Research on metal-free room-temperature phosphorescent (RTP) materials has been a hot topic and made great advances in recent years. However, developing amorphous metal-free RTP compounds whose RTP emission is insensitive to water and oxygen to expand their applications still remains a challenge. Two novel metal-free RTP compounds were facilely designed and synthesized by modifying ureidopyrinidinone (UPy) units on both ends of the phosphors (BrNpA and BrBA). On the basis of their water/oxygen insensitive phosphorescence at different emission wavelengths, pure white-light (CIE coordinates (0.32, 0.33)) emission was obtained by mixing these two compounds. Moreover, visualization of latent fingerprints employing RTP materials as the first application in forensic science, as well as protected quick response codes, was achieved by theses hydrophobic materials.

Molecular probes featuring promising capabilities including specific targeting, high signal-to-noise ratio, and in situ visualization of deep tissues are in great demand for tumor diagnosis and therapy. F-19 magnetic resonance imaging (MRI) techniques incorporating stimuli-responsive probes are anticipated to be highly beneficial for specific detection and imaging of tumors because of negligible background and deep tissue penetration. Herein, we report a cascaded multi-responsive self-assembled nanoprobe, which enables sequential redox-triggered and near-infrared (NIR) irradiation-induced F-19 MR signal activation/amplification for sensing and imaging. Specifically, we designed and synthesized a cascaded multiresponsive F-19-bearing nanoprobe based on the self-assembly of amphiphilic redox-responsive F-19-containing polymers and NIR-absorbing indocyanine green (ICG) molecules. It could realize the activation of F-19 signals in the reducing tumor microenvironment and subsequent signal amplification via the photothermal process. This stepwise two-stage activation/amplification of F-19 signals was validated by F-19 NMR and MRI both in vitro and in vivo. The multiresponsive F-19 nanoprobes capable of cascaded F-19 signal activation/amplification and photothermal effect exertion can provide accurate sensing and imaging of tumors.

A new method of supramolecular emulsion interfacial polymerization is developed, which can be used to fabricate supramolecular polymeric nanospheres. We designed a water-soluble monomer containing two maleimide end groups, acting as both building block and surfactant, and an oil-soluble supramonomer bearing two thiol groups connected by quadruple hydrogen bonds. With the assist of ultrasonication, hollow nanospheres can be controllably prepared by thiol-maleimide reaction of two monomers at the emulsion interface, which exhibit good stability and dynamic property. In addition, the encapsulated guest molecules could be controllably released from the supramolecular polymeric nanospheres, owing to their stimuli-responsiveness. It is anticipated that this approach will enrich the methodology of supramolecular polymerization and can be applied to constructing supramolecular materials with controllable structures and functions.

A series of isocyanate-free polyurethanes (NIPUs), showing an intrinsic thermo-healing performance, have been synthesized from sustainable vegetable oil-based cyclic carbonates and bio-based amines. The bio-based contents of the NIPUs are higher than 78%. Their thermo-mechanical and self-healing performances are tailored simply by varying the feed ratios of the amines employed. The biomass linoleic acid dimer-based diamine plays a positive role in maintaining the higher thermal stability and good elasticity of the prepared NIPUs, the cycloaliphatic isophorone diamine contributes to high tensile strength, and cysteine-derived disulfide-containing cystamine (CA) enhances the self-healing efficiency. The combination of intrinsic hydrogen bonds existed in the NIPU matrix with the dynamic exchange reactions of disulfide bonds and the enhanced flexibility of NIPU networks results in 98.1% self-healing efficiency at 25 degrees C. The contributions of disulfide bonds and hydrogen bonds to the healing efficiency were estimated by adjusting the composition of NIPUs, showing that the contribution of disulfide bonds to the healing ability is about 45.3% even at a low amount of CA (6.0 wt%).

Triptolide (TP) is one of the most common systemic treatments for inflammatory and immune diseases in China for centuries. However, TP exhibits some disadvantages, such as poor solubility in water, poor bioavailability, liver toxicity, renal toxicity, and other side effects. In order to reduce the adverse effects of TP, researchers have developed numerous strategies to address the adverse properties of triptolide. Nano-carrier-based triptolide delivery systems represent an emerging technology and are one of the strategies of nanomedicine that combines diagnostic and therapeutic applications in a single agent. In this approach, we developed a glutathione-activated carrier-free nanodrug of triptolide (CyssTPN) as a trackable drug delivery system. In this system, CyssTP self-assemble to form a carrier-free nanodrug, which possesses a monodisperse spherical morphology with hydrodynamic average sizes of about 50 nm. In addition, CyssTPN had good stability under different physiological conditions (pH, high salt, etc.). Apart from cellular imaging and cell uptake, CyssTPN can be tracked by the activation of TP ability in real-time and applied for cancer cell treatment efficiently. The result showed that CyssTPN could improve solubility, reduce the side effects, and increase the bioavailability of triptolide. It could also track triptolide activation timely and tumor therapy successfully.

Inhibition performance of 2,2′-diaminodiethyl disulfide (DA) was studied against mild steel (MS) corrosion in 0.5 M HCl. Electrochemical impedance spectroscopy (EIS), potentiodynamic (PD) polarization measurements were utilized to investigate the influence of inhibitor concentration and temperature on efficiency, as well as explanation of inhibition mechanism. Scanning electron microscopy (SEM) analysis was utilized to investigate the surface damages due to corrosion, in the absence and presence of inhibitor molecules. PD data revealed that the studied molecule exhibits mixed type inhibitor behavior on steel surface. Moreover, the calculated free adsorption energy (Delta G(ads)(o)) value is 38.45 kJ mol(-1), which indicates to strong adsorptive interaction by both physical and chemical means. UV-Visible study results supported the idea that there is strong interaction between the surface with the molecule, via its -S and -N atoms. As a consequence, 91.7% inhibition efficiency was determined in presence of 1.0 mM DA. (C) 2020 Elsevier B.V. All rights reserved.

RNA interference (RNAi) is an emerging therapeutic modality for tumors. However, lack of a safe and efficient small interfering RNA (siRNA) delivery system limits its clinical application. Here, we report a bioreducible and less-cationic siRNA delivery carrier by conjugating Zn(II)-dipicolylamine complexes (Zn-DPA) onto hyaluronic acid (HA) via a redox-sensitive disulfide (-SS-) linker. Such polymer conjugates can formulate stable siRNA nanomedicines via coordination between zinc ions of DPA and the anionic phosphate of siRNA. After the conjugates are taken up by cells, intracellular reduction stimulus subsequently triggers the release of siRNAs and elucidates the desired RNAi effect. Our studies showed the formulated siRNA nanomedicines can be efficiently delivered into tumor cells/tissues and mediates less cytotoxicities both in vitro and in vivo. More importantly, when applied in a xenograft glioblastoma tumor model, this siRNA nanomedicine demonstrated significantly enhanced antitumor ability comparing to naked siRNA. This work demonstrates that such bioreducible Zn-DPA-functionalized HA conjugates without using cationic material as a siRNA carrier represents a promising direction for RNAi-based cancer therapy.

Doxorubicin (DOX) resistance, which results in a reduced accumulation of DOX in the nucleus and hence decreased DNA damage, is a major challenge for chemotherapy against hepatocellular carcinoma. In this paper, we combined chemotherapy with photodynamic therapy (PDT) to combat DOX-resistant human hepatocellular carcinoma cells. We have prepared the polymeric micelles conjugating with DOX and zinc(II) phthalocyanine (ZnPc) through a pH-responsive hydrazone linker and a glutathione (GSH)-responsive disulfide linker, respectively. The polymeric micelles (DOX-ZnPc-micelles) exhibited a spherical shape with a size of about 98 mn diameter and showed excellent stability in aqueous solution. Due to the self-quenching of the ZnPc inside the micelles, DOX-ZnPc-micelles did not emit fluorescence upon red light irradiation. Drug release experiments verified that DOX and ZnPc could be released under acidic conditions and reducing environments, respectively. A higher concentration of DOX was internalized into DOX-resistant R-HepG2 cells through the delivery of polymeric micelles when compared with the free DOX, hence DOX-ZnPc-micelles exhibited a significant enhancement in anticancer activity. The IC50 value of DOX against R-HepG2 cells was found to be 21 mu M when combined with PDT and it was 5-fold less than that of a single treatment of DOX (102 mu M). The DOX-ZnPc-micelles could induce cell apoptosis and necrosis on R-HepG2 cells by combined therapeutic modalities, while these micelles induced only apoptosis on IiepG2 cells. We have demonstrated that utilization of polymeric micelles can significantly enhance the cellular uptake and cytotoxicity of DOX against R-HepG2 cells when compared with free DOX. Moreover, PDT can act as an adjuvant therapeutic modality and combine with chemotherapy to further improve therapeutic efficacy. Overall speaking, DOX-ZnPc-micelles can overcome DOX resistance and induce a synergistic therapeutic effect against DOX-resistant R-HepG2 cells, hence improving the therapeutic efficacy when compared with monotherapy.

Recent advances in the development of protein-based vaccines have expanded the opportunities for preventing and treating both infectious diseases as well as cancer. However, the development of readily and efficient antigen delivery systems capable of stimulating strong cytotoxic T-lymphocyte (CTL) responses remains a challenge. With the attempt to closely mimic the properties of viruses in terms of their size and molecular organization, we constructed RNA (which is a ligand for Toll-like receptor 7 (TLR7) and TLR8) and antigen-loaded nanoparticles resembling the structural organization of viruses. Cationic polymers containing either azide or bicyclo[6.1.0]nonyne (BCN) groups were synthesized as electrostatic glue that binds negatively charged single stranded RNA (PolyU) to form a self-crosslinked polyplex core. An azide-modified model antigen (ovalbumin, OVA) and a BCN-modified mannosylated or galactosylated polymer were sequentially conjugated to the RNA core via disulfide bonds using copper free click chemistry to form the shell of the polyplexes. The generated reducible virus mimicking particles (VMPs) with a diameter of 200 nm and negatively surface charge (-14 mV) were colloidally stable in physiological conditions. The immunogenicity of these VMP vaccines was evaluated both in vitro and in vivo. The surface mannosylated VMPs (VMP-Man) showed 5 times higher cellular uptake by bone marrow derived DCs (BMDCs) compared to galactosylated VMP (VMP-Gal) counterpart. Moreover, VMP-Man efficiently activated DCs and greatly facilitated MHC I Ag presentation in vitro. Vaccination of mice with VMP-Man elicited strong OVA-specific CTL responses as well as humoral immune responses. These results demonstrate that the modular core-shell polymeric nanoparticles described in this paper are superior in inducing strong and durable immune responses compared to adjuvanted protein subunit vaccines and offer therefore a flexible platform for personalized vaccines.
