Green Synthesis of Novel Naphtho[1,2-e]/benzo[e][1,3]thiazine Derivatives via One-Pot Three-Component Reaction Using Tetra n-Butyl Ammonium Bromide

A simple, convenient and green method catalyzed by tetra- n-butyl ammonium bromide (TBAB) has been reported for the synthesis of a series of novel naphtho[1,2-e]/ benzo[e][1,3]thiazines using a multicomponent reaction of thionaphthol/ thiophenol, aromatic amines and formaldehyde. The advantages of this method are good yields, solvent free condition, and environmentally benign catalyst.

Atom-economical isocyanide-based multicomponent synthesis of 2,5-dioxopyrrolidines, spirobenzothiazinechromans and 1,5-benzothiazepines

An efficient synthesis of new pharmaceutically relevant dioxopyrrolidines, spirobenzo thiazine-2,3’chromans, and benzothiazepines via isocyanide-based multicomponent condensation reactions at room temperature is reported. Synthesis of Some Novel Thiazolo[3,2-a]pyrimidine and Pyrimido[2,1-b][1,3]thiazine Derivatives and their Antimicrobial Evaluation

A 2-(2-Mercapto-4-(4-phenoxyphenyl)-6-(thiophen-2-yl)-1,6-dihydropyrimidin-5-yl) acetic acid was used as a reactive key precursor to design various pyrimidine derivatives such as thiazolo[3,2-a]pyrimidines and pyrimido[2,1-b][1,3]thiazines. The chemical structures of the newly synthesized products were confirmed by their elemental analyses and spectral data (IR, H-1 NMR, C-13 NMR, and mass spectra). The antibacterial and antifungal activities of some of the synthesized products were also evaluated, and it was found that compounds 3, 5, 9, and 11 exhibited potent activity against tested microorganisms in comparison with standard drugs.

NOVEL SYNTHESIS, RING TRANSFORMATION AND ANTICANCER ACTIVITY OF 1,3-THIAZINE, PYRIMIDINE AND TRIAZOLO[1,5-a]PYRIMIDINE DERIVATIVES

Synthesis, heterocyclization and anticancer activity of a new series of heterocyclic compounds are described. Aminothiazine 1 was obtained from the base induced condensation of thiourea, benzaldehyde and ethyl cyanoacetate. The synthesis of N-phenyl amino pyrimidine derivative 2 was obtained as a result of reaction of aniline with compound 1. Compound 2 underwent ring opening and recyclization upon reaction with HCl or H2O2/NaOH to afford the acid derivative 3 or oxazine 4, respectively. Thiazine 1 undergoes ring transformation upon the effect of NH2OH center dot HCl to produce pyrimidine derivative 5. Heterocyclization of compound 1 with thiosemicarbazide followed by oxidation with I-2/AcOH afforded triazolopyrimidine 6 and 7, respectively. Alkylation of compound 1 was promoted by reaction of 1 with ethyl iodide to give alkylated thiazine 8 which in turn undergo ring transformation when subjected to reaction with hydrazine hydrate to give pyrazole derivative 9. Refluxing of amino-1,3-thiazine derivative 1 with ethyl bromoacetate in the presence of Et3N produce the alkylated pyrimidine product 10. Hydrazonolysis of 1,3-thiazine 1 with hydrazine or phenyhydrazine gave pyrimidine derivatives 11a,b, respectively. Compound 11b was cyclized with carbon disulfide or formaldehyde to produce triazolopyrimidines 12 and 13, respectively. Some of the new compounds were screened for anticancer activity and significant results were found for some compounds.

A SIMPLE SYNTHESIS OF 4-HYDROXY-3,4-DIHYDROPYRIDO[3,4-d]PYRIMIDINE-2(1H)-THIONE DERIVATIVES BY THE REACTION OF 3-ISOTHIOCYANATOPYRIDIN-4-YL KETONES WITH PRIMARY AMINES

The reaction of aryl(3-isothiocyanatopyridin-4-yl)methanones, prepared easily from commercially available 3-aminopyridine, with primary amines at room temperature afforded 3-substituted 4-aryl-4-hydroxy-3,4-dihydropyrido[3,4-d]pyrimidine-2(1H)-thiones in good yields. These ketones could be converted to new tricyclic heterocyclic systems, 2,3-dihydro-5H-pyrido[3,4-d]thiazolo [3,2-a]pyrimidine and 3,4-dihydro-2H,6H-pyrido [3′,4′:4,5]-pyrimido[2,1-b] [1,3]thiazine, upon treatment with 2-bromoethanamine hydrobromide and 3-bromopropaneamine hydrobromide, respectively, in the presence two equivalents of triethylamine.

Versatile one-pot synthesis of benzo-fused thiacycles by copper catalysis

A novel, one-pot synthesis of structurally diverse benzo-fused thiacycles via a Cu-catalysed intermolecular C-S coupling/cyclisation tandem process, employing the same catalytic system, has been developed. Thus, 3,4-dihydro-2H-benzo[e][1,3]thiazines and 4H-benzo[e][1,3] thiazines were selectively obtained by this one-pot tandem process from (2-iodophenyl) methanamine and aldehydes or 1-(azido-methyl)-2-iodobenzene, respectively. These reactions proceeded in toluene at 100 degrees C with potassium thioacetate, thiobenzoate or ethyl xanthogenate in moderate to good isolated yields. The first one avoids the imine isolation step. A similar approach to benzo[b] thiophene derivatives from 2-(2- iodophenyl) acetonitrile was also developed.

Copper-Catalyzed Tandem Radical Cyclization of 8-Ethynyl-1-naphthyl-amines for the Synthesis of 2H-Benzo[e][1,2]thiazine 1,1-Dioxides and its Fluorescence Properties

A copper-catalyzed radical cascade dehydrogenative cyclization of N-tosyl-8-ethynyl-1-naphthylamines under air is described herein for the synthesis of thioazafluoranthenes. The reaction proceeds smoothly with high efficiency and a broad reaction scope. The product is indeed a new fluorophore and its photophysical properties are also investigated. Based on the results, we are pleased to find that the Stokes shift of amino-linked thioazafluoranthenes in dilute tetrahydrofuran is determined to be 143 nm (4830 cm(-1)).

Tetrazolo- and 1,2,4-oxadiazolo-fused derivatives of the antitumour, antibiotic, DNA-interactive pyrrolo [2,1-c][1,4]benzodiazepines and their pyrrolobenzothiadiazepine derivatives have been produced as analogues of a 1,2,3-triazolo-fused pyrrolobenzothiadiazepine, which was shown to be a Glut-1 transporter inhibitor with potential as an antitumour agent. The tetrazolo-fused systems were produced by intramolecular 1,3-dipolar cycloaddition between an azide and a nitrile. The 1,2,4-oxadiazolo systems were produced by nitrile oxide cycloadditions to pyrrolobenzothiadiazepines, which were in turn produced from a 2-(azidobenzenesulfonyl)-1,2-thiazine 1-oxide. The latter species underwent a phosphite-mediated one-pot sulfur-extrusion, ring-contraction and azide to amine conversion to form 1-(amino-benzenesulfonyl)pyrroles. An unusual electrochemical oxidation of phenothiazine dye to phenothiazine-bi-1,4-quinone derivative (a donor-acceptor type molecular hybrid) on MWCNT surface and its cysteine electrocatalytic oxidation function

Phenothiazine (PTZ), a thiazine class heterocyclic compound, is a well-known electron donating system and has been widely used as a starting compound to prepare various phenothiazine dyes and pharmaceutically important compounds. Quinones and its derivatives are constituents of biologically active molecules serve as excellent electron-acceptor systems. Oxidation of PTZ by chemical and electrochemical methods often resulted into monohydroxylation of benzene ring moiety, S-oxidized and polymerized compounds as end products. Electrochemical oxidation of PTZ on a multiwalled carbon nanotube (MWCNT) modified glassy carbon electrode in pH 7 phosphate buffers solution (PBS) has been investigated in this work. A highly redox active surface confined PTZ-bi-1,4-quinone derivative (PTZ-biQ) on MWCNT modified glassy carbon electrode, designated as GCE/MWCNT@PTZ-biQ, as a product was unusually observed. The GCE/MWCNT@PTZ-biQ showed well-defined redox peaks at E-1/2 = -0.07 and +0.29 V vs Ag/AgCl corresponding to surface confined electron-transfer behavior of the bi-quinone (acceptor) and PTZ-cationic radical species (donor) respectively. No such electrochemical characteristics were noticed when unmodified GCE was subjected to the electrochemical oxidation of PTZ. Existence of PTZ-biQ was confirmed by XRD, Raman spectroscopy, FT-IR and GC-MS (methanolic extract of the active layer) analyses. Position of biQ in PTZ-biQ as 1,4-quinone isomer was confirmed by observation of absence of copper-complexation with 1,4-quinone and H2O2 electrochemical reduction reactions at -0.1 V vs Ag/AgCl unlike to the specific copper-complexation and H2O2 reduction with 1,2-quinone isomer in pH 7. Cysteine (CySH) oxidation was studied as a model system to understand the electron-transfer function of the MWCNT@PTZ-biQ. A highly selective electrocatalytic oxidation and sensing by amperometric i-t and flow injection analysis of CySH at low oxidation potential, 0.3 V vs Ag/AgCl in pH 7 PBS with detection limit values (signal-to-noise ratio = 3) of 11.10 mM and 110 nM respectively, without any interference from other biochemicals like uric acid, dopamine, nitrite, citric acid and H2O2, unlike the conventional chemically modified electrodes with serious interference’s, have been demonstrated.
