Characterization of a Novel Human-Specific STING Agonist that Elicits Antiviral Activity Against Emerging Alphaviruses

Pharmacologic stimulation of innate immune processes represents an attractive strategy to achieve multiple therapeutic outcomes including inhibition of virus replication, boosting antitumor immunity, and enhancing vaccine immunogenicity. In light of this we sought to identify small molecules capable of activating the type I interferon (IFN) response by way of the transcription factor IFN regulatory factor 3 (IRF3). A high throughput in vitro screen yielded 4-(2-chloro-6-fluorobenzyl)-N-(furan-2-ylmethyl)-3-oxo-3,4-dihydro-2H-benzo[b][1,4]thiazine-6-carboxamide (referred to herein as G10), which was found to trigger IRF3/IFN-associated transcription in human fibroblasts. Further examination of the cellular response to this molecule revealed expression of multiple IRF3-dependent antiviral effector genes as well as type I and III IFN subtypes. This led to the establishment of a cellular state that prevented replication of emerging Alphavirus species including Chikungunya virus, Venezuelan Equine Encephalitis virus, and Sindbis virus. To define cellular proteins essential to elicitation of the antiviral activity by the compound we employed a reverse genetics approach that utilized genome editing via CRISPR/Cas9 technology. This allowed the identification of IRF3, the IRF3-activating adaptor molecule STING, and the IFN-associated transcription factor STAT1 as required for observed gene induction and antiviral effects. Biochemical analysis indicates that G10 does not bind to STING directly, however. Thus the compound may represent the first synthetic small molecule characterized as an indirect activator of human STING-dependent phenotypes. In vivo stimulation of STING-dependent activity by an unrelated small molecule in a mouse model of Chikungunya virus infection blocked viremia demonstrating that pharmacologic activation of this signaling pathway may represent a feasible strategy for combating emerging Alphaviruses.

Zero-Power, Low-Cost Ultraviolet-C Colorimetric Sensor Using a Gallium Oxide and Reduced Graphene Oxide Hybrid via Photoelectrochemical Reactions

A zero-power, low-cost ultraviolet (UV)-C colorimetric sensor is demonstrated using a gallium oxide and reduced graphene oxide (rGO) hybrid via photoelectrochemical reactions. A wide bandgap semiconductor (WBS) such as gallium oxide with an energy bandgap of 4.9 eV generates electron-hole pairs (EHPs) when exposed under a mercury lamp emitting 254 nm. While the conventional UVC sensors employing WBS convert the generated EHPs into an electrical signal via a solid-state junction device (SSD), our newly proposed UVC sensory system works by converting EHPs into an electrochemical reaction. The electrochemical reaction causes the degradation of a cationic thiazine redox dye, methylene blue (MB) and thereby spontaneously changes its color. As more rGO was hybridized with the gallium oxide, MB degradation was effectively expedited. Thus, the level of MB degradation under UVC can be evaluated as a UVC indicator. Unlike conventional SSD-based UVC sensors, our responsive colorimetric sensor can be applied where needed inexpensively and zero power.

A comparative study for removal of different dyes over M/TiO2 (M = Cu, Ni, Co, Fe, Mn and Cr) photocatalysts under visible light irradiation

The purpose of this work is to design and develop a series of stable and effective TiO2 photocatalysts prepared by a modified precipitation method. In order to improve the photo response of the TiO2 to the visible region, the effect of addition of six successive transition metal elements of fourth period, namely chromium (Cr), manganese (Mn), iron (Fe), cobalt (Co), nickel (Ni), and copper (Cu) was studied. Two different types of dyes, methyl orange (MO, azo dye) and methylene blue (MB, thiazine dye group), were used to investigate the influence of structure of dyes on the photocatalytic degradation rates. The results showed that loading titania with 0.3 wt.% of different transition metal elements was found to have significant influences on the crystallographic structure, physical properties and optical absorption properties of titania based catalysts, as well as catalytic activity during dye degradation reactions. The optical reflection edge was obviously shifted to the visible light range with the transition metal additives. The band gap energies of the doped TiO2 samples were considerably narrower than that of plain TiO2. The different activity levels of the catalysts for both dyes clearly show that the photocatalytic activity of samples strongly depended on the metal dopant used. Among all the catalysts, Cu/TiO2 sample exhibited the highest photocatalytic activity under visible light for both of the dyes owing to the low band gap energy and delayed electron-hole recombination. A series of new derivatives of dipyrimido[4,5-b:5,4-e][1,4] thiazine were synthesized by treatment of 5-amino-6-methyl-2-morpholino-4-pyrimidinethiol (1) with 5-bromo-2,4-dichloro-6-methylpyrimidine (2) in the presence of triethylamine and ethanol and subsequently with various secondary amines in EtOH/DMF. The 15-LO inhibitory activities of the new synthesized compounds were also evaluated. The results show that compound 4e has the best IC50 of 15-LO inhibition (IC50 = 14.4 A mu M) but when all the products were theoretically docked into 15-LO, the GOLD scores of compounds 4e and 4d are 43.67 and 43.39 respectively, which show the best results. We suggest that the hydrogen bond interaction between Ser489 of 15-LO and the nitrogen of piperazine ring of compound 4e appears to play major role in lipoxygenase inhibition.

Residualization Rates of Near-Infrared Dyes for the Rational Design of Molecular Imaging Agents

Near-infrared (NIR) fluorescence imaging is widely used for tracking antibodies and biomolecules in vivo. Clinical and preclinical applications include intraoperative imaging, tracking therapeutics, and fluorescent labeling as a surrogate for subsequent radiolabeling. Despite their extensive use, one of the fundamental properties of NIR dyes, the residualization rate within cells following internalization, has not been systematically studied. This rate is required for the rational design of probes and proper interpretation of in vivo results. In this brief report, we measure the cellular residualization rate of eight commonly used dyes encompassing three core structures (cyanine, boron-dipyrromethene (BODIPY), and oxazine/thiazine/carbopyronin). We identify residualizing (half-life > 24 h) and non-residualizing (half-life < 24 h) dyes in both the far-red (similar to 650-680 nm) and near-infrared (similar to 740-800 nm) regions. This data will allow researchers to independently and rationally select the wavelength and residualizing nature of dyes for molecular imaging agent design. Media polarity and concentration roles on the third order nonlinear behaviors of thiazine dyes

Nonlinear optical materials play important roles in optics and photonics. In order to improve the performance of these materials, in this paper, we tried to find effective methods for controlling the nonlinear responses of thiazine dyes. Our experimental results indicate that molecular linear properties, media polarity, concentration and excitation beam properties with different contributions play significant roles on the saturable and reverse saturable absorption characteristics of thiazine dyes. In real, competition between these effective parameters leads to appearance of various third order nonlinear responses. Although by excitation wavelengths near the absorption peaks and at high powers, dye molecules tend to indicate reverse saturable absorption characteristics in polar protic solvents by increasing the concentration, saturable absorption characteristics were observed in polar aprotic solvents. Furthermore, the samples displayed the same behaviors in different solvent media at low powers and for excitation wavelengths far from absorption peaks. Therefore, by controlling the nonlinear responses of thiazine dyes, these materials can indicate both saturable and reverse saturable absorption characteristics that increase their applications in optics and photonics devices.
