In Vitro Antiproliferative Evaluation of Synthetic Meroterpenes Inspired by Marine Natural Products

Several marine natural linear prenylquinones/hydroquinones have been identified as anticancer and antimutagenic agents. Structure-activity relationship studies on natural compounds and their synthetic analogs demonstrated that these effects depend on the length of the prenyl side chain and on the type and position of the substituent groups in the quinone moiety. Aiming to broaden the knowledge of the underlying mechanism of the antiproliferative effect of these prenylated compounds, herein we report the synthesis of two quinones 4 and 5 and of their corresponding dioxothiazine fused quinones 6 and 7 inspired to the marine natural product aplidinone A (1), a geranylquinone featuring the 1,1-dioxo-1,4-thiazine ring isolated from the ascidian Aplidium conicum. The potential effects on viability and proliferation in three different human cancer cell lines, breast adenocarcinoma (MCF-7), pancreas adenocarcinoma (Bx-PC3) and bone osteosarcoma (MG-63), were investigated. The methoxylated geranylquinone 5 exerted the highest antiproliferative effect exhibiting a comparable toxicity in all three cell lines analyzed. Interestingly, a deeper investigation has highlighted a cytostatic effect of quinone 5 referable to a G0/G1 cell-cycle arrest in BxPC-3 cells after 24 h treatment.

In addition, thioamide 3 was used as starting material for preparation of a new series of thiadiazole derivatives via its reaction with hydrazonoyl chlorides 5a-c in dioxane using triethylamines as catalyst. DECOLORIZATION KINETICS OF ACID AZO DYE AND BASIC THIAZINE DYE IN AQUEOUS SOLUTION BY UV/H2O2 AND UV/FENTON: EFFECTS OF OPERATIONAL PARAMETERS

The photochemical decolorization of two dyes, namely Acid Yellow 54 and Basic Blue 9, was studied using the UV/H2O2 and UV/Fenton processes. The effects of the amount of H2O2 and FeSO4 as well as the initial pH solution on decolorization kinetics of both the dyes were investigated. The pseudo-first order kinetic model was applied to predict the decolorization of the selected dyes at the different operational conditions and results showed that this model fitted very well with the experimental data. The obtained results also showed the efficiency of UV/Fenton process to quickly degrade aqueous effluents polluted by Acid Yellow 54 and Basic Blue 9 compared to the UV/H2O2 process.

OBJECTIVES: To compare transmitted fluorescence (Olympus CX21 (TM)/FRAEN FluoLED (TM)) with epi-fluorescence (Zeiss Primostar iLED (TM)) light-emitting diode microscopes (LED-FM) and various auramine staining and destaining/counterstaining techniques for the detection of acid-fast bacilli. DESIGN: Multicentre blinded reading of routine smears on both types of microscopes using different staining techniques in multiple phases. LED-FM rechecking of discordant series with and without restaining to calculate operating characteristics. RESULTS: Among 64 874 smears, both instruments detected 9.6% positives. Compared to the standard technique, the stronger auramine-O formulation did not perform better. Thiazine red counterstaining tended to yield more false-positive as well as false-negative errors. Combined destaining/counterstaining (sensitivity 93%, positive predictive value [PPV] 98%) proved significantly less effective. Both destaining with 1% hydrochloric acid (HCl) and 10% alcohol and the standard 0.5% HCl and 70-95% alcohol were equally accurate (sensitivity 95-96%, PPV 99%). The sturdiness of the microscopes in field conditions was sub-optimal: only 5/16 instruments did not break down. CONCLUSIONS: Both microscopes performed equally well. The standard staining technique is as good as the more complicated and expensive variations. A destaining solution containing only 10% alcohol works perfectly well. The inferior quality of LED-FM microscope components is an obstacle to FM expansion.

Prolongation of skin graft survival in mice by an azaphenothiazine derivative

Azaphenothiazines are predominantly immunosuppressive compounds. We evaluated the efficacy of an azaphenothiazine derivative, 6-chloroetlaylureidoethyldiquino[3,2-b;2′,3′-e][1,4]thiazine (DQT) in prolongation of survival of skin allografts between BALB/c and C5781/6 mice. The mice were treated intraperitoneally (i.p.) with 100 mu g of DQT on alternate days, on days 1-13 of the experiment (7 doses). The effect of DQT on a two-way mixed lymphocyte reaction (MLR) in the human model, as well as its effect on production of TNF alpha and IL-10 in a whole blood cell culture, stimulated by lipopolysaccharide (LPS), were evaluated. In addition, DQT effects were investigated regarding the proportion of T cell subsets in human peripheral blood lymphocytes (PBMC) by flow cytometry. Lastly, the effect of DQT on expression of signaling molecules involved in pro apoptotic pathways was determined by RT PCR. The results showed that DQT significantly extended skin graft survival. The compound also strongly suppressed two-way MLR in the human model at a concentration range of 2.5-5.0 mu M. In addition, DQT inhibited LPS-inducible TNF alpha, but not IL-10 production. The compound preferentially caused a loss of the CD3-CD8 +CD11b + PBMC cell subset, and transformed CD3 + CD8 +(high) into CD3 + CD8+(low) cells. Lastly, we demonstrated significant increases in expression of caspases (in particular caspase 8) and of p53 in a culture of Jurkat T cells. We conclude that the immunosuppressive actions of the compound in allograft rejection may be predominantly associated with induction of cell apoptosis and inhibition of TNF a production. The apoptosis could be predominantly selective for the CD3-CD8 + CD11b + cell phenotype.
