For next-generation electronics, it is crucial to be able to accurately mimic the nature of human skin and develop skin-inspired materials and devices for electronic skin (e-skin) applications. However, it is a great challenge to design such a materials because they require the complex integration of stretchability, skin adhesiveness, transparency, healability, and tactile sensing. Herein, we present the first dual covalently reversible dynamic bond-based polymer that can be integrated with various functional materials and used as a skin-like sensor. A series of polymers was designed using polypropylene glycol (PPG)-based polyurethane (PU) as the polymer backbone, and these polymers could integrate two types of covalent reversible dynamic bonds, Diels-Alder and disulfide bonds, in one polymer for the first time. Based on our design, a polymer was created with the advantages of high stretchability, fast healing abilities (60 degrees C, 5 min), skin adhesion, recyclability, high transparency, and tactile sensing abilities. We provide an alternative strategy for the construction of skin-inspired multifunctional materials for e-skin applications.

Molecular probes featuring promising capabilities including specific targeting, high signal-to-noise ratio, and in situ visualization of deep tissues are in great demand for tumor diagnosis and therapy. F-19 magnetic resonance imaging (MRI) techniques incorporating stimuli-responsive probes are anticipated to be highly beneficial for specific detection and imaging of tumors because of negligible background and deep tissue penetration. Herein, we report a cascaded multi-responsive self-assembled nanoprobe, which enables sequential redox-triggered and near-infrared (NIR) irradiation-induced F-19 MR signal activation/amplification for sensing and imaging. Specifically, we designed and synthesized a cascaded multiresponsive F-19-bearing nanoprobe based on the self-assembly of amphiphilic redox-responsive F-19-containing polymers and NIR-absorbing indocyanine green (ICG) molecules. It could realize the activation of F-19 signals in the reducing tumor microenvironment and subsequent signal amplification via the photothermal process. This stepwise two-stage activation/amplification of F-19 signals was validated by F-19 NMR and MRI both in vitro and in vivo. The multiresponsive F-19 nanoprobes capable of cascaded F-19 signal activation/amplification and photothermal effect exertion can provide accurate sensing and imaging of tumors.

A multifunctional drug delivery system (MDDS) based on near-infrared (NIR) imaging was developed in various conformations for the accurate diagnosis and therapy of cancer. However, it remains challenging to develop an ideal structure for an MDDS with both exact and variable functionality. Herein, we report a fluorescence color-changeable branched-form heptamethine cyanine dye (PEG-4-CyP) that has both a simple structure and various functionality. The PEG-4-CyP was not only able to construct the nanoparticle (NP) structure by encapsulating DOX (PEG-4-CyP/DOX), but also showed change in the NIR fluorescence color by reacting with glutathione (GSH). The PEG-4-CyP/DOX was broken down in the redox microenvironment inside cancers because of the reduction reaction of PEG-4-CyP to GSH. As a result, the NP disruption, DOX release, and reactivity of PEG-4-CyP to GSH were visualized by NIR fluorescence color-change. Moreover, PEG-4-CyP/DOX showed an excellent chemophototherapeutic effect in cancer cells. This NIR fluorescence color-change strategy demonstrates the applicability of PEG-4-CyP as a novel MDDS able to provide a variety of information with a simple NP structure. 5-Carboxylcytosine (5caC) plays a vital role in the dynamics of DNA demethylation, and sequencing of its sites will help us dig out more biological functions of 5caC. Herein, we present a novel chemical method to efficiently label 5caC distinguished from other bases in DNA. Combined with bisulfite sequencing, 5caC sites can be located at single-base resolution, and the efficiency of 5caC labeling is 92% based on the Sanger sequencing data. Furthermore, dot blot assays have confirmed that 5caC-containing DNA isolated from HeLa cells was successfully labeled using our method. We expect that our strategy can be further applied to selectively tagging other carboxyl-modified bases and mapping their sites in RNA.

Objective: To determine the pattern of presentations and characteristics of patients at a psychiatric emergency services facility. Methods: The cross-sectional study was conducted in November 2016 at the Department of Psychiatry and Behavioural Sciences, Jinnah Postgraduate Medical Centre, Karachi, and comprised all patients presenting to the departmental emergency services. Data was documented on a pre-designed semi-structured proforma, and analysed using SPSS 22. Results: Of the 316 cases, 162(51.3%) were males and 154(48.7%) were females. The overall mean age was 30.78+/-13.09 years (range: 8-80 years). Psychiatric emergencies comprised suicidal attempt, excitement, violence, altered sensorium, altered/inappropriate behaviour and extrapyramidal symptoms / acute dystonia together constituting for 123(38.9%) of the total presentations, while there were 20(6.33%) cases of hysterical fits and 6(1.89%) with acute anxiety symptoms. Overall, 56(17.7%) subjects were found to have no psychiatric diagnosis, and 46(81.5%) of these were referred cases. Conclusion: Non-emergency visits constituted high percentage of emergency presentation.

Multidrug resistance (MDR) of cancer cells is a significant challenge in chemotherapy, highlighting the urgent medical need for simple and reproducible strategies to reverse this process. Here, we report the development of an active tumor-targeting and redox-responsive nanoplatform (PA-ss-NP) using hyaluronic acid-g-cystamine dihydrochloride-poly-epsilon-(benzyloxycarbonyl)-L-lysine (HA-ss-PLLZ) to co-deliver paclitaxel (PTX) and apatinib (APA) for effective reversal of MDR. This smart nanoplatform specifically bound to CD44 receptors, leading to selective accumulation at the tumor site and uptake by MCF-7/ADR cells. Under high concentrations of cellular glutathione (GSH), the nanocarrier was degraded rapidly with complete release of its encapsulated drugs. Released APA effectively inhibited the function of the P-glycoprotein (P-gp) drug pump and improved the sensitivity of MDR cells to chemotherapeutic agents, leading to the recovery of PTX chemosensitivity in MDR cells. As expected, this newly developed intelligent drug delivery system could effectively control MDR, both in vitro and in vivo.

Molecular machines are ubiquitous in nature and function away from equilibrium by consuming fuels to produce appropriate work. Clinical application of drug cocktails for cancer therapy is limited by their severe systemic toxicity. To solve a catch-22 dilemma between safety and efficacy for drug cocktails, a heterotargeted nanococktail (PPPDMA) with traceless linkers is developed. In the PPPDMA nanogel, a heterotargeting strategy is employed to improve its tumor selective targeting efficacy by overcoming the cancer cell monoligand density limitation. Benefitting from its glutathione and reactive oxygen species responsiveness, the loaded paclitaxel and doxorubicin can be quickly and tracelessly released into the cytoplasm in their original form, which bestows upon PPPDMA nanogels the capability to overwhelm the processing capacity of the cancer cell’s P-glycoprotein efflux pump, and ultimately kill them without inducing side effects. The PPPDMA treatment reduced its tumor burden over 99% (in tumor weight) and 96% (in tumor number). Most importantly, no detectable tumor in more than half of the PPPDMA treated mice was observed. It is concluded that traceless linker and heterotargeted nanococktail strategy can be a safe and effective approach for cancer treatment.

Galectin-3 (gal-3) plays a crucial role in various cellular events associated to tumor metastasis and progression. In this direction, gal-3 binding core-shell glyconanoparticles based on citrus pectin (CP) have been designed for targeted, trigger-responsive combination drug delivery. Depolymerization via periodate oxidation in heterogeneous medium yielded low-molecular weight dialdehyde oligomers (CPDA) of CP with a gal-3 binding property (K-d = 160.90 mu M). CPDA-based core-shell nanoparticles prepared to enhance the gal-3 binding specificity via a multivalent ligand presentation have shown to reduce homotypic cellular aggregation, tumor cell binding with endothelial cells, and endothelial tube formation, the major steps involved in the progression of cancer. Immune-fluorescence and flow cytometric analysis confirmed significant reduction in gal-3 expression on MDA-MB 231 cancer cells upon incubation with nanoparticles. An on-demand tumor microenvironment-responsive release of drugs at low pH and high concentrations of glucose and glutathione prevailing in tumor milieu was achieved by introducing a cleavable Schiff’s base, a boronate ester, and disulfide linkages within the shell of the nanoparticles. Nanoparticles with encapsulated sulindac in the core and doxorubicin (DOX) in the shell demonstrated target specificity and enhanced internalization with synergistic cytotoxic effects with a 30-fold reduction in IC50 in DOX-resistant, triple-negative MDA-MB 231 breast cancer cells. Nanoparticles were radiolabeled with 131I radioisotopes with >= 80% efficiency while retaining its gal-3 binding property. Biodistribution studies of radiolabeled placebo nanoparticles and drug-loaded CPDA nanoparticles demonstrated proof of concept of gal-3 targeting seen as preferential accumulation in the gal-3-expressing tissues of the gastric tract. The CPDA core-shell nanoparticles are thus promising platforms for gal-3 targeting and inhibition of gal-3-mediated processes involved in cancer progression with a potential of radiolabeling for in vivo monitoring or delivering therapeutic doses of radiation and on-demand triggered, target-specific drug release.
