybrid association of thionine molecules and Ag2S QDs adversely affects the nonlinear optical properties of the latter, which manifests itself in inverse saturated absorption by the action of 10-ns second-harmonic pulses (532 nm) of a Nd3+:YAG laser. For the associates of Ag2S QDs with methylene-blue molecules, the radiation focusing caused by the transfer of charge carriers from the dye and the change in the population of small traps in nanocrystals is found. It is concluded that the direction of the transfer of electronic excitations and the photophysical processes in these objects are determined by the mutual arrangement of the HOMO-LUMO levels of the dye with respect to the levels of dimensional quantization of the Ag2S QDs.

10H-3,6-Diazaphenothiazines triggered the mitochondrial-dependent and cell death receptor-dependent apoptosis pathways and further increased the chemosensitivity of MCF-7 breast cancer cells via inhibition of AKT1 pathways

Breast cancer is one of the leading causes of death in cancer categories, followed by lung, colorectal, and ovarian among the female gender across the world. 10H-3,6-diazaphenothiazine (PTZ) is a thiazine derivative compound that exhibits many pharmacological activities. Herein, we proceed to investigate the pharmacological activities of PTZ toward breast cancer MCF-7 cells as a representative in vitro breast cancer cell model. The PTZ exhibited a proliferation inhibition (IC50 = 0.895 mu M) toward MCF-7 cells. Further, cell cycle analysis illustrated that the S-phase checkpoint was activated to achieve proliferation inhibition. In vitro cytotoxicity test on three normal cell lines (HEK293 normal kidney cells, MCF-10A normal breast cells, and H9C2 normal heart cells) demonstrated that PTZ was more potent toward cancer cells. Increase in the levels of reactive oxygen species results in polarization of mitochondrial membrane potential (Delta psi m), together with suppression of mitochondrial thioredoxin reductase enzymatic activity suggested that PTZ induced oxidative damages toward mitochondria and contributed to improved drug efficacy toward treatment. The (RTPCR)-P-2 Profiler Array (human apoptosis pathways) proved that PTZ induced cell death via mitochondria-dependent and cell death receptor-dependent pathways, through a series of modulation of caspases, and the respective morphology of apoptosis was observed. Mechanistic studies of apoptosis suggested that PTZ inhibited AKT1 pathways resulting in enhanced drug efficacy despite it preventing invasion of cancer cells. These results showed the effectiveness of PTZ in initiation of apoptosis, programmed cell death, toward highly chemoresistant MCF-7 cells, thus suggesting its potential as a chemotherapeutic drug.

Media polarity and concentration roles on the third order nonlinear behaviors of thiazine dyes

Nonlinear optical materials play important roles in optics and photonics. In order to improve the performance of these materials, in this paper, we tried to find effective methods for controlling the nonlinear responses of thiazine dyes. Our experimental results indicate that molecular linear properties, media polarity, concentration and excitation beam properties with different contributions play significant roles on the saturable and reverse saturable absorption characteristics of thiazine dyes. In real, competition between these effective parameters leads to appearance of various third order nonlinear responses. Although by excitation wavelengths near the absorption peaks and at high powers, dye molecules tend to indicate reverse saturable absorption characteristics in polar protic solvents by increasing the concentration, saturable absorption characteristics were observed in polar aprotic solvents. Furthermore, the samples displayed the same behaviors in different solvent media at low powers and for excitation wavelengths far from absorption peaks. Therefore, by controlling the nonlinear responses of thiazine dyes, these materials can indicate both saturable and reverse saturable absorption characteristics that increase their applications in optics and photonics devices. Enhancement of nonlinear optical response of methylene blue and azure a during association with colloidal CdS quantum dots

The nonlinear optical response of thiazine dyes during association with colloidal CdS quantum dots (QDs) applying the z-scan technique at wavelength of 532 nm with use 10 ns pulses of Nd:YAG laser second harmonic were studied. Increasing in nonlinear absorption intensity in hybrid associates in comparison with the initial components were found. Moreover, UV-vis absorption spectrum shows the effective formation of dyes H-aggregates during association with colloidal QDs. Two-photon mechanism of observed nonlinear absorption in hybrid associates is proposed, based on a sharper dependence of the normalized transmittance on the incident intensity (distance between the sample and focal plane of focusing lens). And for pure solutions, the reverse saturated absorption is preferred nonlinear absorption mechanism. The nonlinear absorption coefficients for MB and AzA in the hybrid associates were found. They are beta approximate to 8.4 x 10(-9) cm W-1 (sic) approximate to 1.34 x 10(-8) cm W-1, respectively. It was shown that it is possible to control the parameters of nonlinear absorption of thiazine dyes via the hybrid association.

A simple and efficient pathway to conjugate monosaccharides to thienopyrimidines via click chemistry establishing a 1,2,3-triazole linker was reported. Design, synthesis, characterization and antimicrobial evaluation of some novel hydrazinecarbothioamide, 4-thiazolidinone and 1,2,4-triazole-3-thione derivatives
