The quantification of proteins and peptides becomes more important besides mere identification in modern life sciences. Therefore, we have developed a new reagent that adds to the known metall-coded affinity tagging strategy employed in molecular and elemental mass spectrometry containing a photocleavable linker. A synthesis route was developed that provides the new reagent in good yields. The stability of the synthesized reagents was assessed under different temperature and illumination conditions. Labeling reactions were carried out at peptide and protein level, while also the fragmentation behavior of labeled peptides was assessed. In additional experiments, the photocleavability of the new reagent was examined. Upon irradiation with ultraviolet light, the photoproducts were liberated and could be used for quantification of labeled peptides.

The aim of this work was to obtain a novel onco-theranostic system for early diagnosis and improved chemotherapeutic outcome. Hybrid nanogels with magnetic and dual responsive properties were fabricated by covalently attaching superparamagnetic iron oxide nanoparticles (SPIONs) with a disulfide-modified alginate derivative, while simultaneously encapsulating the anticancer drug doxorubicin. The resulting nanogels exhibited magnetic-targeted characteristics, high drug loading content, co-triggered release behavior, high toxicity to tumor cells, low side effects to normal cells, and magnetic resonance imaging (MRI) functions. These findings proved that the hybrid nanogels have great potential as novel tumor-targeting nano-theranostic agents for simultaneous MRI imaging and efficient anti-tumor treatment.

BACKGROUND Decontamination of water by means of photocatalysis is seen as a sustainable solution. However, catalysts with low recovery from liquid suspension remain the main predicament that obstructs their industrial applications. To overcome this shortcoming, a methodology is reported here for the functionalization of nanospinel nickel zinc ferrite by cystamine ligand to develop Ni0.5Zn0.5Fe2O4@cystamine magnetic recyclable catalyst for water decontamination applications. RESULTS The diffraction patterns of bare and functionalized nanoferrites indicated that the cubic inverse spinel structure of the particles was not changed after cystamine functionalization. X-ray photoelectron spectroscopy (XPS) and Raman spectroscopy analyses confirmed the chemical bonding between cystamine and spinel ferrites. The spherical morphology and elemental composition were studied through high-resolution transmission electron spectroscopy (HRTEM) images and energy-dispersive X-ray (EDX) analysis respectively. Deeper understanding of the catalyst’s optical behavior and calculation of the energy band gap were achieved by means of photoluminescence spectroscopy. Magnetic saturation values were calculated from vibrating sample magnetometer (VSM) analysis, and almost negligible change was found. A possible photocatalytic mechanism for the removal of organic dye is proposed. CONCLUSION The present findings imply that the presence of amine groups on the catalyst surface, good dispersibility stability and outstanding catalytic activity make functionalized nanoferrite a suitable candidate for industrial wastewater treatment. Easy synthesis, easy work-up and short reaction times are some of the advantages of the methodology. (c) 2019 Society of Chemical Industry

A series of isocyanate-free polyurethanes (NIPUs), showing an intrinsic thermo-healing performance, have been synthesized from sustainable vegetable oil-based cyclic carbonates and bio-based amines. The bio-based contents of the NIPUs are higher than 78%. Their thermo-mechanical and self-healing performances are tailored simply by varying the feed ratios of the amines employed. The biomass linoleic acid dimer-based diamine plays a positive role in maintaining the higher thermal stability and good elasticity of the prepared NIPUs, the cycloaliphatic isophorone diamine contributes to high tensile strength, and cysteine-derived disulfide-containing cystamine (CA) enhances the self-healing efficiency. The combination of intrinsic hydrogen bonds existed in the NIPU matrix with the dynamic exchange reactions of disulfide bonds and the enhanced flexibility of NIPU networks results in 98.1% self-healing efficiency at 25 degrees C. The contributions of disulfide bonds and hydrogen bonds to the healing efficiency were estimated by adjusting the composition of NIPUs, showing that the contribution of disulfide bonds to the healing ability is about 45.3% even at a low amount of CA (6.0 wt%).

A water-soluble cyclophane dimer having two disulfide groups as a reduction-responsive cleavable bond as well as several acidic and basic functional groups as a pH-responsive ionizable group 1 was successfully synthesized. It was found that 1 showed pH-dependent guest-binding behavior. That is, 1 strongly bound an anionic guest, 6-p-toluidinonaphthalene-2-sulfonate (TNS) with binding constant (K/M-1) for 1:1 host-guest complexes of 9.6 x 10(4) M-1 at pH 3.8, which was larger than those at pH 7.4 and 10.7 (6.0 x 10(4) and 2.4 x 10(4) M-1, respectively), indicating a favorable electrostatic interaction between anionic guest and net cationic 1. What is more, release of the entrapped guest molecules by 1 was easily controlled by pH stimulus. Large favorable enthalpies (Delta H) for formation of host-guest complexes were obtained under the pH conditions employed, suggesting that electrostatic interaction between anionic TNS and 1 was the most important driving force for host-guest complexation. Such contributions of Delta H for formation of host-guest complexes decreased along with increased pH values from acidic to basic solutions. Upon addition of dithiothreitol (DTT) as a reducing reagent to an aqueous PBS buffer (pH 7.4) containing 1 and TNS, the fluorescence intensity originating from the bound guest molecules decreased gradually. A treatment of 1 with DTT gave 2, having less guest-binding affinity by the cleavage of disulfide bonds of 1. Consequently, almost all entrapped guest molecules by 1 were released from the host. Moreover, such reduction-responsive cleavage of 1 and release of bound guest molecules was performed more rapidly in aqueous buffer at pH 10.7.

Crosslinked polymer nanomaterials have attracted great attention due to their stability and highly controllable drug delivery; herein, a series of well-defined amphiphilic PDPA-b-P(NMS-co-OEG) diblock terpolymers (P1-P3) were designed and prepared via RAFT polymerization and were self-assembled into non-cross-linked (NCL) nanomicelles, which were further prepared into shell-cross-linked (SCL) micelles via cystamine-based in situ shell cross-linking. The application of combination immune-chemotherapy makes up for the limitation of monotherapy and achieves superior antitumor activity against cancer. However, combinational therapy is always restricted by poor tumor targeted drug delivery efficacy. Herein, novel T cell membrane cloaking tumor microenvi-ronment-responsive nanoparticles (PBA modified T cell membrane cloaking hyaluronic acid (HA)-disulfide bond-vitamin E succinate/curcumin, shortened as RCM@T) were developed. T cell membrane cloaking not only serves as a protection shell for sufficient drug delivery but also acts as a programmed cell death-1(PD-1) antibody to selectively bind the PD-Li of tumor cells. When RCM@T is intravenously administrated into the blood stream, it accumulates at tumor sites and responds to an acidic pH to achieve a ‘membrane escape effect and expose the HA residues of RCM for tumor targeted drug delivery. RCM accumulates in the cytoplasm via CD44 receptor mediated endocytosis and intracellularly releases antitumor drug in the intracellular redox microenvironment for tumor chemotherapy. T cell membrane debris targets the PD-Llof tumor cells for tumor immunotherapy, which not only directly kills tumor cells, but also improves the CD8(+) T cell level and facilitates effector cytokine release. Taken together, the as-constructed RCM@T creates a new way for the rational design of a drug delivery system via the combination of stimuli-responsive drug release, chemotherapeutical agent delivery and cell membrane based immune checkpoint blockade immunotherapy.

X-ray photoelectron spectroscopy (XPS), contact angle, and protein adsorption measurements revealed that the R-f groups and PEG chains in PMFP were segregated to the outermost surface of the dip-coated layer. The surface segregation of these moieties produced a low-fouling surface on the PP substrate. In addition, we synthesized a terpolymer that contained R-f groups and PEG chains with carboxy groups at their termini (PMFB) and used it to dip-coat a PP substrate. The surface segregation of side chains in PMFB induced the presentation of carboxy groups at the outermost surface, which were used as reactive sites for enzyme immobilization.
