Facile, Odourless, Quantitative Synthesis of 3-Hydroxy-3,4-dihydro-2H-1,4-thiazines

A sulfa-Michael addition/formal [3+3] annulation cascade reaction between 1,4-dithiane-2,5-diol and 1,2-diaza-1,3-dienes by a one-pot base-catalysed strategy was developed. This metal-free and odourless method provides a practical and facile approach to 1,4-thiazines obtained in total atom economy and in quantitative yields. Furthermore, the synthesis could be scaled-up to the gram scale.

Heterocyclization of Cyanoacetamide Derivatives: Synthesis and Biological Activity of Novel Azoles and Azines

The intermolecular cyclization of N-benzyl-2-cyanoacetamide with carbon disulfide followed by intramolecular cyclization gave thioxothiazinone 3. This compound was used to synthesize a series of novel fused furopyrrole, pyridine, pyrimidine and other azine and azole derivatives. The Michael-type reaction of compound 3 with maleic anhydride followed by pyrrole and furan cyclizations and aromatization yielded polycyclic compound 7. The [3+3]-cycloaddition of benzylidene malononitrile and its derivative to compound 3 gave pyridothiazines 10-12. The ring opening in compound 3 under the action of urea or thiourea followed by pyrimidine cyclization and subsequent air oxidation resulted in the synthesis of oxa- and thiadiazolopyrimidinones 15 and 16, respectively. The reaction of compound 3 with H2O2 in a basic medium provided pyrimidine derivative 17. The oxidation of compound 3 with Br-2 in an acid medium led to bromo derivative 19. The synthesized novel compounds were characterized by elemental analysis and IR and H-1 and C-13 NMR spectroscopy and tested antibacterial and anticancer activities.

Synthesis and antimicrobial activity of new derivatives of pyrano[4”,3”:4′,5′]pyrido[3′,2′:4,5]thieno[3,2-d]pyrimidine and new heterocyclic systems

Taking into account previously obtained biological results on some polyheterocyclic compounds (containing different heteroatoms) and in particular on several 8-amino-5-isopropyl-2,2-dimethyl-10-(methylthio)-1,4-dihydro-2H-pyrano[4”,3”:4′,5′]pyrido[3′,2′:4,5]thieno[3,2-d]pyrimidines Ia-v we have carried out the synthesis of twentyone 8-amino-5-isobutyl-2,2-dimethyl-10-(methylthio)-1,4-dihydro-2H-pyrano[4”,3”:4′,5′]pyrido[3′,2′:4,5]thieno[3,2-d]pyrimidines 6. Therefore we have slightly modified the structure of the previously studied I introducing at C-5 an isobutyl group instead of the previously examined isopropyl ones in order to see if this variation (changing a little the lipophilicity) will affect the biological activity. Furthermore thieno[3,2-d]pyrimidine-8-thione 7 and their S-alkylated 8 were synthesized. Finally by alkylation of 5-isobutyl-2,2-dimethyl-10-thioxo-1,4,10,11-tetrahydro-2H-pyrano[4”,3”:4′,5′]pyrido[3′,2′:4,5]thieno[3,2-d]pyrimidin-8(9H)-one 3 with alkyl dichlorides (bifunctional reagents) we realized the cyclization of a thiazole or thiazine ring on the [b] side of the pyrimidine ring with formation of the new condensed pentaheterocyclic systems: pyrano[4”,3”:4′,5′]pyrido[3′,2′:4,5]thieno[3,2-d][1,3]thiazolo[3,2-a]pyrimidin-8-one 11 and pyrano[4”’,3”’:4”,5”]pyrido[3”,2”:4′,5′]thieno[3′,2′:4,5]pyrimido[2,1-b][1,3]thiazin-8-one 12. It was found that some of the synthesized compounds showed interesting antimicrobial activity (by agar diffusion method) against some gram-positive and gram-negative bacilli strains. [GRAPHICS] .

Crystal structure of 2-(4-methylphenyl)-4H-1,3-benzothiazine

In the title compound, C15H13NS, the thiazine ring adopts a boat conformation. The dihedral angle between the planes of the benzene ring of the benzothiazine unit and the tolyl ring is 19.52 (9)degrees. In the crystal, molecules are linked by weak C-H center dot center dot center dot pi interactions into a tape structure along the b-axis direction.

Inhibition of heat shock protein (Hsp) 90 potentiates the antiproliferative and pro-apoptotic effects of 2-(4 ‘ fluoro-phenylamino)-4H-1,3-thiazine[6,5-b]indole in A2780cis cells

Ovarian carcinoma is initially sensitive to platinum-based therapy, but become resistant over time. The study of cancer sensitizing substance is therefore the major challenge for a number of scientific groups. Our experiments were carried out on human ovarian adenocarcinoma A2780cis cells resistant to cisplatin and their response to 2-(4’fluoro-phenylamino)-4H-1,3-thiazine[6,5-b] indole (thiazine[6,5-b] indole) and/or heat shock protein (Hsp) 90 inhibitor 17-dimethylaminoethylamino-17-demethoxygeldanamycin (17-DMAG) using proliferation assay, cell cycle analysis and monitoring of apoptosis were examined. A2780cis cells revealed the same fold of resistance to Hsp90 inhibitor 17-DMAG as it is declared for cisplatin (18 times), but only 3.2 times for thiazine[6,5-b] indole. Our results showed that the combination of thiazine[6,5-b] indole and 17-DMAG significantly reduced proliferation of A2780cis cells and led to their accumulation in G2/M phase of the cell cycle. Moreover, both thiazine[6,5-b] indole as well as 17-DMAG increased the number of annexin V positive A2780cis cells in time dependent manner. Interestingly, thiazine[6,5-b] indole treatment significantly activated also caspase-3 compared to untreated or 17-DMAG-treated cells and reduced mitochondrial membrane potential (MMP) of A2780cis cells with more significant decline after combined treatment. In this regard, the incubation of A2780cis cells with thiazine[6,5-b] indole induced PARP protein cleavage as well as an increased level of Bad protein with more pronounced changes after combined treatment. Importantly, Hsp70 protein was not upregulated in A2780cis cells neither by individual treatment nor by mutual combination. Our results signify antiproliferative and pro-apoptotic effects of novel thiazine[6,5-b] indole potentiated by Hsp90 inhibitor 17-DMAG in ovarian adenocarcinoma cells resistant to cisplatin and therefore represents new strategy in cancer treatment. Synthesis of microgels based on carboxymethylcellulose cross-linked with zinc(II) ions and heterocyclic effectors of NO-synthase

Convenient synthesis path of nanostructured water-soluble microgels (PMG) based on carboxymethylcellulose cross-linked with zinc and ligand N-(5,6-dihydro-4H-1,3-thiazine-2 yl) benzamide and N-(4- isopropyl-phenyl)-N-(1-iminoethyl)piperidine-1-carbothioamide hydrobromides is described. Being insoluble in aqueous solutions the complex of zinc with the ligand acquires the solubility in water when dispersed in nanocontainers of PMG. Microgels were obtained by varying the content of zinc-ions in the reaction mixture. A new method for the determination of zinc in PMG composite using sulfarsazene is proposed. The resulting nanoparticles have been tested for cytotoxicity. It was established cytotoxicity of nanocontainers strongly depends on the concentration of zinc and ligand in composition of PMG.

Synthesis and selected immunological properties of 10-substituted 1,8-diazaphenothiazines

A new type of tricyclic azaphenothiazines-1,8-diazaphenothiazines-was obtained in the reaction of 2,3- and 3,4-disubstituted pyridines. The reaction ran as the Smiles rearrangement. The 1,8-diazaphenothiazine system was determined using NOE experiment and 2D NMR spectra (COSY, HSQC, HMBC). 10H-1,8-diazaphenothiazine was transformed into 10-derivatives with alkyl, aminoalkyl, amidoalkyl, sulfonamidoalkyl, and nitrogen half-mustard groups. The compounds were tested for their effects on phytohemagglutinin A-induced proliferative response of human peripheral blood mononuclear cells (PBMC) and lipopolysaccharide-induced tumor necrosis factor alpha production by human whole blood cultures. The compounds exhibited differential, dose-dependent inhibitory activities in these tests. All the compounds were low toxic against PBMC. The compounds showing the highest antiproliferative activity strongly inhibited the growth of leukemia L-1210 and colon cancer SW-948 cell lines, similarly as cisplatin, a reference drug.

A New Metabolite with a Unique 4-Pyranone-gamma-Lactam-1,4-Thiazine Moiety from a Hawaiian-Plant Associated Fungus

An endophytic fungus Paraphaeosphaeria neglecta FT462 isolated from the Hawaiian-plant Lycopodiella cernua (L.) Pic. Serm produced one unusual compound (1, paraphaeosphaeride A) with the 4-pyranone-gamma-lactam-1,4-thiazine moiety, along With two new compounds (2 and 3, paraphaeosphaerides B and C, respectively) and the known compound (4). Compounds 1-3 were characterized by NMR and MS spectroscopic analysis. The absolute configuration of the 3-position of compound 1 was determined as S by electronic circular dichroism (ECD) calculations. Compound 3 also Showed STAT3 inhibition at 10 mu M.

Reinvestigations into synthesis of allyldithiocarbamates and their intramolecular cyclization: synthesis and antihyperglycemic activity of 2-thioxothiazolidine-4-alkanoates

Reinvestigating the synthesis of allylthiocarbamates from MBH derivatives revealed that they are obtained more efficiently from allylbromides in isopropanol rather than acetates in water. Further unlike reported, the base-mediated intramolecular cyclization of these allyldithiocarbamates give substituted 2-thioxothiazolidine-4-alkanoates instead of substituted 1,3-thiazines and the reaction was restricted to 2- or 4-nitrophenyl carrying substrates. Screening of these products for antihyperglycemic activity revealed that 5bC displays significant activity via partial agonist activity on PPAR gamma at transcriptional level, which was corroborated by molecular docking studies. A Single-Reagent- Driven Multistep One-Pot Preparation of Thiazolines and 1,3-Thiazines from Aldoximes, Nitriles, and Carboxylic Acids

N-(omega-Bromoalkyl)dichlorothiophosphoramidates have been designed as a new class of reagent for the single-reagent-driven multistep preparation of 2-substituted thiazolines and 1,3-thiazines from aldoximes, nitriles, or carboxylic acids. A wide range of substrates both aromatic as well as aliphatic were investigated and found suitable for the developed protocol.
